Investigating the DNA-Binding Site for VirB, a Key Transcriptional Regulator of Shigella Virulence Genes, Using an In Vivo Binding Tool
The transcriptional anti-silencing and DNA-binding protein, VirB, is essential for the virulence of Shigella species and, yet, sequences required for VirB-DNA binding are poorly understood. While a 7-8 bp VirB-binding site has been proposed, it was derived from studies at a single VirB-dependent promoter, icsB. Our previous in vivo studies at a different VirB-dependent promoter, icsP, found that the proposed VirB-binding site was insufficient for regulation. Instead, the required site was found to be organized as a near-perfect inverted repeat separated by a single nucleotide spacer. Thus, the proposed 7-8 bp VirB-binding site needed to be re-evaluated. Here, we engineer and validate a molecular tool to capture protein-DNA binding interactions in vivo. Our data show that a sequence organized as a near-perfect inverted repeat is required for VirB-DNA binding interactions in vivo at both the icsB and icsP promoters. Furthermore, the previously proposed VirB-binding site and multiple sites found as a result of its description (i.e., sites located at the virB, virF, spa15, and virA promoters) are not sufficient for VirB to bind in vivo using this tool. Determination of the electron velocity distribution from the soft and hard x-ray emission during lower-hybrid current drive on PLT
During lower-hybrid heating in low-density-tokamak discharges, a nonMaxwellian tail of high-energy electrons is formed. This tail carries the plasma current. Utilizing the fact that relativistic electrons emit bremsstrahlung predominantly in the forward direction, we investigate the shape of the electron distribution by measuring the dependence of the x-ray emission on the angle between the magnetic field and the line of sight. The experimental data indicate that the distribution function is predominantly peaked in the forward direction, although a small fraction of the electrons is in the backward cone. The energy dependence of the x-ray spectra is consistent with that of a velocity distribution which has a plateau extending out to several hundred kiloelectron volts. Radial profiles show that the hot electrons are located in the central plasma region and form a high-conductivity plasma with the current profile frozen in. The slope of the spectrum depends on the rf power and on the phasing of the waveguide grill, but not on the externally applied plasma voltage. Relaxation oscillations occur shortly after switching the rf off. They also appear during the rf for low rf power and at the high-density limit of the lower-hybrid current drive. The x-ray spectra confirm that parallel energy is transferred to perpendicular energy during the instability, suggesting an instability due to the anomalous Doppler effect
Nonlinear Radiation Pressure and Stochasticity in Ultraintense Laser Fields
The radiation force on a single electron in an ultraintense plane wave () is calculated and shown to be proportional to in the
high- limit for arbitrary waveform and polarization. The cyclotron motion of
an electron in a constant magnetic field and an ultraintense plane wave is
numerically found to be quasiperiodic even in the high- limit if the
magnetic field is not too strong, as suggested by previous analytical work. A
strong magnetic field causes highly chaotic electron motion and the boundary of
the highly chaotic region of parameter space is determined numerically.

Negative pressures in full-scale distribution system: field investigation, modelling, estimation of intrusion volumes and risk for public health
International audienceVarious investigations encompassing microbial characterization of external sources of contamination (soil and trenchwater surrounding water mains, flooded air-valve vaults), field pressure monitoring, and hydraulic and transient analyses were conducted in the same distribution system where two epidemiological studies showing an increase in gastrointestinal illness for people drinking tap water were conducted in the 1990's. Interesting results include the detection of microorganisms indicators of fecal contamination in all external sources investigated but at a higher frequency in the water from flooded air-valve vaults, and the recording of 18 negative pressure events in the distribution system during a 17-month monitoring period. Transient analysis of this large and complex distribution system was challenging and highlighted the need to consider field pressure data in the process
Marital Histories and Economic Well-Being
Compared to unmarried individuals married individuals report greater average wealth. A restricted focus on current marital status risks misrepresenting the effects of marriage on wealth, as an increasing proportion of older adults have been divorced and remarried, having lived through the dramatic upheavals in family structure from the 1960s through the 1980s. To shed light on the associations between a lifetime of marriage events and wealth near retirement, we used panel data from the Health and Retirement Study and developed categories of marital experiences that acknowledged current status, type, number and date of past marital disruptions and total duration of time spent married across the lifespan. We found that the route individuals took to get to their current marital status were important predictors of wealth levels near retirement and were different for males and females. Observable differences in lifetime earnings, mortality risk, risk aversion, other characteristics such as education and number of children, explained much of the wealth difference between married and remarried individuals however neither observable characteristics nor sources of other wealth from pensions and Social Security were enough to explain the large differences in wealth accumulation between single and married women and individuals experiencing more than one marital disruption. Given the higher divorce rate, prevalence of multiple divorces and earlier age of divorce of the Baby Boomer cohort compared to earlier cohorts, an understanding of how marriage disruptions over the lifecycle impact savings is increasingly important for understanding the economic security of retirees.Social Security Administrationhttp://deepblue.lib.umich.edu/bitstream/2027.42/61806/1/wp180.pd
A Framework for Incorporating Dyads in Models of HIV-Prevention
Although HIV is contracted by individuals, it is typically transmitted in dyads. Most efforts to promote safer sex practices, however, focus exclusively on individuals. The goal of this paper is to provide a theoretical framework that specifies how models of dyadic processes and relationships can inform models of HIV-prevention. At the center of the framework is the proposition that safer sex between two people requires a dyadic capacity for successful coordination. According to this framework, relational, individual, and structural variables that affect the enactment of safer sex do so through their direct and indirect effects on that dyadic capacity. This dyadic perspective does not require an ongoing relationship between two individuals; rather, it offers a way of distinguishing between dyads along a continuum from anonymous strangers (with minimal coordination of behavior) to long-term partners (with much greater coordination). Acknowledging the dyadic context of HIV-prevention offers new targets for interventions and suggests new approaches to tailoring interventions to specific populations
Universal diffusion near the golden chaos border
We study local diffusion rate in Chirikov standard map near the critical
golden curve. Numerical simulations confirm the predicted exponent
for the power law decay of as approaching the golden curve via principal
resonances with period (). The universal
self-similar structure of diffusion between principal resonances is
demonstrated and it is shown that resonances of other type play also an
