Wave function-dependent mobility and suppression of interface roughness scattering in a strained SiGe p-channel field-effect structure

    Get PDF
The 4 K Hall mobility has been measured in a top-gated, inverted, modulation-doped Si/Si0.8Ge0.2 structure having a Si:B doping layer beneath the alloy. From comparisons with theoretical calculations, we argue that, unlike an ordinary enhancement-mode SiGe p-channel metal–oxide–semiconductor structure, this configuration leads to a decrease of interface roughness scattering with increasing sheet carrier density. We also speculate on the nature of the interface charge observed in these structures at low temperature

An engineered Tetrahymena tRNA(Gln) for in vivo incorporation of unnatural amino acids into proteins by nonsense suppression

    Get PDF
A new tRNA, THG73, has been designed and evaluated as a vehicle for incorporating unnatural amino acids site-specifically into proteins expressed in vivo using the stop codon suppression technique. The construct is a modification of tRNAGln(CUA) from Tetrahymena thermophila, which naturally recognizes the stop codon UAG. Using electrophysiological studies of mutations at several sites of the nicotinic acetylcholine receptor, it is established that THG73 represents a major improvement over previous nonsense suppressors both in terms of efficiency and fidelity of unnatural amino acid incorporation. Compared with a previous tRNA used for in vivo suppression, THG73 is as much as 100-fold less likely to be acylated by endogenous synthetases of the Xenopus oocyte. This effectively eliminates a major concern of the in vivo suppression methodology, the undesirable incorporation of natural amino acids at the suppression site. In addition, THG73 is 4-10-fold more efficient at incorporating unnatural amino acids in the oocyte system. Taken together, these two advances should greatly expand the range of applicability of the in vivo nonsense suppression methodology

Increased gravitational force reveals the mechanical, resonant nature of physiological tremor

    Get PDF
Human physiological hand tremor has a resonant component. Proof of this is that its frequency can be modified by adding mass. However, adding mass also increases the load which must be supported. The necessary force requires muscular contraction which will change motor output and is likely to increase limb stiffness. The increased stiffness will partly offset the effect of the increased mass and this can lead to the erroneous conclusion that factors other than resonance are involved in determining tremor frequency. Using a human centrifuge to increase head-to-foot gravitational field strength, we were able to control for the increased effort by increasing force without changing mass. This revealed that the peak frequency of human hand tremor is 99% predictable on the basis of a resonant mechanism. We ask what, if anything, the peak frequency of physiological tremor can reveal about the operation of the nervous system. This work was funded by a BBSRC Industry Interchange Award to J.P.R.S. and R.F.R. Growth pattern to the end of the mating period influences the reproductive performance of Merino ewe lambs mated at 7 to 8 months of age

The reproductive performance of Merino ewe lambs is highly variable and generally poor in comparison to older ewes. In this study, we determined the impacts of growth pattern to the end of the mating period and sire genetics on the reproductive performance of Merino ewe lambs. Five hundred ewe lambs with full pedigree records were managed under commercial conditions from weaning and weighed 43.5 kg at the start of the mating period with an average age of 224 days. The ewe lambs were offered a moderate or high feed allowance to achieve target growth rates of 100 or 200 g/day during a 46-day mating period. They were then recombined and scanned for pregnancy status 60 days after the mating period. At the individual animal level, a 5 kg greater live weight at the start of the mating period increased reproductive rate (foetuses per 100 ewes joined) by about 20% (P < 0.001). Regardless of their live weight at the start of the mating period, gaining an extra 100 g/day of live weight during the mating period further increased their reproductive rate by about 20% (P < 0.001). Ewe lambs from sires with higher Australian Sheep Breeding Values for fat measured post-weaning achieved a higher fertility (P < 0.05) and reproductive rate (P < 0.01) regardless of feed allowance treatment. The effects of sire fatness was significant even when the sire breeding values for live weight measured post-weaning were included in the model. The effects of managing growth pattern and sire genetics were additive, so improving the reproductive performance of Merino ewe lambs mated at 7 to 8 months of age requires improving their feed allowance and rate of live weight gain until the end of the mating period and using sires with higher breeding values for fatness

On the Inelastic Collapse of a Ball Bouncing on a Randomly Vibrating Platform

We study analytically the dynamics of a ball bouncing inelastically on a randomly vibrating platform, as a simple toy model of inelastic collapse. Of principal interest are the distributions of the number of flights n_f till the collapse and the total time \tau_c elapsed before the collapse. In the strictly elastic case, both distributions have power law tails characterised by exponents which are universal, i.e., independent of the details of the platform noise distribution. In the inelastic case, both distributions have exponential tails: P(n_f) ~ exp[-\theta_1 n_f] and P(\tau_c) ~ exp[-\theta_2 \tau_c]. The decay exponents \theta_1 and \theta_2 depend continuously on the coefficient of restitution and are nonuniversal; however as one approches the elastic limit, they vanish in a universal manner that we compute exactly. An explicit expression for \theta_1 is provided for a particular case of the platform noise distribution.Comment: 32 page

Human exercise-induced circulating progenitor cell mobilization is nitric oxide-dependent and is blunted in South Asian men

This article is available open access through the publisher’s website. Copyright @ 2010 American Heart Foundation. Objective— Circulating progenitor cells (CPC) have emerged as potential mediators of vascular repair. In experimental models, CPC mobilization is critically dependent on nitric oxide (NO). South Asian ethnicity is associated with reduced CPC. We assessed CPC mobilization in response to exercise in Asian men and examined the role of NO in CPC mobilization per se. Methods and Results— In 15 healthy, white European men and 15 matched South Asian men, CPC mobilization was assessed during moderate-intensity exercise. Brachial artery flow-mediated vasodilatation was used to assess NO bioavailability. To determine the role of NO in CPC mobilization, identical exercise studies were performed during intravenous separate infusions of saline, the NO synthase inhibitor l-NMMA, and norepinephrine.  Flow-mediated vasodilatation (5.8%±0.4% vs 7.9%±0.5%; P=0.002) and CPC mobilization (CD34+/KDR+ 53.2% vs 85.4%; P=0.001; CD133+/CD34+/KDR+ 48.4% vs 73.9%; P=0.05; and CD34+/CD45− 49.3% vs 78.4; P=0.006) was blunted in the South Asian group. CPC mobilization correlated with flow-mediated vasodilatation and l-NMMA significantly reduced exercise-induced CPC mobilization (CD34+/KDR+ −3.3% vs 68.4%; CD133+/CD34+/KDR+ 0.7% vs 71.4%; and CD34+/CD45− −30.5% vs 77.8%; all P<0.001). Conclusion— In humans, NO is critical for CPC mobilization in response to exercise. Reduced NO bioavailability may contribute to imbalance between vascular damage and repair mechanisms in South Asian men.British Heart Foundatio
