    PCR detection assay for sex determination in papaya using SCAR marker

    Papaya (Carica papaya L., 2n = 18), a polygamous angiosperm, is a major fruit crop in tropical and subtropical regions. It is trioecious with three sex forms: male, female, and hermaphrodite, where sex determination is controlled by the XY chromosome pair with two slightly different Y chromosomes i.e. Y for male and Yh for hermaphrodite. Sex type determination in papaya, which cannot be determined either by embryo shape or morphology at the juvenile developmental stage, is an essential pre-requisite for crop improvement processes as it helps in identification of fruitful plants. Hence, molecular profiling could be used as an alternative that provides a quick and reliable identifi cation of sex types in plantlets at initial stages only. In the present study we have validated the sex linked sequence characterized amplified region (SCAR) marker W11 using PCR detection assay among different cultivars of papaya i.e. dioecious with either female or male and gynodioecious with either female or hermaphrodites and also performed a double-blind test for validating the seedlings of 84 F1 plants, which resulted in their sex determination. The assay clearly gives 800 bp band in male plants in dioecious types and hermaphrodite in gynodioecious plants

    Not AvailableIntroduction-An attempt was made to analyze the morphological, biochemical and molecular diversity in karonda (Carissa carandas L.) populations maintained at Indian Institute of Horticultural Research in Bangalore, India, using a multivariate analysis approach. Material and methods-Morphological and biochemical traits were recorded and evaluated for variance and mean comparisons. Sequence Related Amplified Polymorphism (SRAP) molecular markers were used for generating information on genetic variation and relationships among the accessions. Mantel test was used for comparing the distance matrices derived from morphological, biochemical and molecular markers. Results and discussion-Significant variation was found in fruit related morphological characters and fruit quality traits. The combined analysis used in this study successfully categorized the different karonda accessions into various clusters based on genetic diversity and also established relatedness among them. A high correlation was found between Euclidean distance matrices of biochemical and morphological data. Conclusion-This work will be useful for the breeders working on this future crop for characterization, genotype identification and selection of parents.Not Availabl

    Development and evaluation of ready to serve (RTS) beverage from bael (Aegle marmelose Correa.)

    A research study was carried out to develop a RTS beverage by exploiting the nutritional and organoleptic properties of bael fruit pulp. Six treatment combinationsof bael RTS with 10, 15 and 20% of pulp concentration and 10 and 15°B of TSS were prepared based on the review of literature. The biochemical and organoleptic properties of the prepared RTS were evaluated during storage. The pH, ascorbic acid and antioxidant activity of the RTS decreased with the storage, while acidity and total sugars increased. Results of the sensory evaluation showed that there was a significant difference between treatments in terms of color, flavor, taste, body and overall acceptability. From the results of quality assessments, the formulated bael RTS beverage with 15% pulp and 15°B TSS was found to be superior and suitable for consumption up to 12 weeks without any significant changes in the quality characteristics

    Not AvailableThe purpose of this study was to investigate the effect of various DNA methylation modifying agents on genome wide global DNA methylation status and to develop papaya cv. Surya plants with useful and stable horticulturally important variations. DNA methylation-status modifying agents (Zebularine), 5-azacytidine, 5-aza deoxycytidine (DNA hypomethylating agents), 3’-azido-deoxythymidine (DNA hypermethylating agent) were used at two concentrations (0.3mM and 0.5mM) on papaya cv. Surya zygotic embryos for incubation periods ranging from 0-120 hrs. The technique of MS-RAPD-PCR which resolves global DNA methylation pattern was employed. A total of 87 primers were tested on the control samples, 55 primers showed PCR amplification, of which 11 showed polymorphic bands. Six primers showing clear and reproducible bands were selected further for analysing methylation-modifying-agent treated samples. This technique was found to be reliable and efficient for detecting changes in DNA methylation at both the scale and pattern in a genome wide standpoint.Not Availabl

    Not AvailableIn this paper we report the fruit characteristics and seed polyembryony in langsat, Lansium parasiticum (syn. Lansium domesticum Corr.). This fruit tree belongs to the family Meliaceae in order Sapindales and is considered to be native of western South East Asia and is common in both wild and cultivated forms throughout Malaysia and Philippines where the fruits are very popular and the tree is being utilized in reforestation efforts. It is also grown in southern Thailand and Vietnam and flourishes in the Nilgiris and other humid areas of South India. In the present investigation, morphological and biochemical parameters of the plants and fruits were investigated obtained from State Horticultural Farm, Buliar (latitude 11.34; longitude 76.79) in Tamil Nadu, at elevation of 360 m MSL and receiving average annual rainfall of 125.14 cm. The plantation was established in the year 1900 and consists of various tropical trees like mangosteen, langsat, arecanut, coffee, silveroak, pepper, cinnamon in tier system of planting.Not Availabl
