Evidence for vivianite formation and its contribution to long-term phosphorus retention in a recent lake sediment: a novel analytical approach
Vivianite, Fe3(PO4)2 ¡ 8 H2O, is a ferrous iron
phosphate mineral which forms in waterlogged soils and
sediments. The phosphorus (P) bound in its crystal lattice is
considered to be immobilised because vivianite is stable under
anoxic, reducing, sedimentary conditions. Thus, vivianite formation
can make a major contribution to P retention during early
diagenesis. Much remains unknown about vivianite in sediments,
because technical challenges have rendered direct identification and
quantification difficult. To identify vivianite and assess its
significance for P burial during early diagenesis we studied the
consequences of a 1992/1993 in-lake application of FeCl3 and
Fe(OH)3 aimed at restoring Lake GroĂ-Glienicke (Berlin,
Germany). In a novel approach, we firstly applied a heavy-liquid
separation to the iron-rich surface sediments which allowed direct
identification of vivianite by X-ray diffraction in the high-density
(ρ > 2.3 g cm−3) sediment fraction. Secondly, we
assessed the contribution of vivianite to P retention, combining
results from chemical digestion with magnetic susceptibility data
derived from magnetic hysteresis measurements. Scanning electron
microscopy revealed that the dark blue spherical vivianite nodules
were 40â180 Îźm in diameter, and formed of platy- and
needle-shaped crystal aggregates. Although equilibrium calculations
indicated supersaturation of vivianite throughout the upper
30 cm of the sediment, the vivianite deposits were
homogeneously distributed within, and restricted to, the upper
23 cm only. Thus, supersaturated pore water alone cannot
serve as a reliable predictor for the in situ formation of
vivianite. In Lake GroĂ -Glienicke, vivianite formation continues
to be triggered by the artificial iron amendment more than
20 yr ago, significantly contributing to P retention in
surface sediments
Progressive decline of decision-making performances during multiple sclerosis
The purpose of this study was to evaluate longitudinally, using the Iowa Gambling Task (IGT), the dynamics of decision-making capacity at a two-year interval (median: 2.1 years) in a group of patients with multiple sclerosis (MS) (n = 70) and minor neurological disability [Expanded Disability Status Scale (EDSS) ⤠2.5 at baseline]. Cognition (memory, executive functions, attention), behavior, handicap, and perceived health status were also investigated. Standardized change scores [(score at retest-score at baseline)/standard deviation of baseline score] were computed. Results showed that IGT performances decreased from baseline to retest (from 0.3, SD = 0.4 to 0.1, SD = 0.3, p = .005). MS patients who worsened in the IGT were more likely to show a decreased perceived health status and emotional well-being (SEP-59; p = .05 for both). Relapsing rate, disability progression, cognitive, and behavioral changes were not associated with decreased IGT performances. In conclusion, decline in decision making can appear as an isolated deficit in MS. (JINS, 2009, 15, 291-295.
Evaluating inputs for organic farming â a new system. Proposals of the ORGANIC INPUTS EVALUATION project
This volume contains proposals for criteria for evaluation of plant protection products, fertilisers and soil conditioners1 to be used in organic agriculture. These ideas were developed in the course of the European Union (EU) Concerted Action project âORGANIC INPUTS EVALUATIONâ (QLK5-CT-2002-02565). For more information on this project see the end of this volume or visit the project website www.organicinputs.org. The documents in this volume are proposals elaborated by the project consortium and external experts. They were discussed with a broader audience at a public conference held in Brussels on October 13, 2005, and have been amended accordingly.
Our proposals also include a âcriteria matrixâ, which is in Microsoft Excel format, and therefore stands as a separate file. The criteria matrix is discussed in section 5, but we strongly recommend that you consult the original document. To illustrate the use of the matrix, we have further prepared two case studies, which are also separate Excel files. All of these files are contained on the CD, and can also be downloaded from the project website.
Currently, Regulation 2092/91 is under revision. We hope that our ideas can be incorporated into the regulation during this revision! In addition, we strongly encourage national institutions to make use of our proposals at the national level
Development of a botanical plant protection product from Larix by-products to protect grapevine from Plasmopara viticola
Extracts from European Larch (Larix decidua) were shown to be efficient to control grapevine downy mildew (Plasmopara viticola) under controlled and field conditions. Larixyl acetate and larixol were identified as the active compounds
Strong EBV-specific CD8+ T-cell response in patients with early multiple sclerosis
Epstein-Barr virus (EBV) has been associated with multiple sclerosis (MS), however, most studies examining the relationship between the virus and the disease have been based on serologies, and if EBV is linked to MS, CD8+ T cells are likely to be involved as they are important both in MS pathogenesis and in controlling viruses. We hypothesized that valuable information on the link between MS and EBV would be ascertained from the study of frequency and activation levels of EBV-specific CD8+ T cells in different categories of MS patients and control subjects. We investigated EBV-specific cellular immune responses using proliferation and enzyme linked immunospot assays, and humoral immune responses by analysis of anti-EBV antibodies, in a cohort of 164 subjects, including 108 patients with different stages of MS, 35 with other neurological diseases and 21 healthy control subjects. Additionally, the cohort were all tested against cytomegalovirus (CMV), another neurotropic herpes virus not convincingly associated with MS, nor thought to be deleterious to the disease. We corrected all data for age using linear regression analysis over the total cohorts of EBV- and CMV-infected subjects. In the whole cohort, the rate of EBV and CMV infections were 99% and 51%, respectively. The frequency of IFN-Îł secreting EBV-specific CD8+ T cells in patients with clinically isolated syndrome (CIS) was significantly higher than that found in patients with relapsing-remitting MS (RR-MS), secondary-progressive MS, primary-progressive MS, patients with other neurological diseases and healthy controls. The shorter the interval between MS onset and our assays, the more intense was the EBV-specific CD8+ T-cell response. Confirming the above results, we found that EBV-specific CD8+ T-cell responses decreased in 12/13 patients with CIS followed prospectively for 1.0 Âą 0.2 years. In contrast, there was no difference between categories for EBV-specific CD4+ T cell, or for CMV-specific CD4+ and CD8+ T-cell responses. Anti-EBV-encoded nuclear antigen-1 (EBNA-1)-specific antibodies correlated with EBV-specific CD8+ T cells in patients with CIS and RR-MS. However, whereas EBV-specific CD8+ T cells were increased the most in early MS, EBNA-1-specific antibodies were increased in early as well as in progressive forms of MS. Our data show high levels of CD8+ T-cell activation against EBVâbut not CMVâearly in the course of MS, which support the hypothesis that EBV might be associated with the onset of this diseas
The relationship between perceived service quality and patient willingness to recommend at a national oncology hospital network
<p>Abstract</p> <p>Background</p> <p>"Willingness to recommend" questions are being increasingly used to measure and manage patient loyalty. Yet, there is little data in the literature correlating the "willingness to recommend" question with commonly used perceived service quality items in surveys to identify the key drivers of the optimal patient experience. We therefore evaluated the relationship between perceived service quality and subsequent single top box "willingness to recommend" scores among oncology patients.</p> <p>Methods</p> <p>A total of 2018 returning cancer patients treated at Cancer Treatment Centers of AmericaÂŽ (CTCA) responded to an internally developed service quality questionnaire, which covered the following dimensions: operations and services, treatment and care with a multidisciplinary team and patient endorsements. Items were measured on a 7-point Likert-type scale ranging from "completely dissatisfied" to "completely satisfied." Patient willingness to, "recommend this facility to friends and associates" was measured on an 11-point scale ranging from "not at all likely" to "extremely likely", which was subsequently dichotomized into two categories: top box response (10) versus all others (0-9). The relationship between perceived service quality and "willingness to recommend" was assessed via Kendall's tau b correlation and univariate and multivariate logistic regression.</p> <p>Results</p> <p>Of the 2018 patients, 959 were newly diagnosed while 1059 were previously treated. 902 were males and 1116 females. The mean age was 54.2 years and the most frequent diagnoses were breast (412), lung (294), prostate (260), colorectal (179) and pancreas (169). 1553 patients said they were "extremely likely" to recommend CTCA to friends and associates, resulting in 77% "top box" responses while 465 (23%) responded in all other categories. The key service quality drivers that were statistically significant in the final logistic model were "team helping you understand your medical condition", "staff genuinely caring for you as an individual", "whole person approach to patient care" and "CTCA medical oncologist."</p> <p>Conclusions</p> <p>In this multi-center study, we demonstrate the predictive significance of perceived service quality as it relates to patient willingness to recommend an oncology service provider. This study is unique in reporting on the role of perceived service quality as a predictor of patient willingness to recommend in a large sample of cancer patients.</p
Production of a recombinant polyester-cleaving hydrolase from Thermobifida fusca in Escherichia coli
The hydrolase (Thermobifida fusca hydrolase; TfH) from T. fusca was produced in Escherichia coli as fusion protein using the OmpA leader sequence and a His(6) tag. Productivity could be raised more than 100-fold. Both batch and fed-batch cultivations yield comparable cell specific productivities whereas volumetric productivities differ largely. In the fed-batch cultivations final rTfH concentrations of 0.5 g L(â1) could be achieved. In batch cultivations the generated rTfH is translocated to the periplasm wherefrom it is completely released into the extracellular medium. In fed-batch runs most of the produced rTfH remains as soluble protein in the cytoplasm and only a fraction of about 35% is translocated to the periplasm. Migration of periplasmic proteins in the medium is obviously coupled with growth rate and this final transport step possibly plays an important role in product localization and efficacy of the Sec translocation process
