    Development and validation of a UPLC method for quantification of antiviral agent, Acyclovir in lipid-based formulations

    AbstractPurposeThe objective of the current study is to evaluate the Ultra Performance Liquid Chromatography (UPLC) method for quantification of Acyclovir in lipid-based formulations.MethodA simple, rapid, reliable and precise reversed phase UPLC method has been developed and validated according to the regulatory guidelines, which composed of isocratic mobile phase; 0.25% formic acid (FA) in Milli-Q water with a flow rate of 0.5ml/min, and column BEH C18 (2.1×50mm, 1.7μm). The detection was carried out at 254nm.ResultsThe developed UPLC method was found to be rapid (1.2min run time), selective with well resoluted Acyclovir peak (0.89min) from different lipid matrices and sensitive (Limit of Detection (LOD) was 0.3ppm and Lower Limit of Quantification (LLOQ) was 1ppm). The accuracy and precision were determined and were perfectly matching with the standard FDA limits.ConclusionThe study showed that the proposed UPLC method can be used for the assessment of drug purity, stability, solubility and lipid-formulation release profile with no interference of excipients or related substances of active pharmaceutical ingredient

    Improvement of voltage stability and loadability of power system employing the placement of unified power flow controller using artificial neural network

    This paper proposes a voltage stability and loadability improvement model of power systems by incorporating the optimal placement of flexible alternating current transmission systems (FACTS) using an artificial neural network (ANN) called OPFANN. The key aspect of this model is to identify the weakest lines which having the most probability of voltage collapse utilized for placing FACTS devices. As installing a new power system network with rapidly increasing power demand cannot be possible, the operator usually operates the power system close to the stability limit. In this regard, continuous monitoring and improvement of system voltage stability and loadability of the existing system are vital issues for energy management systems nowadays. However, the proposed OPFANN introduces a more straightforward and faster scheme for voltage stability monitoring systems using ANN. Intelligent and reliable data samples have been designed to train the ANN based on two-line voltage stability indices (LVSI) techniques. Compared with other works, OPFANN effectively improves voltage stability and loadability at the load point by installing the unified power flow controller (UPFC) FACTS devices to the weakest lines. OPFANN can provide information on voltage collapse points using ANN and reduce the further computational cost of LVSI. Finally, OPFANN ensures faster and more secure operation of the power system

    Rota virus diarrhea in hospitalized children

    Objective: To determine the frequency and clinical features of Rota virus diarrhea in children presenting in a tertiary care hospital. Study Design: A cross-sectional, observational study. Place and Duration of Study: National Institute of Child Health, Karachi, from January to June 2007. Methodology: A total of three hundred children of either gender aged 1 month to 5 years, who presented with diarrhea of \u3c 7 days as a primary illness were enrolled. Children with bloody diarrhea or nosocomial gastroenteritis acquired duringhospitalization for other disease were not included. Detection of Rota virus in stool was done by enzyme linked immunoassay. Results: Out of 300 children, 188 (63%) tested positive and 112 (37%) tested negative for Rota virus. Positive Rota virus cases in 7 – 12 months of age was (n = 34, 18.08%). Overall, 151 (80.3%) children with Rota virus were less than 3 years old. 182 (60.7%) had fever, 118 (39.3%) had vomiting and 156 (82.9%) children had both fever and vomiting. Conclusion: This study shows that Rota virus is a common organism causing diarrhea in children less than 3 years of age. There is a need to incorporate Rota virus vaccine in the national EPI program to decrease the disease burden as highlighted in this study

    Pattern of relapse in paediatric acute lymphoblastic leukaemia in a tertiary care unit

    OBJECTIVE: To determine the frequency, site and time to relapse from diagnosis, and to see the relationship of relapse with important prognostic factors. METHODS: The prospective descriptive observational study was conducted at the National Institute of Child Health, Karachi, June 2005 to May 2007, and comprised newly-diagnosed cases of acute lymphoblastic leukaemia. Bone marrow aspiration was done on reappearance of blast cells in peripheral smear and cerebrospinal fluid. Detailed report was done each time when intra-thecal chemotherapy was given or there were signs and symptoms suggestive of central nervous system relapse. SPSS 12 was used for data analysis. RESULTS: Of the 60 patients enrolled, 4(6.6%) expired and 1(1.7%) was lost to follow-up. Of the 55(91.6%) who comprised the study sample, 35(58%) were males and 25(42%) females. Mean age of relapse was 6.8±3.27 years. Mean time to relapse from diagnosis was 1.3±0.54 years; 12(20%) patients suffered relapse, and of them 5(14%) were boys. Central nervous system relapse in 8(67%) patients was the most common site, with 3(25%) bone-marrow relapses. Out of 12 patient with relapses, 9(75%) had white blood cell count less than 50,000/cm. CONCLUSIONS: Relapse in acute lymphoblastic leukaemia was common, although treatment modalities are improving day by day

    Doubly Orthogonal Wavelet Packets for Multi-Users Indoor Visible Light Communication Systems

    Visible Light Communication (VLC) is a data communication technology that modulates the intensity of the light to transmit the information mostly by means of Light Emitting Diodes (LEDs). The data rate is mainly throttled by the limited bandwidth of the LEDs. To combat, Multi-carrier Code Division Multiple Access (MC-CDMA) is a favorable technique for achieving higher data rates along with reduced Inter-Symbol Interference (ISI) and easy access to multi-users at the cost of slightly reduced compromised spectral efficiency and Multiple Access Interference (MAI). In this article, a multi-user VLC system is designed using a Discrete Wavelet Transform (DWT) that eradicates the use of cyclic prefix due to the good orthogonality and time-frequency localization properties of wavelets. Moreover, the design also comprises suitable signature codes, which are generated by employing double orthogonality depending upon Walsh codes and Wavelet Packets. The proposed multi-user system is simulated in MATLAB software and its overall performance is assessed using line-of-sight (LoS) and non-line-of-sight (NLoS) configurations. Furthermore, two sub-optimum multi-users detection schemes such as zero forcing (ZF) and minimum-mean-square-error (MMSE) are also used at the receiver. The simulated results illustrate that the doubly orthogonal signature waveform-based DWT-MC-CDMA with MMSE detection scheme outperforms the Walsh code-based multi-user system

    Engagement with consumer smartwatches for tracking symptoms of individuals living with multiple long-term conditions (multimorbidity)::A longitudinal observational study

    INTRODUCTION: People living with multiple long-term conditions (multimorbidity) (MLTC-M) experience an accumulating combination of different symptoms. It has been suggested that these symptoms can be tracked longitudinally using consumer technology, such as smartphones and wearable devices. AIM: The aim of this study was to investigate longitudinal user engagement with a smartwatch application, collecting survey questions and active tasks over 90 days, in people living with MLTC-M. METHODS: ‘Watch Your Steps’ was a prospective observational study, administering multiple questions and active tasks over 90 days. Adults with more than one clinician-diagnosed long-term conditions were loaned Fossil® Sport smartwatches, pre-loaded with the study app. Around 20 questions were prompted per day. Daily completion rates were calculated to describe engagement patterns over time, and to explore how these varied by patient characteristics and question type. RESULTS: Fifty three people with MLTC-M took part in the study. Around half were male ( = 26; 49%) and the majority had a white ethnic background (n = 45; 85%). About a third of participants engaged with the smartwatch app nearly every day. The overall completion rate of symptom questions was 45% inter-quartile range (IQR 23–67%) across all study participants. Older patients and those with greater MLTC-M were more engaged, although engagement was not significantly different between genders. CONCLUSION: It was feasible for people living with MLTC-M to report multiple symptoms per day over 3 months. User engagement appeared as good as other mobile health studies that recruited people with single health conditions, despite the higher daily data entry burden

    The Soybean Genome Database (SoyGD): a browser for display of duplicated, polyploid, regions and sequence tagged sites on the integrated physical and genetic maps of Glycine max

    Genomes that have been highly conserved following increases in ploidy (by duplication or hybridization) like Glycine max (soybean) present challenges during genome analysis. At the Soybean Genome Database (SoyGD) genome browser has, since 2002, integrated and served the publicly available soybean physical map, bacterial artificial chromosome (BAC) fingerprint database and genetic map associated genomic data. The browser shows both build 3 and build 4 contiguous sets of clones (contigs) of the soybean physical map. Build 4 consisted of 2854 contigs that encompassed 1.05 Gb and 404 high-quality DNA markers that anchored 742 contigs. Many DNA markers anchored sets of 2–8 different contigs. Each contig in the set represented a homologous region of related sequences. GBrowse was adapted to show sets of homologous contigs at all potential anchor points, spread laterally and prevented from overlapping. About 8064 minimum tiling path (MTP2) clones provided 13 473 BAC end sequences (BES) to decorate the physical map. Analyses of BES placed 2111 gene models, 40 marker anchors and 1053 new microsatellite markers on the map. Estimated sequence tag probes from 201 low-copy gene families located 613 paralogs. The genome browser portal showed each data type as a separate track. Tetraploid, octoploid, diploid and homologous regions are shown clearly in relation to an integrated genetic and physical map
