Diet-induced Obesity In Rats Leads To A Decrease In Sperm Motility.
Obesity is rapidly becoming a worldwide epidemic that affects children and adults. Some studies have shown a relationship between obesity and infertility, but until now it remains controversial. Thus, the aim of the present study was to investigate the effect of high-fat diet-induced obesity on male reproductive parameters. In a first experiment, male Wistar rats were fed a high-fat diet (HFD) or standard chow (SD) for 15, 30 or 45 weeks, after which they were evaluated by adiposity index, serum leptin levels, reproductive organ weights and sperm counts. In a second experiment, rats received HFD or SD only for 15 weeks, long enough to cause obesity. Sexual hormones and sexual behavior were evaluated in these animals, as well as fertility after natural mating. Another group of rats was submitted to motility analysis and fertility evaluation after in utero insemination. After 15, 30 or 45 weeks, HFD-fed animals presented significant increases in obesity index and serum leptin levels. Reproductive organ weights and sperm counts in the testis and epididymis were similar between the two groups at all timepoints studied. Sexual behavior was not altered by the diet regimen, and HFD fertility after natural mating was also similar to SD-fed animals. Intergroup testosterone levels were also comparable, but estradiol levels were increased in HFD rats. Furthermore, sperm quality was reduced in HFD animals as evidenced by their decreased percentage of sperm with progressive movement. This altered motility parameter was followed by a trend toward reduction in fertility potential after artificial in utero insemination. The results reported herein showed that obesity can affect sperm quality, by reducing sperm motility, without affecting other sperm parameters. The low sperm quality caused a slight reduction in fertility potential, showing that obesity may lead to impairment in male fertility.93
Fenvalerate, A Pyrethroid Insecticide, Adversely Affects Sperm Production and Storage in Male Rats
The aim of this study was to investigate the potential estrogenic activity of fenvalerate by examining reproductive and fertility capabilities in Wistar rats. Adult male animals were treated for 30 d with 20 or 40 mg/kg/d fenvalerate or corn oil (vehicle) by oral gavage. Further, a possible estrogenic activity of fenvalerate (0.4, 1, 4, 8, or 40 mg/kg) was tested after a 3-d treatment of immature female rats using the uterotrophic assay. Exposure to the higher dose of fenvalerate was toxic to testis and epididymis as shown by a decrease in the absolute weights and sperm counts in both organs. Although the sperm counts were reduced, the fertility and sexual behavior were similar in control rats and rats treated with 40 mg/kg pesticide. Fenvalerate did not exert estrogenic activity in vivo at the tested doses. Effect of essential oil from Citrus aurantium in maternal reproductive outcome and fetal anomaly frequency in rats
Citrus aurantium L., commonly known as bitter orange, is widely used in folk medicine, but there is little data in the literature about the s on pregnancy. The aim of the present study was to evaluate the influence of essential oil obtained from fruits of Citrus aurantium on the maternal reproductive outcome and fetal anomaly incidence in rats. Pregnant Wistar rats were randomized into four groups (n minimum = 12 animals/group): G1 = control, G2 to G4 = treated with essential oil from C. aurantium at dose 125, 250 and 500 mg/kg, respectively. Rats were orally treated, by gavage, with plant essential oil or vehicle during pre-implantation and organogenic period (gestational day 0-14). On gestational day 20 the rats were anaesthetized and the gravid uterus was weighed with its contents and the fetuses were analyzed. Results showed that the treated group with 500 mg/kg presented decreased placental weights and placental index, although the treatment with bitter orange essential oil did not show any alteration in maternal reproductive performance, toxicological , changes in ossification sites, and malformation index. In conclusion, the treatment of Citrus aurantium essential oil was not teratogenic and did not alter the maternal reproductive outcome.Citrus aurantium L., conhecida popularmente como laranja amarga, é amplamente utilizada na medicina popular, mas há poucos dados na literatura sobre seus efeitos na gestação. O objetivo do presente estudo foi avaliar a influencia do óleo essencial obtido das frutas de Citrus aurantium no desempenho reprodutivo materno e na incidência de anomalias fetais em ratos. Ratas Wistar prenhes foram randomizadas em quarto grupos (n mínimo = 12 animais/grupo): G1 = controle, G2 à G4 = tratados com óleo essencial de C. aurantium nas doses de 125, 250 e 500 mg/kg, respectivamente. Ratas foram tratadas oralmente, por gavage, com óleo essencial da planta ou veículo durante os períodos de pré-implantação e organogênese (dias de prenhez 0-14). No dia 20 de prenhez as ratas foram anestesiadas e o útero foi pesado com seu conteúdo e os fetos foram analisados. Resultados mostraram que o grupo tratado com 500 mg/kg apresentou diminuição do peso e índice placentário, embora o tratamento com óleo essencial de laranja amarga não mostrou nenhuma alteração no desempenho reprodutivo materno, efeito tóxico, mudanças na ossificação e nas taxas de malformações. Concluindo, o tratamento com óleo essencial de Citrus aurantium não foi teratogênico e não alterou o desempenho reprodutivo materno
Androgen Deprivation From Pre-puberty To Peripuberty Interferes In Proteins Expression In Pubertal And Adult Rat Epididymis.
Few studies have focused on experimental testosterone deprivation in immature animals. Therefore, this study used sexually immature rats aiming to evaluate the testes and epididymis histology and proteins expression in these organs on PND50 and 75, after premature antiandrogen exposure, from PND21 to 44. Although the androgen deprivation from pre-puberty up to peripuberty did not alter the histological organization of the testes and epididymis either at puberty or at adulthood, the treatment impaired the expression of specific proteins in epididymal tissue at puberty and adulthood (androgen receptor, calmodulin, Rab11A). These changes may be related to impaired epididymal function, sperm quality and fertility capacity as observed in a previous study. Further studies are necessary to better investigate the molecular mechanisms involved in the impairment on reproductive competence of male rats after precocious hormonal injury.3865-7
Can vitamins C and E restore the androgen level and hypersensitivity of the vas deferens in hyperglycemic rats?
Diabetic neuropathy can affect the male reproductive system. The aim of this study was therefore to evaluate whether antioxidant (vitamins C and/or E) treatment could attenuate reproductive dysfunctions in hyperglycemic adult male rats. The animals were randomly assigned to one of four experimental groups: hyperglycemic control (Hy), hyperglycemic + 150 mg/day vitamin C (HyC), hyperglycemic + 100 mg/day vitamin E (HyE) or hyperglycemic + vitamins C and E (HyCE). The normoglycemic group (n = 10) received only the vehicles. The testosterone level and noradrenergic response of the vas deferens were analyzed. Both vitamins significantly decreased the TBARS (thiobarbituric acid reactive species) level in the hyperglycemic groups. There was a significant reduction in the testosterone level in the Hy and HyE groups when compared to the normoglycemic group. However, the testosterone levels were partially recovered in the HyC and HyCE groups. In addition, an increased sensitivity of the alpha-1 adrenoceptor in the vas deferens of the hyperglycemic control group was observed. Treatment with vitamins partially restored (vitamin E or in combination with vitamin C) or totally (vitamin C alone) this dysfunction. Moreover, the maximum response values to norepinephrine were similar among all groups. This paper addresses the questions, perplexities and results of an ongoing project that involves mixed methods analysis and that has as its main purpose to understand the role of the communication professionals in the non for profit organisations in Portugal. The specific research questions that this paper addresses may be summarised as an interrogation about the role that the communication professionals are called to fulfill in these institutions.
