    Complex history and differentiation patterns of the t-haplotype, a mouse meiotic driver

    The t-haplotype, a mouse meiotic driver found on chromosome 17, has been a model for autosomal segregation distortion for close to a century, but several questions remain regarding its biology and evolutionary history. A recently published set of population genomics resources for wild mice includes several individuals heterozygous for the t-haplotype, which we use to characterize this selfish element at the genomic and transcriptomic level. Our results show that large sections of the t-haplotype have been replaced by standard homologous sequences, possibly due to occasional events of recombination, and that this complicates the inference of its history. As expected for a long genomic segment of very low recombination, the t-haplotype carries an excess of fixed nonsynonymous mutations compared to the standard chromosome. This excess is stronger for regions that have not undergone recent recombination, suggesting that occasional gene flow between the t and the standard chromosome may provide a mechanism to regenerate coding sequences that have accumulated deleterious mutations. Finally, we find that t-complex genes with altered expression largely overlap with deleted or amplified regions, and that carrying a t-haplotype alters the testis expression of genes outside of the t-complex, providing new leads into the pathways involved in the biology of this segregation distorter

    Many studies suggest that balanced budget rules can restrain sovereign debt and lower sovereign borrowing costs, even if those rules are never enforced in court. Typically, this is explained as a result of a legal deterrence logic, in which the threat of judicial enforcement deters sovereigns from violating the rules. By contrast, we argue that balanced budget rules work by coordinating decentralized punishment of sovereigns by bond markets, rather than by posing a credible threat of judicial enforcement. Therefore, the clarity of the focal point provided by the rule, rather than the strength of its judicial enforcement mechanisms, determines its effectiveness. We develop a formal model that captures the logic of our argument, and we assess this model using data on US states. We then consider implications of our argument for the impact of the balanced budget rules recently imposed on eurozone states in the Fiscal Compact Treaty

    Novel patterns of expression and recruitment of new genes on the t-haplotype, a mouse selfish chromosome

    The t-haplotype of mice is a classical model for autosomal transmission distortion. A largely non-recombining variant of the proximal region of chromosome 17, it is transmitted to more than 90% of the progeny of heterozygous males through the disabling of sperm carrying a standard chromosome. While extensive genetic and functional work has shed light on individual genes involved in drive, much less is known about the evolution and function of the rest of its hundreds of genes. Here, we characterize the sequence and expression of dozens of t-specific transcripts and of their chromosome 17 homologues. Many genes showed reduced expression of the t-allele, but an equal number of genes showed increased expression of their t-copy, consistent with increased activity or a newly evolved function. Genes on the t-haplotype had a significantly higher non-synonymous substitution rate than their homologues on the standard chromosome, with several genes harbouring dN/dS ratios above 1. Finally, the t-haplotype has acquired at least two genes from other chromosomes, which show high and tissue-specific expression. These results provide a first overview of the gene content of this selfish element, and support a more dynamic evolutionary scenario than expected of a large genomic region with almost no recombination

    Dynamic response of herbaceous vegetation to gap opening in a Central European beech stand

    Herbaceous ground vegetation in artificially-created gaps was studied in a managed beech (Fagus sylvatica L.) forest over a period of eight years in Northern Hungary, Central Europe. These gaps were being used as an alternative to the regular shelterwood system to create uneven-aged stands. The effects of gap size (15 and 40 m diameter) and canopy openness on herbaceous species colonization and persistence were assessed in a systematic grid of 5 5 m. Overall, herbaceous cover was low before gap creation, increased soon afterwards, and continued to rise over time. The number of herb species increased in the gaps and, to a lesser extent, in adjacent areas under the remaining tree canopy. Colonization of gaps was rapid and there was substantial turnover of species i.e. various species disappeared from the gaps over time whilst others colonized. Species with both long-term persistent seed banks and long distance dispersal abilities were the most successful types colonizing gaps. Six species occurred preferentially in large gaps, while only one species was found to prefer small gaps. Species present before gap creation survived in both gap sizes. Smaller gaps with a diameter of half the height of canopy trees also tended to remain free of common weed species, whereas large cover of Rubus fruticosus L. and Calamagrostis epigejos (L.) Roth could hamper natural regeneration in larger gaps. For the successful regeneration of beech we recommend the use of small gaps complemented by few large gaps.</ja:p

    Clustering of activated CD8 T cells around Malaria-infected hepatocytes is rapid and is driven by antigen-specific cells

    Malaria, a disease caused by parasites of the Plasmodium genus, begins when Plasmodium-infected mosquitoes inject malaria sporozoites while searching for blood. Sporozoites migrate from the skin via blood to the liver, infect hepatocytes, and form liver stages which in mice 48 h later escape into blood and cause clinical malaria. Vaccine-induced activated or memory CD8 T cells are capable of locating and eliminating all liver stages in 48 h, thus preventing the blood-stage disease. However, the rules of how CD8 T cells are able to locate all liver stages within a relatively short time period remains poorly understood. We recently reported formation of clusters consisting of variable numbers of activated CD8 T cells around Plasmodium yoelii (Py)-infected hepatocytes. Using a combination of experimental data and mathematical models we now provide additional insights into mechanisms of formation of these clusters. First, we show that a model in which cluster formation is driven exclusively by T-cell-extrinsic factors, such as variability in “attractiveness” of different liver stages, cannot explain distribution of cluster sizes in different experimental conditions. In contrast, the model in which cluster formation is driven by the positive feedback loop (i.e., larger clusters attract more CD8 T cells) can accurately explain the available data. Second, while both Py-specific CD8 T cells and T cells of irrelevant specificity (non-specific CD8 T cells) are attracted to the clusters, we found no evidence that non-specific CD8 T cells play a role in cluster formation. Third and finally, mathematical modeling suggested that formation of clusters occurs rapidly, within few hours after adoptive transfer of CD8 T cells, thus illustrating high efficiency of CD8 T cells in locating their targets in complex peripheral organs, such as the liver. Taken together, our analysis provides novel insights into and attempts to discriminate between alternative mechanisms driving the formation of clusters of antigen-specific CD8 T cells in the liver

    Activity of 50 Long-Period Comets Beyond 5.2 AU

    Remote investigations of the ancient solar system matter has been traditionally carried out through the observations of long-period (LP) comets that are less affected by solar irradiation than the short-period counterparts orbiting much closer to the Sun. Here we summarize the results of our decade-long survey of the distant activity of LP comets. We found that the most important separation in the dataset is based on the dynamical nature of the objects. Dynamically new comets are characterized by a higher level of activity on average: the most active new comets in our sample can be characterized by afrho values >3--4 higher than that of our most active returning comets. New comets develop more symmetric comae, suggesting a generally isotropic outflow. Contrary to this, the coma of recurrent comets can be less symmetrical, ocassionally exhibiting negative slope parameters, suggesting sudden variations in matter production. The morphological appearance of the observed comets is rather diverse. A surprisingly large fraction of the comets have long, teniouos tails, but the presence of impressive tails does not show a clear correlation with the brightness of the comets.Comment: 21 pages, 4 figures, accepted for publication in A

    Overtone and multi-mode RR Lyrae stars in the globular cluster M3

    The overtone and multi-mode RR Lyrae stars in the globular cluster M3 are studied using a 200-d long, B,VB,V and ICI_{\mathrm C} time-series photometry obtained in 2012. 70\% of the 52 overtone variables observed show some kind of multi-periodicity (additional frequency at f0.61=f1O/0.61{f_{0.61}}={f_{\mathrm {1O}}}/0.61 frequency ratio, Blazhko effect, double/multi-mode pulsation, period doubling). A signal at 0.587 frequency ratio to the fundamental-mode frequency is detected in the double-mode star, V13, which may be identified as the second radial overtone mode. If this mode-identification is correct, than V13 is the first RR Lyrae star showing triple-mode pulsation of the first three radial modes. Either the Blazhko effect or the f0.61{f_{0.61}} frequency (or both of these phenomena) appear in 7 double-mode stars. The P1O/PFP_{\mathrm{1O}}/P_{\mathrm{F}} period ratio of RRd stars showing the Blazhko effect are anomalous. A displacement of the main frequency component at the fundamental-mode with the value of modulation frequency (or its half) is detected in three Blazhko RRd stars parallel with the appearance of the overtone-mode pulsation. The f0.61{f_{0.61}} frequency appears in RRc stars that lie at the blue side of the double-mode region and in RRd stars, raising the suspicion that its occurrence may be connected to double-mode pulsation. The changes of the Blazhko and double-mode properties of the stars are also reviewed using the recent and archive photometric data.Comment: accepted for publication in ApJ Suppl. 26 pages, 25 figure

    Environmentally conscious consumption patterns in Hungarian households

    This article provides a comprehensive review of the literature on the theoretical aspects of sustainable consumption. The conditions for consumers’ social responsibility and the formation of environmentally conscious behavior patterns will also be discussed, along with possible methods for motivating behavioral changes. The authors have completed a primary research study with the purpose of surveying environmentally conscious consumption patterns in Hungary. They also examined how the provision of appropriate information and the raising of awareness might encourage sustainable consumption. According to their findings, the respondents’ knowledge on environmentally conscious behavior was rather limited, and reinforcement was needed in identifying appropriate activity alternatives. This paper provides a summary of the qualitative research phase which employed in-depth interviews, logging and focus groups. The consecutive application of these methods enabled the authors to keep track of the process and the consequences of raising awareness

    During the last few years high power diode laser arrays have become well established for direct material processing due to their high efficiency of more than 50%. But standard broad-area waveguide designs are susceptible to modal instabilities and filamentations resulting in low beam qualities. The beam quality increases by more than a factor of four by using tapered laser arrays, but so far they suffer from lower efficiencies. Therefore tapered lasers are mainly used today as single emitters in external resonator configurations. With increased output power and lifetime, they will be much more attractive for material processing and for pumping of fiber amplifiers. High efficiency tapered mini bars emitting at a wavelength of 980 nm are developed, and in order to qualify the bars, the characteristics of single emitters and mini bars from the same wafer have been compared. The mini bars have a width of 6 mm with 12 emitters. The ridge waveguide tapered lasers consist of a 500 µm long ridge and a 2000 µm long tapered section. The results show very similar behavior of the electro-optical characteristics and the beam quality for single emitters and bars. Due to different junction temperatures, different slope efficiencies were measured: 0.8 W/A for passively cooled mini bars and 1.0 W/A for actively cooled mini-bars and single emitters. The threshold current of 0.7 A per emitter is the same for single emitters and emitter arrays. Output powers of more than 50 W in continuous wave mode for a mini bar with standard packaging demonstrates the increased power of tapered laser bars
