Src Homology 2–containing 5-Inositol Phosphatase (SHIP) Suppresses an Early Stage of Lymphoid Cell Development through Elevated Interleukin-6 Production by Myeloid Cells in Bone Marrow
The Src homology (SH)2–containing inositol 5-phosphatase (SHIP) negatively regulates a variety of immune responses through inhibitory immune receptors. In SHIP(−/−) animals, we found that the number of early lymphoid progenitors in the bone marrow was significantly reduced and accompanied by expansion of myeloid cells. We exploited an in vitro system using hematopoietic progenitors that reproduced the in vivo phenotype of SHIP(−/−) mice. Lineage-negative marrow (Lin(−)) cells isolated from wild-type mice failed to differentiate into B cells when cocultured with those of SHIP(−/−) mice. Furthermore, culture supernatants of SHIP(−/−) Lin(−) cells suppressed the B lineage expansion of wild-type lineage-negative cells, suggesting the presence of a suppressive cytokine. SHIP(−/−) Lin(−) cells contained more IL-6 transcripts than wild-type Lin(−) cells, and neutralizing anti–IL-6 antibody rescued the B lineage expansion suppressed by the supernatants of SHIP(−/−) Lin(−) cells. Finally, we found that addition of recombinant IL-6 to cultures of wild-type Lin(−) bone marrow cells reproduced the phenotype of SHIP(−/−) bone marrow cultures: suppression of B cell development and expansion of myeloid cells. The results identify IL-6 as an important regulatory cytokine that can suppress B lineage differentiation and drive excessive myeloid development in bone marrow
Effect of Students’ Reflection on their Teaching Practice Experiences: Discussions during Case Conferences in Teaching Practice
The purpose of this study was to clarify the effect of reflecting by student A, who participated in a teaching practice. As a result, the following three points were clarified. First, student A made more comments at case conferences than student teacher B. Moreover, student A made more comments concerning “alternatives” in which students’ reflections appear in case conferences than student teacher B. Second, the students’ reflections at case conferences revealed a commonly shared viewpoint on “teaching skills”. Student A also held the viewpoint of an “approach to the objective” and that the content of the class was appropriate to the goal of the lesson. Regarding the difference between student A and student teacher B, it was suggested that the mentoring received by student A from mentor D during the teaching practice had some degree of influence. Third, both student A and student teacher B has difficulties with “schoolchild understanding” despite mentor D talking at case conferences about this issue. Based on these findings, it is suggested that it is difficult to engage in reflection that enables student to comprehend “schoolchild understanding” in teaching practice

Opossum APOBEC1 is a DNA mutator with retrovirus and retroelement restriction activity
APOBEC3s (A3s) are single-stranded DNA cytosine deaminases that provide innate immune defences against retroviruses and mobile elements. A3s are specific to eutherian mammals because no direct homologs exist at the syntenic genomic locus in metatherian (marsupial) or prototherian (monotreme) mammals. However, the A3s in these species have the likely evolutionary precursors, the antibody gene deaminase AID and the RNA/DNA editing enzyme APOBEC1 (A1). Here, we used cell culture-based assays to determine whether opossum A1 restricts the infectivity of retroviruses including human immunodeficiency virus type 1 (HIV-1) and the mobility of LTR/non-LTR retrotransposons. Opossum A1 partially inhibited HIV-1, as well as simian immunodeficiency virus (SIV), murine leukemia virus (MLV), and the retrotransposon MusD. The mechanism of inhibition required catalytic activity, except for human LINE1 (L1) restriction, which was deamination-independent. These results indicate that opossum A1 functions as an innate barrier to infection by retroviruses such as HIV-1, and controls LTR/non-LTR retrotransposition in marsupials
