    Who Are Social Entrepreneurs? Connecting the Stories of Women in the Global Textile and Apparel Industry

    Current definitions of social entrepreneurs appear limited in view, delineating social-entrepreneurs as outside elites with special qualities and their work to be complex and lavish. Definitions of social entrepreneurs fail to capture and illustrate the multitudes and diversity of social entrepreneurship. Thus, social entrepreneurship needs refashioning to address the multiple types of intentions (feasibility and desirability) to act, opportunities, and capacities. The present interpretation lacks a holistic standpoint. Using a scenario of analysis of the textile and apparel industry, it becomes evident that micro-entrepreneurs engage daily in solving the complex problem of poverty, unemployment, exploitation, and other social issues through self-employment. They are by their very nature practicing social entrepreneurship. The purpose of this concept paper is not to dispute current definitions of social entrepreneurs but to help make definitions more holistic, by recognizing the contributions of the multiple types of people and organization who attempt to solve societal concerns

    Temporal and spatial variation in temperature experienced by macrofauna at main endeavour Hydrothermal vent field

    A significant focus of hydrothermal vent ecological studies has been to understand how species cope with various stressors through physiological tolerance and biochemical resistance. Yet, the environmental conditions experienced by vent species have not been well characterized. This objective requires continuous observations over time intervals that can capture environmental variability at scales that are relevant to animals. We used autonomous temperature logger arrays (four roughly parallel linear arrays of 12 loggers spaced every 10–12 cm) to study spatial and temporal variations in the thermal regime experienced by hydrothermal vent macrofauna at a diffuse flow vent. Hourly temperatures were recorded over eight months from 2010 to 2011 at Grotto vent in the Main Endeavour vent field on the Juan de Fuca Ridge, a focus area of the Ocean Networks Canada cabled observatory. The conspicuous animal assemblages in video footage contained Ridgeia piscesae tubeworms, gastropods (primarily Lepetodrilus fucensis), and polychaetes (polynoid scaleworms and the palm worm Paralvinella palmiformis). Two dimensional spatial gradients in temperature were generally stable over the deployment period. The average temperature recorded by all arrays, and in some individual loggers, revealed distinctive fluctuations in temperature that often corresponded with the tidal cycle. We postulate that this may be related to changes in bottom currents or fluctuations in vent discharge. A marked transient temperature increase lasting over a period of days was observed in April 2011. While the distributions and behavior of Juan de Fuca Ridge vent invertebrates may be partially constrained by environmental temperature and temperature tolerance, except for the one transient high-temperature event, observed fluid temperatures were generally similar to the thermal preferences for some species, and typically well below lethal temperatures for all species. Average temperatures of the four arrays ranged from 4.1 to 11.0 °C during the deployment, indicating that on an hourly timescale the temperature conditions in this tubeworm community were fairly moderate and stable. The generality of these findings and behavioural responses of vent organisms to predictable rhythmicity and non-periodic temperature shifts are areas for further investigation

    Peroxisome proliferator-activated receptors (PPARs; PPAR-alpha, PPAR-delta, and PPAR-gamma) comprise a family of nuclear receptors that sense fatty acid levels and translate this information into altered gene transcription. Previously, it was reported that treatment of mice with a synthetic ligand activator of PPAR-delta, GW0742, ameliorates experimental autoimmune encephalomyelitis (EAE), indicating a possible role for this nuclear receptor in the control of central nervous system (CNS) autoimmune inflammation. We show that mice deficient in PPAR-delta (PPAR-delta(-/-)) develop a severe inflammatory response during EAE characterized by a striking accumulation of IFN-gamma(+)IL-17A(-) and IFN-gamma(+)IL-17A(+) CD4(+) cells in the spinal cord. The preferential expansion of these T helper subsets in the CNS of PPAR-delta(-/-) mice occurred as a result of a constellation of immune system aberrations that included higher CD4(+) cell proliferation, cytokine production, and T-bet expression and enhanced expression of IL-12 family cytokines by myeloid cells. We also show that the effect of PPAR-delta in inhibiting the production of IFN-gamma and IL-12 family cytokines is ligand dependent and is observed in both mouse and human immune cells. Collectively, these findings suggest that PPAR-delta serves as an important molecular brake for the control of autoimmune inflammation

    Obesity-Induced Colorectal Cancer Is Driven by Caloric Silencing of the Guanylin-GUCY2C Paracrine Signaling Axis.

    Obesity is a well-known risk factor for colorectal cancer but precisely how it influences risks of malignancy remains unclear. During colon cancer development in humans or animals, attenuation of the colonic cell surface receptor guanylyl cyclase C (GUCY2C) that occurs due to loss of its paracrine hormone ligand guanylin contributes universally to malignant progression. In this study, we explored a link between obesity and GUCY2C silencing in colorectal cancer. Using genetically engineered mice on different diets, we found that diet-induced obesity caused a loss of guanylin expression in the colon with subsequent GUCY2C silencing, epithelial dysfunction, and tumorigenesis. Mechanistic investigations revealed that obesity reversibly silenced guanylin expression through calorie-dependent induction of endoplasmic reticulum stress and the unfolded protein response in intestinal epithelial cells. In transgenic mice, enforcing specific expression of guanylin in intestinal epithelial cells restored GUCY2C signaling, eliminating intestinal tumors associated with a high calorie diet. Our findings show how caloric suppression of the guanylin-GUCY2C signaling axis links obesity to negation of a universal tumor suppressor pathway in colorectal cancer, suggesting an opportunity to prevent colorectal cancer in obese patients through hormone replacement with the FDA-approved oral GUCY2C ligand linaclotide

    Confirmation of the Electrostatic Self-Assembly of Nanodiamonds

    A reliable explanation for the underlying mechanism responsible for the persistent aggregation and self-assembly of colloidal 5 nm diamond nanoparticles is critical to the development of nanodiamond-based technologies. Although a number of mechanisms have been proposed, validation has been hindered by the inherent difficulty associated with the identification and characterisation of the inter-particle interfaces. In this paper we present results of high resolution aberration corrected electron microscopy and complementary computer simulations to explicate the features involved, and confirm the electrostatic interaction mechanism as the most probable cause for the formation of agglutinates and agglomerates of primary particles.Comment: 9 pages (including Supplementary Information), accepted for publication by Nanoscal

    Tenascin-C is a driver of inflammation in the DSS model of colitis

    Inflammatory Bowel Disease (IBD) is a grouping of chronic inflammatory disorders of the gut. Tenascin-C is a pro-inflammatory, extracellular matrix protein found upregulated in IBD patients and whilst a pathological driver of chronic inflammation, its precise role in the etiology of IBD is unknown. To study tenascin-C’s role in colitis pathology we investigated its expression in a murine model of IBD. Wild-type (WT) or tenascin-C knockout (KO) male mice were left untreated or treated with dextran sodium sulphate (DSS) in their drinking water. Tenascin-C was upregulated at the mRNA level in the colitic distal colon of day eight DSS treated mice, coinciding with significant increases in gross and histological pathology. Immunohistochemistry localized this increase in tenascin-C to areas of inflammation and ulceration in the mucosa. Tenascin-C KO mice exhibited reduced gross pathology in comparison. These differences also extended to the histopathological level where reduced colonic inflammation and tissue damage were found in KO compared to WT mice. Furthermore, the severity of the distal colon lesions were less in the KO mice after 17 days of recovery from DSS treatment. This study demonstrates a role for tenascin-C as a driver of inflammatory pathology in a murine model of IBD and thus suggests neutralizing its pro-inflammatory activity could be explored as a therapeutic strategy for treating IBD

    The trajectory of fidelity in a multiyear trial of the Family Check-Up predicts change in child problem behavior

    Therapist fidelity to evidence-based family interventions has consistently been linked to child and family outcomes. However, few studies evaluate the potential ebb and flow of fidelity of therapists over time. We examined therapist drift in fidelity over four years in the context of a Family Check-Up prevention services in early childhood (age 2–5). At age 2, families engaging in Women, Infants, and Children Nutritional Supplement Program (WIC) services were randomized and offered annual Family Check-Ups. Seventy-nine families with a child in the clinical range of problem behaviors at age 2 were included in this analysis. Latent growth modeling revealed a significant linear decline in fidelity over time (M = ?0.35, SD = 0.35) and steeper declines were related to less improvement in caregiver-reported problem behaviors assessed at ages 7.5/8.5 (b = ?.69, p = .003; ? = ?.95, CI: ?2.11 | ?0.22). These findings add to the literature concerning the need to continually monitor therapist fidelity to an evidence-based practice over time to optimize family benefits. Limitations and directions for future research are discussed

    Uptake of sympagic organic carbon by the Barents Sea benthos linked to sea ice seasonality

    On Arctic shelves, where primary production occurs in both the pelagic and sympagic (ice-associated) habitats, sympagic organic material (OM) can constitute a disproportionate fraction of benthic diets due to higher sinking rates and lower grazing pressure than pelagic OM. Less documented is how sympagic OM assimilation across feeding guilds varies seasonally and in relation to sea ice duation. We therefore investigated the relative abundance of sympagic vs pelagic OM in Barents Sea shelf megabenthos in the summer and winter of 2018 and 2019, from 10 stations where sea ice duration ranged from 0 to 245 days per year. We use highly branched isoprenoids, which are lipid biomarkers produced with distinct molecular structures by diatoms in sea ice and the water column, to determine the ratio of sympagic-to-pelagic OM assimilated by benthic organisms. From 114 samples of 25 taxa analysed, we found that the proportion of sympagic OM assimilated ranged from 0.4% to 95.8% and correlated strongly (r2 = 0.754) with the duration of sea ice cover. The effect of sea ice duration was more evident in fauna collected during summer than winter, indicating that sympagic signals are more evident in the summer than in the winter at higher latitudes. Our data show that sympagic production can supply a high fraction of carbon for Barents Sea benthos, although this is highly variable and likely dependent on availability and patchiness of sympagic OM deposition. These results are comparable to similar studies conducted on benthos in the Pacific Arctic and highlight the variable importance of sympagic OM in the seasonal ice zone of Arctic inflow shelves, which are the Arctic regions with highest rates of sea ice loss.publishedVersio
