Paternal obesity induces metabolic and sperm disturbances in male offspring that are exacerbated by their exposure to an "obesogenic" diet

Obesity and related comorbidities are becoming increasingly prevalent globally. In mice preconception paternal exposure to a high fat diet (HFD) impairs the metabolic and reproductive health of male offspring, despite their control diet (CD) consumption. However, offspring share lifestyle, including diet, with parents. We assessed if male offspring from HFD fathers have a heightened susceptibility to HFD-induced metabolic and reproductive derangements. This 2 × 2 design saw founder males (F0) and their offspring (F1) fed either a HFD or a nutritionally matched CD. Regardless of paternal diet, HFD fed male offspring had greater total body weight and adiposity. Offspring sired by a HFD male and fed a HFD were the heaviest, had the greatest adiposity and had the greatest concentration of serum cholesterol, triglyceride, HDL, and NEFA compared with CD sired/fed littermates. A synergistic increase in serum insulin was unmasked by both father/son HFD consumption, concomitant with increased sera glucose. Either a paternal or offspring HFD was associated with similar reductions to offspring sperm motility. Whereas sperm ROS concentrations and sperm-oocyte binding saw detrimental effects of both F0 HFD and F1 HFD with an interaction evident between both, culminating in the most impaired sperm parameters in this group. This indicates that metabolic and fertility disturbances in male offspring sired by HFD fathers are exacerbated by a "second-hit" of exposure to the same obesogenic environment postnatally. If translatable to human health, this suggests that adverse reproductive and metabolic outcomes may be amplified across generations through a shared calorie dense diet, relevant to the current worldwide obesity epidemic.Tod Fullston, Nicole O. Paternal under-nutrition programs metabolic syndrome in offspring which can be reversed by antioxidant/vitamin food fortification in fathers

There is an ever increasing body of evidence that demonstrates that paternal over-nutrition prior to conception programs impaired metabolic health in offspring. Here we examined whether paternal under-nutrition can also program impaired health in offspring and if any detrimental health outcomes in offspring could be prevented by micronutrient supplementation (vitamins and antioxidants). We discovered that restricting the food intake of male rodents reduced their body weight, fertility, increased sperm oxidative DNA lesions and reduced global sperm methylation. Under-nourished males then sired offspring with reduced postnatal weight and growth but somewhat paradoxically increased adiposity and dyslipidaemia, despite being fed standard chow. Paternal vitamin/antioxidant food fortification during under-nutrition not only normalised founder oxidative sperm DNA lesions but also prevented early growth restriction, fat accumulation and dyslipidaemia in offspring. This demonstrates that paternal under-nutrition reduces postnatal growth but increases the risk of obesity and metabolic disease in the next generation and that micronutrient supplementation during this period of under-nutrition is capable of restoring offspring metabolic health

Rational design of Bi-doped rGO/Co3O4 nanohybrids for ethanol sensing

Gas sensors based on metal oxide semiconductors (MOSCs) and reduced graphene oxide (rGO) for sensing of organic volatile compounds often suffer from high operation temperature, low responses, poor selectivity, or narrow detection range. Herein, we design and fabricate Bi-doped rGO/Co3O4 (BGCO) nanohybrids with a flower morphology, which have been applied as a sensing layer for an ethanol sensor. This BGCO sensor exhibits a maximum p-type response of 178.1 towards 500 ppm ethanol at an optimum working temperature of 120 °C. The sensor’s detection range for the ethanol concentration is from 500 ppb to 500 ppm, and the sensor has an excellent selectivity to ethanol compared to other types of organic volatile gases and oxidizing gas such as NO2. The enhanced ethanol sensing mechanism is attributed to the increased conductivity of Bi doped rGO/Co3O4 material. Additionally, incorporation of Bi dopant can promote the redox reaction, and the rGO/Co3O4 act as the catalyst

Background: Current treatments for chronic hepatitis B are suboptimal. In the search for improved therapies, we compared the efficacy and safety of pegylated interferon alfa plus lamivudine, pegylated interferon alfa without lamivudine, and lamivudine alone for the treatment of hepatitis B e antigen (HBeAg)–positive chronic hepatitis B. Methods: A total of 814 patients with HBeAg-positive chronic hepatitis B received either peginterferon alfa-2a (180 µg once weekly) plus oral placebo, peginterferon alfa-2a plus lamivudine (100 mg daily), or lamivudine alone. The majority of patients in the study were Asian (87 percent). Most patients were infected with hepatitis B virus (HBV) genotype B or C. Patients were treated for 48 weeks and followed for an additional 24 weeks. Results: After 24 weeks of follow-up, significantly more patients who received peginterferon alfa-2a monotherapy or peginterferon alfa-2a plus lamivudine than those who received lamivudine monotherapy had HBeAg seroconversion (32 percent vs. 19 percent [

We report the measurement of the cross sections for e+e−e^+e^-→\rightarrow{nOCH} (nOCH stands for non-open charm hadrons) with improved precision at center-of-mass energies from 3.645 to 3.871 GeV. We observe for the first time a three-resonance structure in the energy-dependent lineshape of the cross sections, which are R(3760)\mathcal R(3760), R(3780)\mathcal R(3780) and R(3810)\mathcal R(3810) with significances of 9.4σ9.4\sigma, 15.7σ15.7\sigma, and 9.8σ9.8\sigma, respectively. The R(3810)\mathcal R(3810) is observed for the first time. We found two solutions in analysis of the cross sections. For solution I [solution II], we measure the mass, the total width and the product of electronic width and nOCH decay branching fraction to be (3805.8±1.1±2.7)(3805.8 \pm 1.1 \pm 2.7) [(3805.8±1.1±2.7)(3805.8 \pm 1.1 \pm 2.7)] MeV/c2c^2, (11.6±2.6±1.9)(11.6 \pm 2.6 \pm 1.9) [(11.5±2.5±1.8)(11.5 \pm 2.5 \pm 1.8)] MeV, and (10.8±3.2±2.3)(10.8\pm 3.2\pm 2.3) [(11.0±2.9±2.4)(11.0\pm 2.9\pm 2.4)] eV for the R(3810)\mathcal R(3810), respectively. In addition, we measure the branching fractions B(R(3760){\mathcal B}({\mathcal R}(3760)→\rightarrow{nOCH})=(24.5±13.4±27.4)%[(6.8±5.4±7.6)%])=(24.5 \pm 13.4 \pm 27.4)\% [(6.8 \pm 5.4 \pm 7.6)\%] for the first time, and B(R(3780){\mathcal B}(\mathcal R(3780)→\rightarrow{nOCH})=(11.6±5.8±7.8)%[(10.3±4.5±6.9)%])=(11.6 \pm 5.8 \pm 7.8)\% [(10.3 \pm 4.5 \pm 6.9)\%]. Moreover, we determine the open-charm (OC) branching fraction B(R{\mathcal B}({\mathcal R}(3760)→(3760)\rightarrow{OC})=(75.5±13.4±27.4)%[(93.2±5.4±7.6)%])=(75.5 \pm 13.4 \pm 27.4)\% [(93.2 \pm 5.4 \pm 7.6)\%], which supports the interpretation of R(3760)\mathcal R(3760) as an OC pair molecular state, but contained a simple four-quark state component. The first uncertainties are from fits to the cross sections, and the second are systematic
