Targeting oncogenic KRasG13C with nucleotide-based covalent inhibitors
Mutations within Ras proteins represent major drivers in human cancer. In this study, we report the structure-based design, synthesis, as well as biochemical and cellular evaluation of nucleotide-based covalent inhibitors for KRasG13C, an important oncogenic mutant of Ras that has not been successfully addressed in the past. Mass spectrometry experiments and kinetic studies reveal promising molecular properties of these covalent inhibitors, and X-ray crystallographic analysis has yielded the first reported crystal structures of KRasG13C covalently locked with these GDP analogues. Importantly, KRasG13C covalently modified with these inhibitors can no longer undergo SOS-catalysed nucleotide exchange. As a final proof-of-concept, we show that in contrast to KRasG13C, the covalently locked protein is unable to induce oncogenic signalling in cells, further highlighting the possibility of using nucleotide-based inhibitors with covalent warheads in KRasG13C-driven cancer

Beam-beam deflection as a beam tuning tool at the SLAC linear collider
To achieve maximum integrated luminosity at the SLAC Linear Collider, a method of noninvasive beam tuning is required. Traditional luminosity monitors based on Bhabha scattering are inadequate because of low instantaneous counting rates. Coherent deflections of one beam by the electromagnetic field of the other are sensitive not only to the relative steering of the two bunches but also to their spot sizes. A high resolution wire scanner for micron-size profile measurements at the SLC
Fine conductive fibers have been used to measure transverse beam dimensions of a few microns at the Stanford Linear Collider (SLC). The beam profile is obtained by scanning a fiber across the beam in steps as small as 1 [mu]m, and recording the secondary emission signal at each step, using a charge sensitive amplifier. We first outline the mechanical construction and the analogue electronics of the wire scanner. We then describe its performance in test beams and in actual operation. Search for charged Higgs decays of the top quark using hadronic tau decays
We present the result of a search for charged Higgs decays of the top quark,
produced in collisions at 1.8 TeV. When the charged
Higgs is heavy and decays to a tau lepton, which subsequently decays
hadronically, the resulting events have a unique signature: large missing
transverse energy and the low-charged-multiplicity tau. Data collected in the
period 1992-1993 at the Collider Detector at Fermilab, corresponding to
18.70.7~pb, exclude new regions of combined top quark and charged

Inclusive jet cross section in collisions at TeV
The inclusive jet differential cross section has been measured for jet
transverse energies, , from 15 to 440 GeV, in the pseudorapidity region
0.10.7. The results are based on 19.5 pb of data
collected by the CDF collaboration at the Fermilab Tevatron collider. The data
are compared with QCD predictions for various sets of parton distribution
functions. The cross section for jets with GeV is significantly
higher than current predictions based on O() perturbative QCD
calculations. Various possible explanations for the high- excess are

Measurement of Dijet Angular Distributions at CDF
We have used 106 pb^-1 of data collected in proton-antiproton collisions at
sqrt(s)=1.8 TeV by the Collider Detector at Fermilab to measure jet angular
distributions in events with two jets in the final state. The angular
distributions agree with next to leading order (NLO) predictions of Quantum
Chromodynamics (QCD) in all dijet invariant mass regions. The data exclude at
95% confidence level (CL) a model of quark substructure in which only up and
down quarks are composite and the contact interaction scale is Lambda_ud(+) <
1.6 TeV or Lambda_ud(-) < 1.4 TeV. For a model in which all quarks are
composite the excluded regions are Lambda(+) < 1.8 TeV and Lambda(-) < 1. 6

Search for New Particles Decaying to Dijets at CDF
We have used 106 pb^-1 of data collected with the Collider Detector at
Fermilab to search for new particles decaying to dijets. We exclude at the 95%
confidence level models containing the following new particles: axigluons and
flavor universal colorons with mass between 200 and 980 GeV/c, excited quarks
with mass between 80 and 570 GeV/c^2 and between 580 and 760 GeV/c^2, color
octet technirhos with mass between 260 and 480 GeV/c^2, W' bosons with mass
between 300 and 420 GeV/c^2, and E_6 diquarks with mass between 290 and 420

The performance of "Virtual Phase" CCDs as detectors of minimum-ionizing particles
The Texas Instruments "Virtual Phase" CCD has been the basis of an ambitious design for a precision vertex detector to be used at the Stanford Linear Collider. The performance of this chip shows promise for future use in electron linear colliders. Relationships between serum adiponectin and soluble TNF-α receptors and glucose and lipid oxidation in lean and obese subjects
Insulin resistance might be associated with an impaired ability of insulin to stimulate glucose oxidation and inhibit lipid oxidation. Insulin action is also inversely associated with TNF-α system and positively related to adiponectin. The aim of the present study was to analyze the associations between serum adiponectin, soluble TNF-α receptors concentrations and the whole-body insulin sensitivity, lipid and glucose oxidation, non-oxidative glucose metabolism (NOGM) and metabolic flexibility in lean and obese subjects. We examined 53 subjects: 25 lean (BMI < 25 kg Ă mâ2) and 28 with overweight or obesity (BMI > 25 kg Ă mâ2) with normal glucose tolerance. Hyperinsulinemic euglycemic clamp and indirect calorimetry were performed. An increase in respiratory exchange ratio in response to insulin was used as a measure of metabolic flexibility. Obese subjects had lower insulin sensitivity, adiponectin and higher sTNFR1 (all P < 0.001) and sTNFR2 (P = 0.001). Insulin sensitivity was positively related to adiponectin (r = 0.49, P < 0.001) and negatively related to sTNFR1 (r = â0.40, P = 0.004) and sTNFR2 (r = â0.52, P < 0.001). Adiponectin was related to the rate of glucose (r = 0.47, P < 0.001) and lipid (r = â0.40, P = 0.003) oxidation during the clamp, NOGM (r = 0.41, P = 0.002) and metabolic flexibility (r = 0.36, P = 0.007). Serum sTNFR1 and sTNFR2 were associated with the rate of glucose (r = â0.45, P = 0.001; r = â0.51, P < 0.001, respectively) and lipid (r = 0.52, P < 0.001; r = 0.46, P = 0.001, respectively) oxidation during hyperinsulinemia, NOGM (r = â0.31, P = 0.02; r = â0.43, P = 0.002, respectively) and metabolic flexibility (r = â0.47 and r = â0.51, respectively, both P < 0.001) in an opposite manner than adiponectin. Our data suggest that soluble TNF-α receptors and adiponectin have multiple effects on glucose and lipid metabolism in obesity
What effect does physician "profiling" have on inpatient physician satisfaction and hospital length of stay?
BACKGROUND: 2002 marked the first time that the rate of hospital spending in the United States outpaced the overall health care spending rate of growth since 1991. As hospital spending continues to grow and as reimbursement for hospital expenses has moved towards the prospective payment system, there is still increasing pressure to reduce costs. Hospitals have a major incentive to decrease resource utilization, including hospital length of stay. We evaluated whether physician profiling affects physician satisfaction and hospital length of stay, and assessed physicians' views concerning hospital cost containment and the quality of care they provide. METHODS: To determine if physician profiling affects hospital length of stay and/or physician satisfaction, we used quasi-experimental with before-versus-after and intervention-versus-control comparisons of length of stay data collected at an intervention and six control hospitals. Intervention hospital physicians were informed their length of stay would be compared to their peers and were given a questionnaire assessing their experience. RESULTS: Nearly half of attending pre-profiled physicians felt negative about the possibility of being profiled, while less than one-third of profiled physicians reported feeling negative about having been profiled. Nearly all physicians greatly enjoyed their ward month. Length of stay at the profiled site decreased by an additional 1/3 of a day in the profiling year, compared to the non-profiled sites (p < 0.001). CONCLUSION: A relatively non-instrusive profiling intervention modestly reduced length of stay without adversely affecting physician satisfaction
