    Quality Protein Maize Germplasm Characterized for Amino Acid Profiles and Endosperm Opacity

    Quality protein maize (QPM) is improved over normal (non-QPM) maize in grain concentrations of the essential amino acids lysine and tryptophan. Quality protein maize has a long history as tropical adapted germplasm, but little effort has been made to incorporate temperate or sub-tropical germplasm for temperate adaptation and interactions between different modifier loci in these backgrounds are poorly understood. A design-II mating scheme including new temperate and subtropical lines produced 69 hybrids. Large hybrid genetic variation components resulted in substantial broad-sense heritability H2 estimates, specifically tryptophan (0.46) and endosperm opacity (0.82). A microbial assay for amino acid estimation proved robust across diverse environments with minimal genotype Ă— environment (GĂ—E) effects. Endosperm opacity had no GĂ—E effects across both Texas and Iowa locations demonstrating stability for this trait. Endosperm opacity primarily followed an additive, midparent trend, with a few hybrids deviating from the trend (36%) suggesting a complex nature of multiple modifier loci across diverse germplasm. The top QPM hybrid outperformed the top commercial hybrid by 35 and 30% for lysine and tryptophan as a proportion of grain, respectively. QPM line Tx832 was a parent of top hybrids for lysine and tryptophan, and the highest noncommercial hybrids for methionine. Minimal correlations with yield and other traits suggest that future breeding should result in QPM hybrids with increasingly competitive yields

    Choline acetyltransferase-expressing T cells are required to control chronic viral infection.

    peer reviewedAlthough widely studied as a neurotransmitter, T cell-derived acetylcholine (ACh) has recently been reported to play an important role in regulating immunity. However, the role of lymphocyte-derived ACh in viral infection is unknown. Here, we show that the enzyme choline acetyltransferase (ChAT), which catalyzes the rate-limiting step of ACh production, is robustly induced in both CD4+ and CD8+ T cells during lymphocytic choriomeningitis virus (LCMV) infection in an IL-21-dependent manner. Deletion of Chat within the T cell compartment in mice ablated vasodilation in response to infection, impaired the migration of antiviral T cells into infected tissues, and ultimately compromised the control of chronic LCMV clone 13 infection. Our results reveal a genetic proof of function for ChAT in T cells during viral infection and identify a pathway of T cell migration that sustains antiviral immunity

    Imaging biomarker roadmap for cancer studies.

    Imaging biomarkers (IBs) are integral to the routine management of patients with cancer. IBs used daily in oncology include clinical TNM stage, objective response and left ventricular ejection fraction. Other CT, MRI, PET and ultrasonography biomarkers are used extensively in cancer research and drug development. New IBs need to be established either as useful tools for testing research hypotheses in clinical trials and research studies, or as clinical decision-making tools for use in healthcare, by crossing 'translational gaps' through validation and qualification. Important differences exist between IBs and biospecimen-derived biomarkers and, therefore, the development of IBs requires a tailored 'roadmap'. Recognizing this need, Cancer Research UK (CRUK) and the European Organisation for Research and Treatment of Cancer (EORTC) assembled experts to review, debate and summarize the challenges of IB validation and qualification. This consensus group has produced 14 key recommendations for accelerating the clinical translation of IBs, which highlight the role of parallel (rather than sequential) tracks of technical (assay) validation, biological/clinical validation and assessment of cost-effectiveness; the need for IB standardization and accreditation systems; the need to continually revisit IB precision; an alternative framework for biological/clinical validation of IBs; and the essential requirements for multicentre studies to qualify IBs for clinical use.Development of this roadmap received support from Cancer Research UK and the Engineering and Physical Sciences Research Council (grant references A/15267, A/16463, A/16464, A/16465, A/16466 and A/18097), the EORTC Cancer Research Fund, and the Innovative Medicines Initiative Joint Undertaking (grant agreement number 115151), resources of which are composed of financial contribution from the European Union's Seventh Framework Programme (FP7/2007-2013) and European Federation of Pharmaceutical Industries and Associations (EFPIA) companies' in kind contribution

    The Debate over Police Reform: Examining Minority Support for Citizen Oversight and Resistance by Police Unions

    Researchers have argued that the creation of citizen oversight often involves debate between those that support its use and the police which do not. Police unions, for example, have a long history of objecting to the creation of oversight, especially during collective bargaining. Minority demands for police reform, on the other hand, can lend support for its implementation, especially after a highly publicized case of misconduct between the police and minority citizens. Using a retrospective approach, this study examined the extent to which these opposing forces influenced the existence of oversight. Findings suggest that departments that engage in collective bargaining were no more likely to use an oversight agency than departments that did not engage in collective bargaining. Cities with large percentages of African Americans, however, were more likely to have an existing oversight agency

    Public Support for Punishment and Progressive Criminal Justice Policy Preferences: The Role of Symbolic Racism and Negative Racial Stereotype

    Prior studies have found that symbolic racism and negative African-American stereotypes are linked to public preferences for punitive criminal justice policy. But prior studies have mostly focused attention on White respondents and have not adequately examined whether the effects of symbolic racism and negative African American stereotypes are the same across race and ethnicity. This study used the 2000 American National Election Study data to fill this gap in the empirical literature. The study found that the effects of symbolic racism were broad and generally impact Whites, African-Americans, and members of other races/ethnicities the same. The effects of negative African-American stereotype were more limited. This variable predicted punishment policy preference for members of other races/ethnicities and there were significant differences in how stereotypes impacted policy preferences across race and ethnicity.     Prior research has established a strong and enduring “racial divide” in support for capital punishment, but little research has explored the processes that explain the racial divide. Following the lead of Unnever and Cullen (Social Forces 85:1281–1301, 2007a), this research explores whether racist sentiment and core values (individualism, egalitarianism, symbolic patriotism, and authoritarianism) can partially explain the racial divide in public support for capital punishment. The findings suggest that racist sentiment by Whites and belief in core values by Whites partially explains the racial divide in support for capital punishment

    Public views of illegal migration policy and control strategies: A test of the core hypotheses

    Illegal migration into the United States continues to be an important and contentious issue in the early stages of the twenty-first century. An important aspect of the contemporary migration debate is public opinion toward the various policy and control initiatives that have recently been discussed. This study used public opinion data from a 2006 study conducted by the Pew Research Center to test seven core hypotheses generated by prior academic research to explain variation in public support for cracking down on illegal migration into the United States (economic threat, culture threat, ethnic affect, core values, cultural affinity, contact, and group threat). The analysis found support for the economic threat, cultural threat, ethnic affect, core values, and cultural affinity hypotheses. The study found limited support for the contact hypothesis and no support for the group threat hypothesis. Semi-standardized coefficients were generated and suggested that the strongest predictors of support for enhanced controls on illegal migration were the cultural threat and cultural affinity measures. Implications of these findings are discussed.
