Effective software testing with a string-constraint solver

Concolic testing is a paradigm of implementation-based systematic software testing that combines dynamic symbolic execution with constraint-based systematic execution-path enumeration. Concolic testing is easy to use and effective in finding real errors. It is, however, limited by the expressiveness of the underlying constraint solver. Therefore, to date, concolic testing has not been successfully applied to programs with highly-structured inputs (e.g., compilers), or to Web applications. This dissertation shows that the effectiveness and applicability of concolic testing can be greatly improved by using an expressive and efficient string-constraint solver, i.e., a solver for constraints on string variables. We present the design, implementation, and experimental evaluation of a novel string-constraint solver. Furthermore, we show novel techniques for two important problems in concolic testing: getting past input validation in programs with highly-structured inputs, and creating inputs that demonstrate security vulnerabilities in Web applications.by Adam Kieżun.Ph.D
Chemerin Effect on the Endometrial Proteome of the Domestic Pig during Implantation Obtained by LC-MS/MS Analysis
Chemerin (CHEM) is a hormone mainly expressed in adipocytes involved in the regulation of energy homeostasis and inflammatory response. CHEM expression has been demonstrated in the structures of the porcine hypothalamic-pituitary-gonadal axis, as well as in the uterus, trophoblasts and conceptuses of pigs. In this study, we performed high-throughput proteomic analyses (liquid chromatography with tandem mass spectrometry, LC-MS/MS) to examine the influence of CHEM (400 ng/mL) on differentially regulated proteins (DRPs) in the porcine endometrial tissue explants during implantation (15 to 16 days of gestation). Among all 352 DRPs, 164 were up-regulated and 188 were down-regulated in CHEM-treated group. DRPs were assigned to 47 gene ontology (GO) terms (p-adjusted < 0.05). Validation of four DRPs (IFIT5, TGFβ1, ACO1 and PGRMC1) by Western blot analysis confirmed the veracity and accuracy of the LC-MS/MS method used in the present study. We suggest that CHEM, by modulating various protein expressions, takes part in the endometrial cell proliferation, migration and invasion at the time of implantation. It also regulates the endometrial immune response, sensitivity to P4 and the formation of new blood vessels. Additionally, CHEM appears to be an important factor involved in endothelial cell dysfunction during the pathogenesis of preeclampsia. The identification of a large number of DRPs under the influence of CHEM provides a valuable resource for understanding the molecular mechanisms of this hormone action during implantation, which is a prerequisite for better control of pig reproduction

Correction ofvitamin D deficiency/insufficiency and assessment of 24-hour blood pressure parameters in perimenopausal women with arterial hypertension
Both arterial hypertension and vitamin D deficiency are highly prevalent in the general population. Aim of the study was to
assess blood pressure and its dynamics in 24-hour ambulatory blood pressure monitoring (ABPM) while correcting
the vitamin D deficiency/insufficiency in perimenopausal women, who have grade 2 arterial hypertension. Material and
methods: The study group consisted of 50 premenopausal women [48 (45–50) years] and 52 women in early (up to 5 years)
postmenopausal period [52 (50–54) years]. Total 25-(OH)D blood plasma levels were assessed by immunoenzyme assay using
DRG kit (Marburg, Germany). The total 25-(OH)D blood plasma levels below 20 ng/ml were considered as vitamin D
insufficiency, the concentration of 20–30 ng/ml – as deficiency, and the levels 30–60 ng/ml were considered normal. In the
group of women with 25-(OH)D deficiency/insufficiency two subgroups were isolated: IB subgroup (n = 25) and IIB (n = 21),
which were introduced with treatment algorithm, including cholecalciferol administration at a daily dose of 2000 IU for
3 months. Results: Cholecalciferol therapy at a dose of 2000 IU per day for a period of 3 months has allowed reaching the optimum concentration of vitamin D in 80% of premenopausal hypertensive women, and in 76.2% of women in the early
postmenopausal period. Correction of plasma vitamin D deficiency achieved by taking cholecalciferol, while using combined
antihypertensive therapy (ramipril + indapamide) allows to improve the following parameters assessed in ambulatory blood
pressure monitoring in premenopausal women: the mean daytime and nighttime systolic blood pressure, time index
of 24-hour and daytime systolic blood pressure, nighttime blood pressure variability, systolic and diastolic blood pressure
morning surge, and normalization of nighttime:daytime blood pressure ratio; in postmenopausal women the treatment
allowed: the reduction of morning surge of diastolic blood pressure, normalization of daytime systolic blood pressure
variability, and nighttime diastolic blood pressure time index
High-Dose Fenofibrate Stimulates Multiple Cellular Stress Pathways in the Kidney of Old Rats
We investigated the age-related effects of the lipid-lowering drug fenofibrate on renal stress-associated effectors. Young and old rats were fed standard chow with 0.1% or 0.5% fenofibrate. The kidney cortex tissue structure showed typical aging-related changes. In old rats, 0.1% fenofibrate reduced the thickening of basement membranes, but 0.5% fenofibrate exacerbated interstitial fibrosis. The PCR array for stress and toxicity-related targets showed that 0.1% fenofibrate mildly downregulated, whereas 0.5% upregulated multiple genes. In young rats, 0.1% fenofibrate increased some antioxidant genes’ expression and decreased the immunoreactivity of oxidative stress marker 4-HNE. However, the activation of cellular antioxidant defenses was impaired in old rats. Fenofibrate modulated the expression of factors involved in hypoxia and osmotic stress signaling similarly in both age groups. Inflammatory response genes were variably modulated in the young rats, whereas old animals presented elevated expression of proinflammatory genes and TNFα immunoreactivity after 0.5% fenofibrate. In old rats, 0.1% fenofibrate more prominently than in young animals induced phospho-AMPK and PGC1α levels, and upregulated fatty acid oxidation genes. Our results show divergent effects of fenofibrate in young and old rat kidneys. The activation of multiple stress-associated effectors by high-dose fenofibrate in the aged kidney warrants caution when applying fenofibrate therapy to the elderly
