    Biological Effect of Gas Plasma Treatment on CO 2

    Porous polycaprolactone (PCL) scaffolds were fabricated by using the CO2 gas foaming/salt leaching process and then PCL scaffolds surface was treated by oxygen or nitrogen gas plasma in order to enhance the cell adhesion, spreading, and proliferation. The PCL and NaCl were mixed in the ratios of 3 : 1. The supercritical CO2 gas foaming process was carried out by solubilizing CO2 within samples at 50°C and 8 MPa for 6 hr and depressurization rate was 0.4 MPa/s. The oxygen or nitrogen plasma treated porous PCL scaffolds were prepared at discharge power 100 W and 10 mTorr for 60 s. The mean pore size of porous PCL scaffolds showed 427.89 μm. The gas plasma treated porous PCL scaffolds surface showed hydrophilic property and the enhanced adhesion and proliferation of MC3T3-E1 cells comparing to untreated porous PCL scaffolds. The PCL scaffolds produced from the gas foaming/salt leaching and plasma surface treatment are suitable for potential applications in bone tissue engineering

    Co-occurrence matrix analysis-based semi-supervised training for object detection

    One of the most important factors in training object recognition networks using convolutional neural networks (CNNs) is the provision of annotated data accompanying human judgment. Particularly, in object detection or semantic segmentation, the annotation process requires considerable human effort. In this paper, we propose a semi-supervised learning (SSL)-based training methodology for object detection, which makes use of automatic labeling of un-annotated data by applying a network previously trained from an annotated dataset. Because an inferred label by the trained network is dependent on the learned parameters, it is often meaningless for re-training the network. To transfer a valuable inferred label to the unlabeled data, we propose a re-alignment method based on co-occurrence matrix analysis that takes into account one-hot-vector encoding of the estimated label and the correlation between the objects in the image. We used an MS-COCO detection dataset to verify the performance of the proposed SSL method and deformable neural networks (D-ConvNets) as an object detector for basic training. The performance of the existing state-of-the-art detectors (DConvNets, YOLO v2, and single shot multi-box detector (SSD)) can be improved by the proposed SSL method without using the additional model parameter or modifying the network architecture.Comment: Submitted to International Conference on Image Processing (ICIP) 201

    Differences in the Gene Expression Profiles of Slow- and Fast-Forming Preinduced Pluripotent Stem Cell Colonies

    Induced pluripotent stem cells (iPSCs) are generated through a gradual process in which somatic cells undergo a number of stochastic events. In this study, we examined whether two different doxycycline-inducible iPSCs, slow-forming 4F2A-iPSCs and fast-forming NGFP-iPSCs, have equivalent levels of pluripotency. Multiplex reverse-transcriptase PCR generated gene expression profiles (GEPs) of 13 pluripotency genes in single initially formed-iPSC (if-iPSC) colonies of NGFP and 4F2A group. Assessment of GEP difference using a weighted root mean square deviation (wRMSD) indicates that 4F2A if-iPSCs are more closely related to mESCs than NGFP if-iPSCs. Consistently, Nanog and Sox2 genes were more frequently derepressed in 4F2A if-iPSC group. We further examined 20 genes that are implicated in reprogramming. They were, overall, more highly expressed in NGFP if-iPSCs, differing from the pluripotency genes being more expressed in 4F2A if-iPSCs. wRMSD analysis for these reprogramming-related genes confirmed that the 4F2A if-iPSC colonies were less deviated from mESCs than the NGFP if-iPSC colonies. Our findings suggest that more important in attaining a better reprogramming is the mode of action by the given reprogramming factors, rather than the total activity of them exerting to the cells, as the thin-but-long-lasting mode of action in 4F2A if-iPSCs is shown to be more effective than its full-but-short-lasting mode in NGFP if-iPSCs

    Effect of rhBMP-2 applied with a 3D-printed titanium implant on new bone formation in rabbit calvarium

    Objective: This study sought to compare the biocompatibility of a three-dimensional (3D)-printed titanium implant with a conventional machined titanium product, as well as the effect of such implant applied with recombinant human Bone Morphogenetic Protein Type 2 (rhBMP-2) for guided bone regeneration. Methodology: Disk-shaped titanium specimens fabricated either by the conventional machining technique or by the 3D-printing technique were compared by MC3T3-E1 cells cytotoxicity assay. New bone formation was evaluated using a rapid prototype titanium cap applied to the calvaria of 10 rabbits, which were divided into two groups: one including an atelopeptide collagen plug on one side of the cap (group I) and the other including a plug with rhBMP-2 on the other side (group II). At six and 12 weeks after euthanasia, rabbits calvaria underwent morphometric analysis through radiological and histological examination. Results: Through the cytotoxicity assay, we identified a significantly higher number of MC3T3-E1 cells in the 3D-printed specimen when compared to the machined specimen after 48 hours of culture. Moreover, morphometric analysis indicated significantly greater bone formation at week 12 on the side where rhBMP-2 was applied when evaluating the upper portion immediately below the ca p. Conclusion: The results suggest that 3D-printed titanium implant applied with rhBMP-2 enables new bone formation

    Effect of 660 nm Light-Emitting Diode on the Wound Healing in Fibroblast-Like Cell Lines

    Light in the red to near-infrared (NIR) range (630–1000 nm), which is generated using low energy laser or light-emitting diode (LED) arrays, was reported to have a range of beneficial biological effects in many injury models. NIR via a LED is a well-accepted therapeutic tool for the treatment of infected, ischemic, and hypoxic wounds as well as other soft tissue injuries in humans and animals. This study examined the effects of exposure to 660 nm red LED light at intensities of 2.5, 5.5, and 8.5 mW/cm2 for 5, 10, and 20 min on wound healing and proliferation in fibroblast-like cells, such as L929 mouse fibroblasts and human gingival fibroblasts (HGF-1). A photo illumination-cell culture system was designed to evaluate the cell proliferation and wound healing of fibroblast-like cells exposed to 600 nm LED light. The cell proliferation was evaluated by MTT assay, and a scratched wound assay was performed to assess the rate of migrating cells and the healing effect. Exposure to the 660 nm red LED resulted in an increase in cell proliferation and migration compared to the control, indicating its potential use as a phototherapeutic agent
