Evidence that polymorphonuclear neutrophils infiltrate into the developing corpus luteum and promote angiogenesis with interleukin-8 in the cow
<p>Abstract</p> <p>Background</p> <p>After ovulation in the cow, the corpus luteum (CL) rapidly develops within a few days with angiogenesis and progesterone production. CL formation resembles an inflammatory response due to the influx of immune cells. Neutrophils play a role in host defense and inflammation, and secrete chemoattractants to stimulate angiogenesis. We therefore hypothesized that neutrophils infiltrate in the developing CL from just after ovulation and may play a role in angiogenesis of the CL.</p> <p>Methods and Results</p> <p>Polymorphonuclear neutrophils (PMN) were detected in CL tissue by Pas-staining, and interleukin-8 (IL-8, a neutrophil-specific chemoattractant) was measured in supernatant of the CL tissue culture: considerable amounts of PMNs and the high level of IL-8 were observed during the early luteal phase (days 1-4 of the estrous cycle). PMNs and IL-8 were low levels in the mid and late luteal phases, but IL-8 was increased during luteal regression. The PMN migration in vitro was stimulated by the supernatant from the early CL but not from the mid CL, and this activity was inhibited by neutralizing with an anti-IL-8 antibody, indicating the major role of IL-8 in inducing active PMN migration in the early CL. Moreover, IL-8 stimulated proliferation of CL-derived endothelial cells (LECs), and both the supernatant of activated PMNs and IL-8 stimulated formation of capillary-like structures of LECs.</p> <p>Conclusion</p> <p>PMNs migrate into the early CL partially due to its major chemoattractant IL-8 produced at high levels in the CL, and PMNs is a potential regulator of angiogenesis together with IL-8 in developing CL in the cow.</p
Periductal Induction of High Endothelial Venule-Like Vessels in Type 1 Autoimmune Pancreatitis
信州大学博士(医学)・学位論文・平成24年3月31日授与(甲第946号)・丸山 雅史This is a non-final version of an article published in final form in PANCREAS. 42(1):53-59 (2013).Objectives: Type 1 autoimmune pancreatitis (AIP) is histologically characterized by dense lymphoplasmacytic infiltration and marked storiform fibrosis, manifestations associated with pancreatic ducts. Such periductal lymphocyte recruitment is thought to be elicited by dysregulation of mechanisms governing physiological lymphocyte homing. The present study was undertaken to determine whether vascular addressins including peripheral lymph node addressin and mucosal addressin cell adhesion molecule 1 (MAdCAM-1) play a role in type 1 AIP histogenesis. Methods: Tissue sections of type 1 AIP and tumor-associated non-AIP chronic pancreatitis, as well as normal pancreas, were subjected to immunohistochemical analysis using vascular addressin-related antibodies. Results: The number of periductal mouse endothelial cell antigen 79-positive high endothelial venule (HEV)-like vessels was increased in type 1 AIP relative to that seen in non-AIP chronic pancreatitis, whereas the number of MAdCAM-1-positive HEV-like vessels did not differ between the 2 conditions. Mouse endothelial cell antigen 79 antigens are expressed on duct-forming epithelial cells not only in pancreas but also in salivary glands, which often harbor extrapancreatic lesions in type 1 AIP. Conclusions: Type 1 AIP can be characterized by periductal induction of MECA-79-positive HEV-like vessels. MECA-79-positive 6-sulfo sialyl Lewis X-related carbohydrate antigens expressed on duct-forming epithelial cells could be associated with type 1 AIP pathogenesis.ArticlePANCREAS. 42(1):53-59 (2013)journal articl
Magnetostriction studies up to megagauss fields using fiber Bragg grating technique
We here report magnetostriction measurements under pulsed megagauss fields
using a high-speed 100 MHz strain monitoring system devised using fiber Bragg
grating (FBG) technique with optical filter method. The optical filter method
is a detection scheme of the strain of FBG, where the changing Bragg wavelength
of the FBG reflection is converted to the intensity of reflected light to
enable the 100 MHz measurement. In order to show the usefulness and reliability
of the method, we report the measurements for solid oxygen, spin-controlled
crystal, and volborthite, a deformed Kagom\'{e} quantum spin lattice, using
static magnetic fields up to 7 T and non-destructive millisecond pulse magnets
up to 50 T. Then, we show the application of the method for the
magnetostriction measurements of CaVO, a two-dimensional
antiferromagnet with spin-halves, and LaCoO, an anomalous spin-crossover
oxide, in the megagauss fields.Comment: 9pages, 6 figures, Conference proceedings for MegaGauss16 at Kashiwa,

A survey on recognition of the "bryophytes" targeting university students
The aim of the present study was to grasp the current situation of university students' understanding about the bryophytes. The survey was conducted on 49 students of National University "A", Faculty of Education. The contents of the questionnaire were (1) ability to distinguish the bryophytes, (2) knowledge about bryophytes, and (3) attitudes toward the bryophytes and science classes, nature experiences. The results were as follows. In the question (1), it was difficult to distinguish the bryophytes from the lichens or the algae. In the question (2), many students did not understand the classification and reproduction of the bryophytes. Factor analysis was conducted on the result of the question (3), and calculated Pearson’s correlation coefficient with the question (1) or (2). As the results, weak correlations (.227, .235) were observed between the total score of question (1) and the factors "interests in the bryophytes" or "interests in science (biology)". Weak correlation (.246) was also observed between "knowledge on reproduction of bryophytes" and "positive nature experiences". These results suggested that there are relevance between distinguishing or understanding and attitude of the bryophytes. However, more detailed examination is required.本研究の一部は,JSPS科研費17H01980および16K00966の助成を受けて行った
The Inhibitory Effect of Kakkonto, Japanese Traditional (Kampo) Medicine, on Brain Penetration of Oseltamivir Carboxylate in Mice with Reduced Blood-Brain Barrier Function
Oseltamivir phosphate (OP) is used to treat influenza virus infections. However, its use may result in central nervous system (CNS) adverse effects. In Japan, OP is used with Kampo formulations to improve clinical effectiveness. We evaluated the potential for using Kampo formulations to reduce CNS adverse effects by quantifying the CNS distribution of oseltamivir and its active metabolite oseltamivir carboxylate (OC) when administered with maoto and kakkonto. We administered lipopolysaccharide (LPS) by intraperitoneal injection to C57BL/6 mice to reduce blood-brain barrier function. Saline, maoto, and kakkonto were administered orally at the same time as LPS. OP was orally administered 4 hours after the last LPS injection and the migration of oseltamivir and OC was examined. Additionally, we examined the brain distribution of OC following intravenous administration. Changes in OC concentrations in the brain suggest that, in comparison to LPS-treated control mice, both Kampo formulations increased plasma levels of OC, thereby enhancing its therapeutic effect. Additionally, our findings suggest kakkonto may not only improve the therapeutic effect of oseltamivir but also reduce the risk of CNS-based adverse effects. Considering these findings, it should be noted that administration of kakkonto during periods of inflammation has led to increased OAT3 expression
