    Hepatocellular Carcinoma with Foamy Histiocyte-Like Appearance: A Deceptively Clear Cell Carcinoma Appearing Variant

    Hepatocellular carcinoma (HCC) shows many pathological features, and it varies architecturally and cytologically. There have been many reports and discussions of the morphological features of HCC. A 63-year-old man was found to have a solitary tumor in liver segment 7 that was diagnosed as HCC. A partial resection of liver segment 7 was performed. Microscopically, the tumor lesion showed a moderately differentiated HCC. There was also a lesion with foamy histiocyte-like cells corresponding to the white lesion in the face of the cut tumor. Immunohistochemical staining showed that they were negative for CD68, S-100, vimentin, and HMB-45. The cytoplasm itself was negative on periodic acid Schiff (PAS) and Sudan staining. Without immunohistological analysis, it is difficult to distinguish this HCC variant from clear cell carcinoma or metastases of renal cell carcinoma. It is important to recognize this type as a specific cytological variant of HCC that requires confirmation by immunohistochemistry. This report describes the case of a patient with a morphologically distinctive pattern of HCC with prominent cell cytoplasm that had a foamy histiocyte-like appearance.     p53 Expression in Pretreatment Specimen Predicts Response to Neoadjuvant Chemotherapy Including Anthracycline and Taxane in Patients with Primary Breast Cancer.

    While clinical and pathologic responses are important prognostic parameters, biological markers from core needle biopsy (CNB) are needed to predict neoadjuvant chemotherapy (NAC) response, to individualize treatment, and to achieve maximal efficacy. We retrospectively evaluated the cases of 183 patients with primary breast cancer who underwent surgery after NAC (anthracycline and taxane) at the National Cancer Center Hospital (NCCH). We analyzed EGFR, HER2, and p53 expression and common clinicopathological features from the CNB and surgical specimens of these patients. These biological markers were compared between sensitive patients (pathological complete response;pCR) and insensitive patients (clinical no change;cNC and clinical progressinve disease;cPD). In a comparison between the 9 (5%) sensitive patients and 30 (16%) insensitive patients, overexpression of p53 but not overexpression of either HER2 or EGFR was associated with a good response to NAC. p53 (p=0.045) and histological grade 3 (p=0.011) were important and significant predictors of the response to NAC. The correspondence rates for histological type, histological grade 3, ER, PgR, HER2, p53, and EGFR in insensitive patients between CNB and surgical specimens were 70%, 73%, 67%, 70%, 80%, 93%, and 73%. The pathologic response was significantly associated with p53 expression and histological grade 3. The correspondence rate of p53 expression between CNB and surgical specimens was higher than that of other factors. We conclude that the level of p53 expression in the CNB was an effective and reliable predictor of treatment response to NAC

    High variability of expression profiles of homeologous genes for Wnt, Hh, Notch, and Hippo signaling pathways in Xenopus laevis

    Cell signaling pathways, such as Wnt, Hedgehog (Hh), Notch, and Hippo, are essential for embryogenesis, organogenesis, and tissue homeostasis. In this study, we analyzed 415 genes involved in these pathways in the allotetraploid frog, Xenopus laevis. Most genes are retained in two subgenomes called L and S (193 homeologous gene pairs and 29 singletons). This conservation rate of homeologs is much higher than that of all genes in the X. laevis genome (86.9% vs 60.2%). Among singletons, 24 genes are retained in the L subgenome, a rate similar to the average for all genes (82.8% vs 74.6%). In addition, as general components of signal transduction, we also analyzed 32 heparan sulfate proteoglycan (HSPG)-related genes and eight TLE/Groucho transcriptional corepressors-related genes. In these gene sets, all homeologous pairs have been retained. Transcriptome analysis using RNA-seq data from developmental stages and adult tissues demonstrated that most homeologous pairs of signaling components have variable expression patterns, in contrast to the conservative expression profiles of homeologs for transcription factors. Our results indicate that homeologous gene pairs for cell signaling regulation have tended to become subfunctionalized after allotetraploidization. Diversification of signaling pathways by subfunctionalization of homeologs may enhance environmental adaptability. These results provide insights into the evolution of signaling pathways after polyploidization

    Acoustic and mechanical properties of Nankai accretionary prism core samples

    International audienceWe studied undeformed sediment and accreted strata recently recovered by Ocean Drilling Program/Integrated Ocean Drilling Program (ODP/IODP) drilling in Nankai Trough convergent margin to unravel the changes in physical properties from initial deposition to incipient deformation. We have derived acoustic (Vp) and mechanical (uniaxial poroelastic compliance, compaction amplitude) properties of samples from various drill sites along the Muroto (ODP 1173) and Kii transects (IODP C0001, C0002, C0006, and C0007) from isotropic loading tests where confining and pore pressure were independently applied. We quantified the dependence of Vp on both effective (Peff) and confining (Pc) pressure, which can be used to correct atmospheric pressure measurements of Vp. Experimental Vp obtained on core samples extrapolated to in situ conditions are slightly higher than logging-derived velocities, which can be attributed either to velocity dispersion or to the effect of large-scale faults and weak zones on waves with longer wavelength. In the high-porosity (30%-60%) tested sediments, velocities are controlled at first order by porosity and not by lithology, which is in agreement with our static measurements of drained framework incompressibility, much smaller than fluid incompressibility. Rather than framework incompressibility, shear modulus is probably the second-order control on Vp, accounting for most of the difference between actual Vp and the prediction by Wood's (1941) suspension model. We also quantified the mechanical state of Nankai samples in terms of anisotropy, diagenesis, and consolidation. Both acoustic and mechanical parameters reveal similar values in vertical and horizontal directions, attesting to the very low anisotropy of the tested material. When considering the porous samples of the Upper Shikoku Basin sediments (Site 1173) as examples of diagenetically cemented material, several mechanical and acoustic attributes appeared as reliable experimental indicators of the presence of intergrain cementation. We also detected incipient cementation in samples from IODP Site C0001 (accretionary prism unit). In terms of consolidation, we distinguished two classes of material response (shallow, deformable samples and deep, hardly deformable ones) based on the amount of compaction upon application of a Peff large with respect to the inferred in situ value, with a transition that might be related to a critical porosity

    Internal Radiation Exposure Dose in Iwaki City, Fukushima Prefecture after the Accident at Fukushima Dai-ichi Nuclear Power Plant

    As a result of the accident at the Fukushima Daiichi Nuclear Power Plant (FNPP) on 11 March 2011, a huge amount of radionuclides, including radiocesium, was released and spread over a wide area of eastern Japan. Although three years have passed since the accident, residents around the FNPP are anxious about internal radiation exposure due to radiocesium. In this study, we screened internal radiation exposure doses in Iwaki city of Fukushima prefecture, using a whole-body counter. The first screening was conducted from October 2012 to February 2013, and the second screening was conducted from May to November 2013. Study participants were employees of ALPINE and their families who underwent examination. A total of 2,839 participants (1,366 men and 1,473 women, 1-86 years old) underwent the first screening, and 2,092 (1,022 men and 1,070 women, 1-86 years old) underwent the second screening. The results showed that 99% of subjects registered below 300 Bq per body in the first screening, and all subjects registered below 300 Bq per body in the second screening. The committed effective dose ranged from 0.01-0.06 mSv in the first screening and 0.01-0.02 mSv in the second screening. Long-term follow-up studies are needed to avoid unnecessary chronic internal exposure and to reduce anxiety among the residents by communicating radiation health risks

    Characterization of the correlated background for a sterile neutrino search using the first dataset of the JSNS2^2 experiment

    JSNS2^2 (J-PARC Sterile Neutrino Search at J-PARC Spallation Neutron Source) is an experiment that is searching for sterile neutrinos via the observation of νˉμνˉe\bar{\nu}_{\mu} \to \bar{\nu}_{e} appearance oscillations using muon decay-at-rest neutrinos. Before dedicated data taking in the first-half of 2021, we performed a commissioning run for 10 days in June 2020. Using the data obtained in this commissioning run, in this paper, we present an estimate of the correlated background which imitates the νˉe\bar{\nu}_{e} signal in a sterile neutrino search. In addition, in order to demonstrate future prospects of the JSNS2^2 experiment, possible pulse shape discrimination improvements towards reducing cosmic ray induced fast neutron background are described.Comment: 7 pages, 3 figure

    Genome evolution in the allotetraploid frog Xenopus laevis

    To explore the origins and consequences of tetraploidy in the African clawed frog, we sequenced the Xenopus laevis genome and compared it to the related diploid X. tropicalis genome. We characterize the allotetraploid origin of X. laevis by partitioning its genome into two homoeologous subgenomes, marked by distinct families of ???fossil??? transposable elements. On the basis of the activity of these elements and the age of hundreds of unitary pseudogenes, we estimate that the two diploid progenitor species diverged around 34 million years ago (Ma) and combined to form an allotetraploid around 17-18 Ma. More than 56% of all genes were retained in two homoeologous copies. Protein function, gene expression, and the amount of conserved flanking sequence all correlate with retention rates. The subgenomes have evolved asymmetrically, with one chromosome set more often preserving the ancestral state and the other experiencing more gene loss, deletion, rearrangement, and reduced gene expression.ope
