    Recognition of viral glycoproteins by influenza A-specific cross- reactive cytolytic T lymphocytes

    Two populations of cytolytic T lymphocytes (CTL) generated after influenza A virus infection can be distinguished into one with specificity for the sensitizing hemagglutinin type and a second with cross-reactivity for antigens induced by other type-A influenza viruses. The molecules carrying the antigenic determinants recognized by the cross-reactive CTL were studied. In L-929 cells abortively infected with fowl plague virus, matrix (M) protein synthesis is specifically inhibited, whereas the envelope glycoproteins, hemagglutinin and neuraminidase, are synthesized and incorporated into the plasma membrane. These target cells were lysed by cross-reactive CTL. The envelope proteins of type A/Victoria virus were separated from the other virion components and reconstituted into lipid vesicles that lacked M protein that subsequently were used to prepare artificial target cells. Target-cell formation with vesicles was achieved by addition of fusion-active Sendai virus. These artificial target cells were also susceptible to lysis by cross-reactive CTL. In contrast to previous observations that suggested that the M protein of influenza viruses is recognized by these effector cells, we present evidence that the antigencic determinants induced by the viral glycoproteins are recognized

    A sequential process is used to synthesise CuInS2 absorber layers for photovoltaic application. In this process CuIn precursor layers sputtered on molybdenum coated float glass are converted to CuInS2 via sulphurisation in an elemental sulphur vapour ambient. A re evaluation of process parameters has been performed including fine tuning of numerous minor aspects. Using optimised process conditions has lead to improved device performance, especially a narrowed distribution at higher module efficiencies is achieved.     User's guide for the Solar Backscattered Ultraviolet (SBUV) instrument first year ozone-S data set

    Total-ozone and ozone vertical profile results for Solar Backscattered Ultraviolet/Total Ozone Mapping Spectrometer (SBUV/TOMS) Nimbus 7 operation from November 1978 to November 1979 are available. The algorithm used have been thoroughly tested, the instrument performance has been examined in details, and the ozone results have been compared with Dobson, Umkehr, balloon, and rocket observations. The accuracy and precision of the satellite ozone data are good to at least within the ability of the ground truth to check and are self-consistent to within the specifications of the instrument. The 'SBUV User's Guide' describes the SBUV experiment and algorithms used. Detailed information on the data available on computer tape is provided including how to order tapes from the National Space Science Data Center

    The parathyroid hormone receptor (PTHR) is a class B G protein-coupled receptor (GPCR) that mediates the endocrine and paracrine effects of parathyroid hormone and related peptides through the activation of phospholipase C beta-, adenylyl cyclase-, mitogen-activated protein kinase-, and beta-arrestin-initiated signaling pathways. It is currently not clear how specificity among these downstream signaling pathways is achieved. A possible mechanism involves adaptor proteins that affect receptor/effector coupling. In a proteomic screen with the PTHR C terminus, we identified vav2, a guanine nucleotide exchange factor (GEF) for Rho GTPases, as a PTHR-interacting protein. The core domains of vav2 bound to the intracellular domains of the PTHR independent of receptor activation. In addition, vav2 specifically interacted with activated G alpha(q) but not with G alpha(s) subunits, and it competed with PTHR for coupling to G alpha(q). Consistent with its specific interaction with G alpha(q), vav2 impaired G(q)-mediated inositol phosphate generation but not G(s)-mediated cAMP generation. This inhibition of G(q) signaling was specific for PTHR signaling, compared with other G(q)-coupled GPCRs. Moreover, the benefit for PTHR-mediated inositol phosphate generation in the absence of vav2 required the ezrin binding domain of Na+/H+-exchanger regulatory factor 1. Our results show that a RhoA GEF can specifically interact with a GPCR and modulate its G protein signaling specificity

    Raw data from the Solar Backscattered Ultrviolet/Total Ozone Mapping Spectrometer (SBUV/TOMS) Nimbus 7 operation are available on computer tape. These data are contained on two separate sets of RUTs (Raw Units Tapes) for SBUV and TOMS, labelled RUT-S and RUT-T respectively. The RUT-S and RUT-T tapes contain uncalibrated radiance and irradiance data, housekeeping data, wavelength and electronic calibration data, instrument field-of-view location and solar ephemeris information. These tapes also contain colocated cloud, terrain pressure and snow/ice thickness data, each derived from an independent source. The "RUT User's Guide" describes the SBUV and TOMS experiments, the instrument calibration and performance, operating schedules, and data coverage, and provides an assessment of RUT-S and -T data quality. It also provides detailed information on the data available on the computer tapes

    Preparing for and Responding to Disturbance: Examples from the Forest Sector in Sweden and Canada

    Coping or adaptation following large-scale disturbance may depend on the political system and its preparedness and policy development in relation to risks. Adaptive or foresight planning is necessary in order to account and plan for potential risks that may increase or take place concurrently with climate change. Forests constitute relevant examples of large-scale renewable resource systems that have been directly affected by recent environmental and social changes, and where different levels of management may influence each other. This article views disturbances in the forest sectors of Sweden and Canada, two large forest nations with comparable forestry experiences, in order to elucidate the preparedness and existing responses to multiple potential stresses. The article concludes that the two countries are exposed to stresses that indicate the importance of the governing and institutional system particularly with regard to multi-level systems including federal and EU levels. While economic change largely results in privatization of risk onto individual companies and their economic resources (in Canada coupled with a contestation of institutional systems and equity in these), storm and pest outbreaks in particular challenge institutional capacities at administrative levels, within the context provided by governance and tenure systems.In Sweden, funding from the research agency FORMAS, the MISTRA Arctic Futures programme, and the Future Forests programme (funded by research agency MISTRA, the forest industries, Umeå University and the Swedish University of Agricultural Sciences) as well as from the EU for data collection is acknowledged.     Time resolved investigation of Cu In,Ga Se2 growth and Ga gradient formation during fast selenization of metallic precursors

    Ga segregation at the backside of Cu In,Ga Se2 solar cell absorbers is a commonly observed phenomenon for a large variety of sequential fabrication processes. Here, we investigate the correlation between Se incorporation, phase formation and Ga segregation during fast selenisation of Cu In Ga precursor films in elemental selenium vapour. Se incorporation and phase formation are analysed by real time synchrotron based X ray diffraction and fluorescence analysis. Correlations between phase formation and depth distributions are gained by interrupting the process at several points and by subsequent ex situ cross sectional electron microscopy and Raman spectroscopy. The presented results reveal that the main share of Se incorporation takes place within a few seconds during formation of In Se at the top part of the film, accompanied by outdiffusion of In out of a ternary Cu In Ga phase. Surprisingly, CuInSe2 starts to form at the surface on top of the In Se layer, leading to an intermediate double graded Cu depth distribution. The remaining Ga rich metal phase at the back is finally selenised by indiffusion of Se. On the basis of a proposed growth model, we discuss possible strategies and limitations for the avoidance of Ga segregation during fast selenisation of metallic precursors. Solar cells made from samples selenised with a total annealing time of 6.5 amp; 8201;min reached conversion efficiencies of up to 14.2 total area, without anti reflective coating .     <em>Blastococcus brunescens</em> sp. nov., a member of the <em>Geodermatophilaceae </em>isolated from sandstone collected from the Sahara Desert in Tunisia

    \ua9 2024 The Authors.The taxonomic position of strain BMG 8361T, isolated from sandstone collected in the Sahara Desert of Southern Tunisia, was refined through a polyphasic taxonomic investigation. Colonies of BMG 8361T were pale-orange coloured, irregular with a dry surface and produced a diffusible pink or brown pigment depending on media. The Gram-positive cells were catalase-positive and oxidase-negative. The strain exhibited growth at 10–40\ub0C and pH values ranging from 5.5 to 9.0, with optima at 28–35\ub0C and pH 6.5–8.0. Additionally, BMG 8361T demonstrated the ability to grow in the presence of up to 1% NaCl (w/v) concentration. The peptidoglycan of the cell wall contained meso-diaminopimelic acid, glucose, galactose, xylose, ribose, and rhamnose. The predominant menaquinones consisted of MK-9(H4) and MK-9. The main polar lipids were phosphatidylcholine, phosphatidylinositol, glycophosphatidylinositol, diphosphatidylglycerol, phosphatidylethanolamine, and two unidentified lipids. Major cellular fatty acids were iso-C16:0, iso-C16:1 h, and C17:1 ω8c. Phylogenetic analyses based on both the 16S rRNA gene and whole-genome sequences assigned strain BMG 8361T within the genus Blastococcus. The highest pairwise sequence similarity observed in the 16S rRNA gene was 99.5% with Blastococcus haudaquaticus AT 7-14T. However, when considering digital DNA–DNA hybridization and average nucleotide identity, the highest values, 48.4 and 86.58%, respectively, were obtained with Blastococcus colisei BMG 822T. These values significantly undershoot the recommended thresholds for establishing new species, corroborating the robust support for the distinctive taxonomic status of strain BMG 8361T within the genus Blastococcus. In conjunction with the phenotyping results, this compelling evidence leads to the proposal of a novel species we named Blastococcus brunescens sp. nov. with BMG 8361T (=DSM 46845T=CECT 8880T) as the type strain

    Demand-supply matching through auctioning for the circular economy

    The circular economy aims to reduce the consumption of resources and energy by exploiting multiple use-cycles of components and materials. The creation of new circular businesses hinges on efficient alignment between market demands of circular products with the supply of End-of-life components and materials. In this paper, we address the digitization of a matchmaking tool for the circular economy by defining demand-supply matching (DSM) in context of business link identification and cross-sectorial matchmaking. We further specify a DSM process and p resent our DSM tool, which facilitates publication and search for supplier offerings and demander needs, selection of auctioning candidates, and digitized auctioning and contract definition. By that, this tool supports the alignment of market demands with matching supply offerings. In particular, it combines the steps of publishing, searching, selecting, auctioning and contract definition into one tool, which we argue can make matchmaking more efficient compared to addressing these steps separately. Finally, we present the design of the tool and discuss its merits in light of the needed acceptance for automating business link identification and contractual interactions
