Effects of orange juice formulation on prebiotic functionality using an in vitro colonic model sytem
A three-stage continuous fermentative colonic model system was used to monitor in vitro the effect of different orange juice formulations on prebiotic activity. Three different juices with and without Bimuno, a GOS mixture containing galactooligosaccharides (B-GOS) were assessed in terms of their ability to induce a bifidogenic microbiota. The recipe development was based on incorporating 2.75g B-GOS into a 250 ml serving of juice (65°Brix of concentrate juice). Alongside the production of B-GOS juice, a control juice - orange juice without any additional Bimuno and a positive control juice, containing all the components of Bimuno (glucose, galactose and lactose) in the same relative proportions with the exception of B-GOS were developed. Ion Exchange Chromotography analysis was used to test the maintenance of bimuno components after the production process. Data showed that sterilisation had no significant effect on concentration of B-GOS and simple sugars. The three juice formulations were digested under conditions resembling the gastric and small intestinal environments. Main bacterial groups of the faecal microbiota were evaluated throughout the colonic model study using 16S rRNA-based fluorescence in situ hybridization (FISH). Potential effects of supplementation of the juices on microbial metabolism were studied measuring short chain fatty acids (SCFAs) using gas chromatography. Furthermore, B-GOS juices showed positive modulations of the microbiota composition and metabolic activity. In particular, numbers of faecal bifidobacteria and lactobacilli were significantly higher when B-GOS juice was fermented compared to controls. Furthermore, fermentation of B-GOS juice resulted in an increase in Roseburia subcluster and concomitantly increased butyrate production, which is of potential benefit to the host. In conclusion, this study has shown B-GOS within orange juice can have a beneficial effect on the fecal microbiota

Effect of osmotic dehydration in fructose, sucrose and fructooligosaccharide solutions on the content of saccharides in plums and apples and their energy value
Plums (Najbolia and Stanley) and apples (Idared) were subjected to osmotic dehydration in 50% solutions of fructose, sucrose and fructooligosaccharides (FOS) at 22, 40 and 60 °C for 24 hours. The content of fructooligosaccharides, sucrose and monosaccharides in dried material was determined. Plums osmosed in fructose contained from 22.3% w/w to 29.6%w/w of this saccharide depending on the process temperature. The content of sucrose in plums and apples varied from 18.6% w/w to 30.4% w/w after using sucrose as osmotic agent. Material processed at 40 °C was characterised by the highest content of FOS: 22.624.7% w/w in plums (nystose as osmotic agent) and 13.7% w/w in apples (FOS preparation as osmotic agent). The partial replacement of sucrose and monosaccharides by fructooligosaccharides reduced the energy value of carbohydrates in dried material by 1237% depending on the process conditions.
Production of fructooligosaccharides [FOS] concentrations with various contents of oligomers using enzymatic bioconversion of saccharose
Opisano warunki prowadzenia biokonwersji sacharozy z użyciem komercyjnych preparatów enzymatycznych Fructozyme i Novoferm, zawierających fruktozylotransferazę (FT) do wytwarzania trzech typów syropów FOS i dwóch proszkowych preparatów o zawartości fruktanów odpowiednio 50-60% s.s. i ≥ 90% s.s. Wykazano, że przy aktywności FT 1-2,5 U/g sacharozy proces transglikozylacji reszty fruktozy przebiega w trzech fazach. W pierwszej blisko 80% utworzonych oligomerów stanowi kestoza, w drugiej kestoza ulega transglikozylacji z utworzeniem ponad 50% udziału nystozy w sumie FOS, a w trzeciej ustala się równowagę pomiędzy kestozą, nystozą i fruktozylonystozą wproporcji 2 : 4 : 3. Stwierdzono, że roztwory bogate w nystozę, po usunięciu glukozy na drodze enzymatycznego utleniania mogą być wysuszone metodą rozpyłową w warunkach otrzymywania mleka w proszku. Wykazano, że z syropów nystozowych o zawartości s.s. powyżej 72% (m/m) nystoza krystalizuje z niewielką domieszką kestozy w postaci niehigroskopijnej, drobnokrystalicznej substancji.In the paper, there are described conditions for bioconversion of saccharose into FOS preparations by commercial enzyme preparations (Fructozyme or Novoferm containing fructosyltransferase). The main objective of this bioconversion was to manufacture 3 FOS syrups and 2 powders containing 50%-60%, and over 90% of fructans respectively. It was proved that the transglycosylation of the fructose moiety was a three-phase process when the enzyme was applied in doses of 1.0-2.5 U/g saccharose. During the first phase, kestose is a major fructan formed (nearly 80% of the synthesized FOS). During the second stage, kestose is transformed into nystose (over 50% of the synthesized FOS). Additionally, during the third stage, a balance state among kestose, nystose and F-nystose (their proportion rates are 2:4:3) is settled. It was proved that after the glucose has been removed by an enzymatic oxidation, the nystose-rich solutions of FOS can be dried off using a spray-drying method; the dry-off process shall occur under the conditions similar to the production of powder milk. Furthermore, it was evidenced that from among all the nystose syrups containing over 72% (w/w) of dry matter, the nystose crystallizes with a little amount of kestose applied in the form of a non-hygroscopic fine-crystal substance
Effect of osmotic dehydration in the carbohydrate mixtures containing fructooligosaccharides on the oligosaccharide content of apples
Apples (Idared) were subjected to osmotic dehydration in: a) 50% solutions of fructooligosaccharides (FOS) at 40–70 °C (apple/solution 1/2), b) 50% solution of FOS at 40 °C using different amount of solution (apple-solution ratio from 1/2 to 1/5), c) 50–65% solution of FOS at 40 °C (apple/solution 1/4). The content of fructooligosaccharides in dried material was determined. An increase in temperature and amount of the hypertonic solution intensified the migration of fructooligosaccharides to the fruit tissue. There was no direct relationship between the concentration of the hypertonic solution and the FOS content of dehydrated apples. the contents of fructooligosaccharides and dry substance in fruits dehydrated under different conditions were interlinked. Dehydrated apples of a content of fructooligosaccharides of 7–9% w/w were obtained when the content of dry substance was approximately 30%
