Migraine and Cognitive Decline Among Women: Prospective Cohort Study
Objective: To evaluate the association between migraine and cognitive decline among women. Design Prospective cohort study. Setting Womenâs Health Study, United States. Participants 6349 women aged 65 or older enrolled in the Womenâs Health Study who provided information about migraine status at baseline and participated in cognitive testing during follow-up. Participants were classified into four groups: no history of migraine, migraine with aura, migraine without aura, and past history of migraine (reports of migraine history but no migraine in the year prior to baseline). Main outcome measures Cognitive testing was carried out at two year intervals up to three times using the telephone interview for cognitive status, immediate and delayed recall trials of the east Boston memory test, delayed recall trial of the telephone interview for cognitive status 10 word list, and a category fluency test. All tests were combined into a global cognitive score, and tests assessing verbal memory were combined to create a verbal memory score. Results: Of the 6349 women, 853 (13.4%) reported any migraine; of these, 195 (22.9%) reported migraine with aura, 248 (29.1%) migraine without aura, and 410 (48.1%) a past history of migraine. Compared with women with no history of migraine, those who experienced migraine with or without aura or had a past history of migraine did not have significantly different rates of cognitive decline in any of the cognitive scores: values for the rate of change of the global cognitive score between baseline and the last observation ranged from â0.01 (SE 0.04) for past history of migraine to 0.08 (SE 0.04) for migraine with aura when compared with women without any history of migraine. Women who experienced migraine were also not at increased risk of substantial cognitive decline (worst 10% of the distribution of decline). When compared with women without a history of migraine, the relative risks for the global score ranged from 0.77 (95% confidence interval 0.46 to 1.28) for women with migraine without aura to 1.17 (0.84 to 1.63) for women with a past history of migraine. Conclusion: In this prospective cohort of women, migraine status was not associated with faster rates of cognitive decline
One-pot Enzymatic Synthesis of Deoxy-thymidine-diphosphate (TDP)-2-deoxy-â-d-glucose Using Phosphomannomutase
Production of deoxy-thymidine-diphosphate (TDP)-sugars as substrates of glycosyltransferases, has been one of main hurdles for combinatorial antibiotic biosynthesis, which combines sugar moiety with aglycon of various antibiotics. Here, we report the one-pot enzymatic synthesis of TDP-2-deoxy-glucose employing high efficient TMP kinase (TMK; E.C. 2.7.2.12), acetate kinase (ACK; E.C. 2.7.1.21), and TDP-glucose synthase (TGS; E.C. 2.7.7.24) with phosphomannomutase (PMM; E.C. 5.4.2.8). In this study, replacing phosphoglucomutase (PGM; E.C. 5.4.2) by PMM from Escherichia coli gave four times higher specific activity on 2-deoxy-6-phosphate glucose, suggesting that the activity on 2-deoxy-glucose-6-phosphate was mainly affected by PMM activity, not PGM activity. Using an in vitro system starting from TMP and 2-deoxy-glucose-6-phosphate glucose, TDP-2-deoxy-glucose (63% yield) was successfully synthesized. Considering low productivity of NDP-sugars from cheap starting materials, this paper showed how production of NDP-sugars could be enhanced by controlling mutase activity
Consensus of Corporate E-Learning System Stakeholders Regarding the Satisfaction of End-Users
The purpose of this study is to call attention to the consensus of stakeholders of corporate e-Learning system regarding success. We identified the critical success factors (contents, technical features, management, and organizational support) as major components of corporate eLearning systems and questioned whether stakeholdersâ consensus on the importance of these components facilitates the implementation of these components to achieve good quality or well. We also questioned whether the influence of these components on user satisfaction could be moderated by contextual factors. Based on empirical testing of 18 eLearning user companies, we verified that the consensus of stakeholders regarding the importance of content, technological features, and organizational support has a positive influence on the perceived quality of these factors in their e-Learning systems, which in turn is positively related to user satisfaction. The learning subjects and learning style did significantly moderate the influences of these perceived qualities on user satisfaction
Real-time detection of an airborne microorganism using inertial impaction and mini-fluorescent microscopy
To achieve successful real-time detection of airborne pathogenic microorganisms, the problem must be considered in terms of their physical size and biological characteristics. We developed an airborne microorganism detection chip to realize the detection of microorganisms, ensuring compactness, sensitivity, cost-efficiency, and portability, using three key components: an inertial impaction system, a cartridge-type impaction plate, and a mini-fluorescent microscope. The inertial impaction system was used to separate microorganisms in terms of their aerodynamic particle size, and was fabricated with three impaction stages. Numerical analysis was performed to design the system; the calculated cutoff diameter at each impaction stage was 2.02 (first stage), 0.88 (second stage), and 0.54 ÎŒm (third stage). The measured cutoff diameters were 2.24, 0.91, and 0.49 ÎŒm, respectively. A cartridge-type impaction plate was used, composed of molded polydimethylsiloxane (PDMS) and an actual impaction region made of a SYBR green I dye-stained agar plate. A mini-fluorescent microscope was used to distinguish microbes from non-biological particles. Images of the microorganisms deposited at the impaction zone were obtained via mini-fluorescent microscopy, and fluorescent intensities of the images were calculated using in-house image-processing software. The results showed that the developed system successfully identified aerosolized biological particles from non-biological particles in real time
