    Effects of broadband noise on cortical evoked auditory responses at different loudness levels in young adults

    Young adults with no history of hearing concerns were tested to investigate their /da/-evoked cortical auditory evoked potentials (P1-N1-P2) recorded from 32 scalp electrodes in the presence and absence of noise at three different loudness levels (soft, comfortable, and loud), at a fixed signal-to-noise ratio (+3 dB). P1 peak latency significantly increased at soft and loud levels, and N1 and P2 latencies increased at all three levels in the presence of noise, compared with the quiet condition. P1 amplitude was significantly larger in quiet than in noise conditions at the loudest level. N1 amplitude was larger in quiet than in noise for the soft level only. P2 amplitude was reduced in the presence of noise to a similar degree at all loudness levels. The differential effects of noise on P1, N1, and P2 suggest differences in auditory processes underlying these peaks. The combination of level and signal-to-noise ratio should be considered when using cortical auditory evoked potentials as an electrophysiological indicator of degraded speech processing

    Evaluation of Marine Zooplankton Community Structure Through Environmental DNA Metabarcoding

    Zooplankton dominate the abundance and biomass of multicellular animals in pelagic marine environments; however, traditional methods to characterize zooplankton communities are invasive and laborious. This study compares zooplankton taxonomic composition revealed through metabarcoding of the cytochrome oxidase I (COI) and 18S rRNA genes to traditional morphological identification by microscopy. Triplicates of three different sample types were collected from three coral reef sites in the Florida Keys National Marine Sanctuary: (1) 1 L surface seawater samples prefiltered through 3 ÎŒm filters and subsequently collected on 0.22 ÎŒm filters for eDNA (PF‐eDNA); (2) 1 L surface seawater samples filtered on 0.22 ÎŒm pore‐size filters (environmental DNA; eDNA), and (3) zooplankton tissue samples from 64 ÎŒm, 200 ÎŒm, and 500 ÎŒm mesh size net tows. The zooplankton tissue samples were split, with half identified morphologically and tissue DNA (T‐DNA) extracted from the other half. The COI and 18S rRNA gene metabarcoding of PF‐eDNA, eDNA, and T‐DNA samples was performed using Illumina MiSeq. Of the families detected with COI and 18S rRNA gene metabarcoding, 40% and 32%, respectively, were also identified through morphological assessments. Significant differences in taxonomic composition were observed between PF‐DNA, eDNA, and T‐DNA with both genetic markers. PF‐eDNA resulted in detection of fewer taxa than the other two sample types; thus, prefiltering is not recommended. All dominant copepod taxa (\u3e 5% of total abundance) were detected with eDNA, T‐DNA, and morphological assessments, demonstrating that eDNA metabarcoding is a promising technique for future biodiversity assessments of pelagic zooplankton in marine systems

    A 31-residue peptide induces aggregation of tau's microtubule-binding region in cells

    The self-propagation of misfolded conformations of tau underlies neurodegenerative diseases, including Alzheimer's. There is considerable interest in discovering the minimal sequence and active conformational nucleus that defines this self-propagating event. The microtubule-binding region, spanning residues 244-372, reproduces much of the aggregation behaviour of tau in cells and animal models. Further dissection of the amyloid-forming region to a hexapeptide from the third microtubule-binding repeat resulted in a peptide that rapidly forms fibrils in vitro. We show that this peptide lacks the ability to seed aggregation of tau244-372 in cells. However, as the hexapeptide is gradually extended to 31 residues, the peptides aggregate more slowly and gain potent activity to induce aggregation of tau244-372 in cells. X-ray fibre diffraction, hydrogen-deuterium exchange and solid-state NMR studies map the beta-forming region to a 25-residue sequence. Thus, the nucleus for self-propagating aggregation of tau244-372 in cells is packaged in a remarkably small peptide

    Obesity, type 2 diabetes mellitus (DM) and metabolic syndrome (MS) are common in patients with heart failure (HF). Studies investigating the association between known biomarkers and adiposity in patient populations are limited. The aim of the present study was to investigate the association between C-reactive protein (CRP) and leptin with adiposity in a sub-group of overweight/obese patients with HF, DM and/or MS. A total of 36 patients (mean age, 56.72±9.78 years; ranging between 27 and 76 years of age; 80.6% male; 52.8% Caucasian) were enrolled and their height, weight, waist circumference and body composition (e.g. percentage body fat and lean mass), as well as the levels of CRP and leptin, were assessed.
