    Designing a Tool to Assess Professional Competences: Theoretical Foundations and Potential Applications

    This conceptual paper outlines the descriptive theoretical foundations or kernel theories for designing an information and communication technology (ICT) tool to assess professional competences in the Austrian trade and craft sector. Upon completion, the ICT-tool serves as a boundary object in which applicants and assessors can interact. While this paper consists of a literature review and conceptual discussion, the overall project is methodologically placed within a multidisciplinary design-science paradigm. Design science scaffolds and structures the development of a theoretical model, the generation of assessment-items and the ICT-tool itself. This paper discusses the necessary descriptive knowledge or kernel theories on which the design of the ICT-tool rests. First, we describe the validation of prior learning - a process advocated by the European Union to make professional competences visible. Second, we describe the process how professional competences come about: through formal, non-formal and informal learning. Subsequently, we outline a knowledge-driven discourse on professional competences and discuss how different definitions of professional competence afford different approaches for its assessment. By presenting a use-case, we outline how the ICT-tool may guide applicants and assessors through this process

    In this article, we analyze different approaches to the term global city and reveal the process of formationof this phenomenon in social sciences. We consider the specifics of such concepts as a global city, aworld city, town information, a creative city. The global city is seen as a factor of influence on the socioeconomicprocesses of the modern world, which determines the impact depending not only on the size ofthe city. The modern urban development goal - to become visible on the world map of politics, economy,allowing the city to develop, involving active and creative actors in their own space. This article discussesthe methods and mechanisms that contribute to changes in the urban space by different examples. Theinstability, volatility of urban space, the dependence on a huge number of factors and side effects ofdecisions are the main problems of the modern management of urban space. For the world city, as asystem that defines and forms these processes, the author introduces the concept of a control point, whichis a landmark, setting the direction of development and changing the dynamics of socio-cultural space

    Influence of Specific Energy Inhomogeneity on the CO2 Splitting Performance in a High-Power Plasma Jet

    Plasma-based CO2 conversion is a promising pathway towards greenhouse gas recycling. In the corresponding research field, various types of plasma reactors are applied for carbon dioxide dissociation. So far, spatial inhomogeneities of the specific energy (SEI) distribution in plasma generators, e.g., induced by non-uniform heating or an inhomogeneous mass distribution, are not the focus of the investigations. In this work, the spatial inhomogeneity of mass-specific enthalpy in the plasma jet of the inductive plasma generator IPG4 at the Institute of Space Systems (IRS) is examined. For this, the mean mass-specific enthalpy as well as the radial distribution of the local enthalpy are measured using plasma probes. Moreover, the influence of the determined specific enthalpy inhomogeneity on the CO2 splitting performance is quantified. It is shown that an inhomogeneous radial distribution of the specific energy can significantly lower the carbon dioxide conversion, compared to a homogeneous case.     High-Throughput Molecular Cancer Cell Line Characterization Using Digital Multiplex Ligation-Dependent Probe Amplification for Improved Standardization of in Vitro Research.

    Tumor cell lines are widely used for cancer research, but challenges regarding quality control of cell line identity, cross contamination, and tumor somatic molecular stability remain, demanding novel approaches beyond conventional short tandem repeat profiling. A total of 21 commonly used multiple myeloma cell lines obtained from public repositories were analyzed by digital multiplex ligation-dependent probe amplification (digitalMLPA) to characterize germline single-nucleotide polymorphisms, insertions/deletions, and somatic copy number aberrations (CNAs). Using generated profiles and an in-house developed analytical pipeline, blinded experiments were performed to determine capability of digitalMLPA to predict cell line identity and potential spike-in DNA contamination in 41 anonymized cell line samples. The dominant cell line was correctly identified in all cases, and cross contamination was correctly detected in 33 of 37 samples with spike-in DNA; there were no false-positive predictions. The four samples in which spike in was not detected all carried low levels of contamination (1%), whereas levels of contamination ≥5% were correctly identified in all cases. Unsupervised clustering of CNA profiles identified shared commonalities that correlated with initiating Ig heavy locus translocation events. Longitudinal CNA assessment of nine cell lines revealed changes under standard culturing conditions not detected by insertion/deletion profiling alone. Results suggest that digitalMLPA can be utilized as a high-throughput tool for advanced quality assurance for in vitro cancer research
