Objek dari penelitian ini adalah sebuah perusahaan yang bergerak di bidang migas dan penelitian ini menggunakan jenis penelitian kualitatif dengan menggunakan pendekatan deskriptif. Terdapat banyak faktor yang dapat mempengaruhi proses negosiasi dalam bisnis seperti pada budaya, cara berkomunikasi dan lain-lain. Penelitian ini menunjukkan bahwa dengan pemahaman lintas budaya ini dapat mencapai keberhasilan dalam proses negosiasi bisnis

    Costs and benefits of new apprenticeships

    The introduction of New Apprenticeships in January of 1998 marked an important initiative in Australia's efforts to develop a more encompassing system of employment based vocational education and training. This paper draws on evidence from case studies of 60 employers to assess the impact of these reforms on training outcomes and practices. The provisions most enthusiastically embraced by employers include competency-based assessment and flexibility in time spent in training. However, these positive indicators of the reform process have not translated into an improved cost/benefit outcome for employers. Comparison with previous findings suggests that the net cost of employing trainees has not really changed while the net cost of apprentices is estimated to be higher than it was in 1996

    Flashlights: Properties of Highly Magnified Images Near Cluster Critical Curves in the Presence of Dark Matter Subhalos

    Dark matter subhalos with extended profiles and density cores, and globular stars clusters of mass 106−108M⊙10^6-10^8 M_\odot, that live near the critical curves in galaxy cluster lenses can potentially be detected through their lensing magnification of stars in background galaxies. In this work we study the effect such subhalos have on lensed images, and compare to the case of more well studied microlensing by stars and black holes near critical curves. We find that the cluster density gradient and the extended mass distribution of subhalos are important in determining image properties. Both lead to an asymmetry between the image properties on the positive and negative parity sides of the cluster that is more pronounced than in the case of microlensing. For example, on the negative parity side, subhalos with cores larger than about 50 50\,pc do not generate any images with magnification above ∼100\sim 100 outside of the immediate vicinity of the cluster critical curve. We discuss these factors using analytical and numerical analysis, and exploit them to identify observable signatures of subhalos: subhalos create pixel-to-pixel flux variations of ≳0.1\gtrsim 0.1 magnitudes, on the positive parity side of clusters. These pixels tend to cluster around (otherwise invisible) subhalos. Unlike in the case of microlensing, signatures of subhalo lensing can be found up to 1′′1'' away from the critical curves of massive clusters.Comment: ApJ, submitted, 21 pages, 17 figure

    Novel Interconnections in Lipid Metabolism Revealed by Overexpression of Sphingomyelin Synthase-1

    This study investigates the consequences of elevating sphingomyelin synthase 1 (SMS1) activity, which generates the main mammalian sphingolipid, sphingomyelin. HepG2 cells stably transfected with SMS1 (HepG2-SMS1) exhibit elevated enzyme activity in vitro and increased sphingomyelin content (mainly C22:0- and C24:0-sphingomyelin) but lower hexosylceramide (Hex-Cer) levels. HepG2-SMS1 cells have fewer triacylglycerols than controls but similar diacylglycerol acyltransferase activity, triacylglycerol secretion, and mitochondrial function. Treatment with 1 mm palmitate increases de novo ceramide synthesis in both cell lines to a similar degree, causing accumulation of C16:0-ceramide (and some C18:0-, C20:0-, and C22:0-ceramides) as well as C16:0- and C18:0-Hex-Cers. In these experiments, the palmitic acid is delivered as a complex with delipidated BSA (2:1, mol/mol) and does not induce significant lipotoxicity. Based on precursor labeling, the flux through SM synthase also increases, which is exacerbated in HepG2-SMS1 cells. In contrast, palmitate-induced lipid droplet formation is significantly reduced in HepG2-SMS1 cells. [14C]Choline and [3H]palmitate tracking shows that SMS1 overexpression apparently affects the partitioning of palmitate-enriched diacylglycerol between the phosphatidylcholine and triacylglycerol pathways, to the benefit of the former. Furthermore, triacylglycerols from HepG2-SMS1 cells are enriched in polyunsaturated fatty acids, which is indicative of active remodeling. Together, these results delineate novel metabolic interactions between glycerolipids and sphingolipids
