Extraction of pure components from overlapped signals in gas chromatography-mass spectrometry (GC-MS)
Gas chromatography-mass spectrometry (GC-MS) is a widely used analytical technique for the identification and quantification of trace chemicals in complex mixtures. When complex samples are analyzed by GC-MS it is common to observe co-elution of two or more components, resulting in an overlap of signal peaks observed in the total ion chromatogram. In such situations manual signal analysis is often the most reliable means for the extraction of pure component signals; however, a systematic manual analysis over a number of samples is both tedious and prone to error. In the past 30 years a number of computational approaches were proposed to assist in the process of the extraction of pure signals from co-eluting GC-MS components. This includes empirical methods, comparison with library spectra, eigenvalue analysis, regression and others. However, to date no approach has been recognized as best, nor accepted as standard. This situation hampers general GC-MS capabilities, and in particular has implications for the development of robust, high-throughput GC-MS analytical protocols required in metabolic profiling and biomarker discovery. Here we first discuss the nature of GC-MS data, and then review some of the approaches proposed for the extraction of pure signals from co-eluting components. We summarize and classify different approaches to this problem, and examine why so many approaches proposed in the past have failed to live up to their full promise. Finally, we give some thoughts on the future developments in this field, and suggest that the progress in general computing capabilities attained in the past two decades has opened new horizons for tackling this important problem
What Do Computer Scientists Tweet? Analyzing the Link-Sharing Practice on Twitter
Twitter communication has permeated every sphere of society. To highlight and share small pieces of information with possibly vast audiences or small circles of the interested has some value in almost any aspect of social life. But what is the value exactly for a scientific field? We perform a comprehensive study of computer scientists using Twitter and their tweeting behavior concerning the sharing of web links. Discerning the domains, hosts and individual web pages being tweeted and the differences between computer scientists and a Twitter sample enables us to look in depth at the Twitter-based information sharing practices of a scientific community. Additionally, we aim at providing a deeper understanding of the role and impact of altmetrics in computer science and give a glance at the publications mentioned on Twitter that are most relevant for the computer science community. Our results show a link sharing culture that concentrates more heavily on public and professional quality information than the Twitter sample does. The results also show a broad variety in linked sources and especially in linked publications with some publications clearly related to community-specific interests of computer scientists, while others with a strong relation to attention mechanisms in social media. This refers to the observation that Twitter is a hybrid form of social media between an information service and a social network service. Overall the computer scientists’ style of usage seems to be more on the information-oriented side and to some degree also on professional usage. Therefore, altmetrics are of considerable use in analyzing computer science
Blocking TLR2 Activity Attenuates Pulmonary Metastases of Tumor
Background: Metastasis is the most pivotal cause of mortality in cancer patients. Immune tolerance plays a crucial role in tumor progression and metastasis. Methods and Findings: In this study, we investigated the potential roles and mechanisms of TLR2 signaling on tumor metastasis in a mouse model of intravenously injected B16 melanoma cells. Multiple subtypes of TLRs were expressed on B16 cells and several human cancer cell lines; TLR2 mediated the invasive activity of these cells. High metastatic B16 cells released more heat shock protein 60 than poor metastatic B16-F1 cells. Importantly, heat shock protein 60 released by tumor cells caused a persistent activation of TLR2 and was critical in the constitutive activation of transcription factor Stat3, leading to the release of immunosuppressive cytokines and chemokines. Moreover, targeting TLR2 markedly reduced pulmonary metastases and increased the survival of B16-bearing mice by reversing B16 cells induced immunosuppressive microenvironment and restoring tumor-killing cells such as CD8 + T cells and M1 macrophages. Combining an anti-TLR2 antibody and a cytotoxic agent, gemcitabine, provided a further improvement in the survival of tumor-bearing mice. Conclusions and Significance: Our results demonstrate that TLR2 is an attractive target against metastasis and that targeting immunosuppressive microenvironment using anti-TLR2 antibody is a novel therapeutic strategy for combating

Salah satufilm Perancis yang dianggap menginspirasi adalah film berjudul Le grand voyageyang ditulis dan disutradarai oleh Ismaël Ferroukhi. Film ini bercerita tentang perjalanan seorang muslim keturunan Maroko dan anaknya yang bernama Reda.Mereka menempuh jarak ribuan mil dari Perancis menuju ke kota Makah untukmelaksanakan haji hanya dengan mengendarai sebuah mobil tua. Penelitian inibertujuan untuk mengetahui pesan moral islami apa saja yang terkandung dalamfilm Le grand voyage dan bagaimana pesan tersebut dimunculkan dalam film.Penelitian ini menggunakan teori Struktural untuk menjawab rumusan masalah.Penelitian ini merupakan penelitian kualitatif dengan menggunakan teknik studipustaka serta dokumentasi sebagai metode pengumpulan data, dan teknikdeskriptif dalam proses analisis data.Berdasarkan hasil penelitian ini, terdapat 13 pesan moral islami yang terkandungdalam film Le grand voyage. Semua pesan moral tersebut mengacu pada sebuahproses perbaikan moral dan spiritual antara manusia dengan manusia dan alam,serta antara manusia dengan TuhanPenulis menyarankan pada penelitian selanjutnya untuk meneliti film Le grandvoyage menggunakan pendekatan sosiologi sastra yang nantinya dapat mengupashal apa saja yang melatarbelakangi pembuatan film ini dan tujuan sebenarnyayang ingin dicapai oleh pembuat film Le grand voyage
Social machines:A philosophical engineering
In Weaving the Web (2000), Berners-Lee defines Social Machines as biotechnologically hybrid Web-processes on the basis of which, “high-level activities, which have occurred just within one human’s brain, will occur among even larger more interconnected groups of people acting as if the shared a larger intuitive brain” (201–202). The analysis and design of Social Machines has already started attracting considerable attention both within the industry and academia. Web science, however, is still missing a clear definition of what a Social Machine is, which has in turn resulted in several calls for a “philosophical engineering” (Halpin 2013; Hendler & Berners-Lee 2010); Halpin et al. 2010). This paper is a first attempt to respond to this call, by combining contemporary philosophy of mind and cognitive science with epistemology. The idea of philosophical engineering implies that a sufficiently good conception of Social Machines should be of both theoretical and practical advantage. To demonstrate how the present approach can satisfy both objectives it will be used in order to address one of Wikipedia’s (the most famous Social Machine to date) most worrying concerns—i.e., the current and ongoing decline in the number of its active contributors (Halfacker et al. 2012)
A Crisis of Identity? Contradictions and New Opportunities
Drawing on the previous chapters, this chapter explores four tensions that characterise MOOCs. Although MOOCs are seen as an attempt to democratise education, they often privilege the elite, rather than acting as an equaliser. MOOCS are also considered a way to radically open access to education, yet they tend to offer education to people who are already able to learn rather than providing opportunities for everyone. While MOOCs are positioned as a disrupting force, often they replicate the customs and values associated with formal education, rather than unsettling educational norms.MOOCs are conceived as social networks that allow learners to learn through dialogue with others, yet many learners have limited interactions with others. Even when learners have the ability to learn autonomously, they often are expected to conform to course rules, rather than deciding their own learning strategies. These problems may be accentuated whereMOOCs are viewed as a set of products (content and credentials) on sale to student consumers, rather than as a transformational educational experience for learners. The view of MOOCs as a product for the consumer learner may overly simplify the complex, transformational processes that underscore learning. Particularly where underlying automated systems try to improve progression by quantifying learners’ behaviours and ‘correcting’ these to fit an ‘ideal’ learner profile or where algorithms and metrics are based on convectional education, rather than on future-facing forms of learning. This chapter examines these problems with MOOCs, offering promising future directions
Martigny Bourg au XIXe siècle: inventaire des bâtiments et typologie architecturale
The asynchronous relationship between millennial-scale temperature changes over Greenland and Antarctica during the last glacial period has led to the notion of a bipolar seesaw which acts to redistribute heat depending on the state of meridional overturning circulation within the Atlantic Ocean. Here we present new records from the South Atlantic that show rapid changes during the last deglaciation that were instantaneous (within dating uncertainty) and of opposite sign to those observed in the North Atlantic. Our results demonstrate a direct link between the abrupt changes associated with variations in the Atlantic meridional overturning circulation and the more gradual adjustments characteristic of the Southern Ocean. These results emphasize the importance of the Southern Ocean for the development and transmission of millennial-scale climate variability and highlight its role in deglacial climate change and the associated rise in atmospheric carbon dioxide
