A new type of lordosis and vertebral body compression in Gilthead seabream, Sparus aurata Linnaeus, 1758: aetiology, anatomy and consequences for survival
A new type of vertebral malformation is described, consisting of deformed cartilaginous neural and haemal processes and the compression and fusion of vertebral bodies. The malformation is designated as haemal vertebral compression and fusion (haemal VCF). We studied the aetiology of the malformations and described microanatomical histopathological alterations. The malformations were detected during routine quality control in one of six monitored Gilthead sea bream populations. Haemal VCF affected the posterior part of the vertebral column (haemal vertebrae). In 20% of the deformed specimens, haemal VCF was combined with lordosis. At 35dph (days post-hatching), early anatomical signs of the haemal VCF consisted of abnormal centrum mineralization, malformed cartilaginous neural and haemal processes and developing lordotic alterations. The histological examination of the deformed individuals revealed that haemal VCF is preceded by notochord abnormalities. The frequency of deformed individuals was three times higher at 35 than at 61dph (50.3% vs. 17.2%, n=157 and n=250, respectively). No signs of repair or reversion of malformations have been observed. Thus, the steep decrease in deformities in older animals suggests that haemal VCF is linked to high mortality rates. The results are discussed in respect of the possible causative factors of haemal VCF
Allometric growth patterns in hatchery-reared larvae of the catfish Lophiosilurus alexandri (Pisces: Pseudopimelodidae)
The aim of the present study was to characterize the relationships that occur between morphometric variables of larvae of Lophiosilurus alexandri Steindachner, 1876 through their allometric growth analysis. Total length, head length and body height were correlated with standard length, head height and body height, as well as eye diameter and head height with head length. The results revealed allometric positive relation during the initial development, where the b coefficient varied between 1.10 and 2.81. The variable pre-anal distance in relation to the standard length and the snout length in relation to the head length revealed an allometric negative relation, where the b coefficient was 0.85 and 0.94, respectively
Yolk utilization and growth during the early larval life of the Silver Perch, Bidyanus bidyanus (Mitchell, 1838)
The aim of this research was to investigate the yolk sac and oil globule utilization by silver perch (Bidyanus bidyanus) larvae produced from domesticated broodfish. The larvae were kept unfed in the holding tank, sampled, and investigated by image analysis software to determine various characteristics, such as the diameters of ova, water-hardened eggs, yolk-sac, oil globules, and the total length of larvae. The research illustrated that, with the exception of oil globule diameter, all other morphometric parameters were significantly lower (P < 0.05) when compared to the larvae from the wild broodfish. The yolk sac was completely absorbed at 96 h post-hatching (hph) and the oil globule was visible until 240 hph. The larvae exhibited predatory movements and tried to catch rotifer at 4 days post hatching (dph). However, the onset of feeding took place at 5 dph, while 100% of feeding occurred at 6 dph. During the first 96 h (h), larvae grew significantly faster than the next 144 h. Larvae encountered low mortalities (<10%) during the first 96 hph, before increasing significantly in the next 24 h and no unfed larvae survived post 240 h. The results also suggested that the exogenous feed should be available at 96 hph, which is well after the yolk sac is completely depleted. In addition, although most of eggs and larval performance from domesticated broodfish were inferior compared to the wild one, it has larger oil globule that could make longer of its mixed feeding period and therefore could have better in viability
A next-generation sequencing method for overcoming the multiple gene copy problem in polyploid phylogenetics, applied to Poa grasses
<p>Abstract</p> <p>Background</p> <p>Polyploidy is important from a phylogenetic perspective because of its immense past impact on evolution and its potential future impact on diversification, survival and adaptation, especially in plants. Molecular population genetics studies of polyploid organisms have been difficult because of problems in sequencing multiple-copy nuclear genes using Sanger sequencing. This paper describes a method for sequencing a barcoded mixture of targeted gene regions using next-generation sequencing methods to overcome these problems.</p> <p>Results</p> <p>Using 64 3-bp barcodes, we successfully sequenced three chloroplast and two nuclear gene regions (each of which contained two gene copies with up to two alleles per individual) in a total of 60 individuals across 11 species of Australian <it>Poa </it>grasses. This method had high replicability, a low sequencing error rate (after appropriate quality control) and a low rate of missing data. Eighty-eight percent of the 320 gene/individual combinations produced sequence reads, and >80% of individuals produced sufficient reads to detect all four possible nuclear alleles of the homeologous nuclear loci with 95% probability.</p> <p>We applied this method to a group of sympatric Australian alpine <it>Poa </it>species, which we discovered to share an allopolyploid ancestor with a group of American <it>Poa </it>species. All markers revealed extensive allele sharing among the Australian species and so we recommend that the current taxonomy be re-examined. We also detected hypermutation in the <it>trn</it>H-<it>psb</it>A marker, suggesting it should not be used as a land plant barcode region. Some markers indicated differentiation between Tasmanian and mainland samples. Significant positive spatial genetic structure was detected at <100 km with chloroplast but not nuclear markers, which may be a result of restricted seed flow and long-distance pollen flow in this wind-pollinated group.</p> <p>Conclusions</p> <p>Our results demonstrate that 454 sequencing of barcoded amplicon mixtures can be used to reliably sample all alleles of homeologous loci in polyploid species and successfully investigate phylogenetic relationships among species, as well as to investigate phylogeographic hypotheses. This next-generation sequencing method is more affordable than and at least as reliable as bacterial cloning. It could be applied to any experiment involving sequencing of amplicon mixtures.</p
Effects of temperature in juvenile seabass (Dicentrarchus labrax L.) biomarker responses and behaviour: implications for environmental monitoring
The effects of temperature on European seabass
(Dicentrarchus labrax L.) juveniles were investigated using
a 30-day bioassay carried out at 18 and 25 °C in laboratory
conditions. A multiparameter approach was applied including
fish swimming velocity and several biochemical parameters
involved in important physiological functions. Fish exposed for
four weeks to 25 °C showed a decreased swimming capacity,
concomitant with increased oxidative stress (increased catalase
and glutathione peroxidase activities) and damage (increased
lipid peroxidation levels), increased activity of an enzyme
involved in energy production through the aerobic pathway
(isocitrate dehydrogenase) and increased activities of brain and
muscle cholinesterases (neurotransmission) compared to fish
kept at 18 °C. Globally, these findings indicate that basic
functions, essential for juvenile seabass surviving and well
performing in the wild, such as predation, predator avoidance,
neurofunction and ability to face chemical stress may be compromised
with increasing water temperature. This may be of
particular concern if D. labrax recruitment phase in northwest
European estuaries and coastal areas happens gradually inmore
warm environments as a consequence of global warming.
Considering that the selected endpoints are generally applied
in monitoring studies with different species, these findings also
highlight the need of more research, including interdisciplinary
and multiparameter approaches, on the impacts of temperature
on marine species, and stress the importance of considering
scenarios of temperature increase in environmental monitoring
and in marine ecological risk assessment
