    Effect of Conditioned Medium on the Recovery of Dormant Mycobacteria in Culture

    Mycobacterium avium subsp. paratuberculosis (Mpt) is the etiologic agent of Johne’s disease, a chronic intestinal disease in cattle that threatens the economic viability of dairy farming. Diagnostic culture is typically unrewarding until several years after infection when clinical signs can be observed. This leads to widespread infection within the herd. Difficulty in culturing Mpt may be a result of oxidative damage due to the increased metabolic rate when dormant organisms are recovered in a nutrient rich medium. In order to improve recovery it is believed that some organisms secrete a growth factor in times of environmental stress which enables them to grow more quickly when conditions improve. The purpose of the project was to test conditioned medium (CM) and its components as a method of improving the recovery of Mpt. The conditioned medium from Mpt was separated into 4 fractions based on molecular weight using centrifuge filters. Unsupplemented media (Middlebrook 7H9C or Luria-Bertani broth) and those containing serial dilutions of fractionated or unfractionated Mpt-conditioned medium were inoculated with Mpt or Mycobacterium smegatis (MS); a non-pathogenic species. Growth was monitored by measuring the optical density of the cultures for up to 30 days. Unfractionated Mpt-conditioned medium promoted a two fold or greater enhancement of Mpt growth, but had no effect on that of MS. Results of treatment with conditioned medium fractions are pending.     Mycobacterium avium subsp. paratuberculosis (MAP) is the etiologic agent of Johne’s disease, a chronic intestinal disease of cattle and other ruminants. Diagnostic culture for MAP is typically unrewarding until latter stages of the disease. This may be a consequence of oxidative damage to dormant organisms, which results from culture. The purpose of this investigation was to determine the effect of the commercial reducing agent Oxyrase® for Broth (OB) on the recovery of dormant Mycobacterium smegmatis (MS) and MAP. Dormant organisms were inoculated into medium only or that supplemented with media containing serial dilutions of OB. Growth was monitored by optical density for up to 21 days. Treatment of MAP and MS with OB led to significant increases in recovery and growth yield. However, the concentration of OB necessary to promote recovery was dependent on the number of viable organisms present in seed cultures. Mitigating oxidative damage by using OB can facilitate the recovery of dormant mycobacteria. Whether this is a direct or indirect effect has yet to be be established

    Transverse momentum dependence of semi-inclusive pion production

    Cross sections for semi-inclusive electroproduction of charged pions (π±\pi^{\pm}) from both proton and deuteron targets were measured for 0.2<x<0.50.2<x<0.5, 2<Q2<42<Q^2<4 GeV2^2, 0.3<z<10.3<z<1, and Pt2<0.2P_t^2<0.2 GeV2^2. For Pt<0.1P_t<0.1 GeV, we find the azimuthal dependence to be small, as expected theoretically. For both π+\pi^+ and π−\pi^-, the PtP_t dependence from the deuteron is found to be slightly weaker than from the proton. In the context of a simple model, this implies that the initial transverse momenta width of dd quarks is larger than for uu quarks and, contrary to expectations, the transverse momentum width of the favored fragmentation function is larger than the unfavored one.Comment: 15 pages, 4 figures.     A large data set of charged-pion electroproduction from both hydrogen and deuterium targets has been obtained spanning the low-energy residual-mass region. These data conclusively show the onset of the quark-hadron duality phenomenon, as predicted for high-energy hadron electroproduction.     Children struggle beyond preschool-age in a continuous version of the ambiguous figures task

    Children until the age of five are only able to reverse an ambiguous figure when they are informed about the second interpretation. In two experiments, we examined whether children’s difficulties would extend to a continuous version of the ambiguous figures task. Children (Experiment 1: 66 3- to 5-year olds; Experiment 2: 54 4- to 9-year olds) and adult controls saw line drawings of animals gradually morph—through well-known ambiguous figures—into other animals. Results show a relatively late developing ability to recognize the target animal, with difficulties extending beyond preschool-age. This delay can neither be explained with improvements in theory of mind, inhibitory control, nor individual differences in eye movements. Even the best achieving children only started to approach adult level performance at the age of 9, suggesting a fundamentally different processing style in children and adults

    Relativistic Structure of the Deuteron: 1.Electro-disintegration and y-scaling

    Realistic solutions of the spinor-spinor Bethe-Salpeter equation for the deuteron with realistic interaction kernel including the exchange of pi, sigma, omega, rho, eta and delta mesons, are used to systematically investigate relativistic effects in inclusive quasi-elastic electron-deuteron scattering within the relativistic impulse approximation. Relativistic y-scaling is considered by generalising the non relativistic scaling function to the relativistic case, and it is shown that y-scaling does occur in the usual relativistic scaling variable resulting from the energy conservation in the instant form of dynamics. The present approach of y-scaling is fully covariant, with the deuteron being described by eight components, viz. the 3S_1^{++}, 3S_1^{--}, 3D_1^{++}, 3D_1^{--}, 3P_1^{+-}, 3P_1^{-+}, 1P_1^{+-}, 1P_1^{-+} waves. It is demonstrated that if the negative relative energy states 1P_1, 3P_1 are disregarded, the concept of covariant momentum distributions N(p_0,p), with p_0=M_D/2-\sqrt{p^2+m^2}, can be introduced, and that calculations of lectro-disintegration cross section in terms of these distributions agree within few percents with the exact calculations which include the 1P_1, 3P_1 states, provided the nucleon three momentum |p|\<= 1 GeV/c; in this momentum range, the asymptotic relativistic scaling function is shown to coincide with the longitudinal covariant momentum distribution.Comment: 32 LaTeX pages, 18 eps-figures.     Quantitative analyses and modelling to support achievement of the 2020 goals for nine neglected tropical diseases

    Quantitative analysis and mathematical models are useful tools in informing strategies to control or eliminate disease. Currently, there is an urgent need to develop these tools to inform policy to achieve the 2020 goals for neglected tropical diseases (NTDs). In this paper we give an overview of a collection of novel model-based analyses which aim to address key questions on the dynamics of transmission and control of nine NTDs: Chagas disease, visceral leishmaniasis, human African trypanosomiasis, leprosy, soil-transmitted helminths, schistosomiasis, lymphatic filariasis, onchocerciasis and trachoma. Several common themes resonate throughout these analyses, including: the importance of epidemiological setting on the success of interventions; targeting groups who are at highest risk of infection or re-infection; and reaching populations who are not accessing interventions and may act as a reservoir for infection,. The results also highlight the challenge of maintaining elimination ‘as a public health problem’ when true elimination is not reached. The models elucidate the factors that may be contributing most to persistence of disease and discuss the requirements for eventually achieving true elimination, if that is possible. Overall this collection presents new analyses to inform current control initiatives. These papers form a base from which further development of the models and more rigorous validation against a variety of datasets can help to give more detailed advice. At the moment, the models’ predictions are being considered as the world prepares for a final push towards control or elimination of neglected tropical diseases by 2020

    Electroproduction of Eta Mesons in the S11(1535) Resonance Region at High Momentum Transfer

    The differential cross-section for the process p(e,e'p)eta has been measured at Q2 ~ 5.7 and 7.0 (GeV/c)2 for centre-of-mass energies from threshold to 1.8 GeV, encompassing the S11(1535) resonance, which dominates the channel. This is the highest momentum transfer measurement of this exclusive process to date. The helicity-conserving transition amplitude A_1/2, for the production of the S11(1535) resonance, is extracted from the data.
