    The neurobiological mechanism underlying the negative motivational component of withdrawal from acute opiate dependence is far from understood. Our objectives were to determine whether the glutamatergic system is involved in the motivational component of morphine withdrawal in acutely dependent rats and such an involvement is associated with dopaminergic neurotransmission. We examined the effects of various kinds of glutamate receptor antagonists on conditioned place aversion (CPA) induced by naloxone-precipitated withdrawal from a single morphine exposure 24 h before. Furthermore, the influence of pretreatment with the dopamine receptor antagonist haloperidol on those effects of glutamate receptor antagonists was also investigated. CPA was attenuated in a dose-dependent manner by all glutamate receptor antagonists examined including the NMDA receptor antagonists (+)-5-methyl-10,11-dihydro-5H-dibenzo[a,d]cyclo-hepten-5,10-imine maleate (MK-801) and phencyclidine hydrochloride (PCP), AMPA receptor antagonist 1-(4-aminophenyl)4-methyl-7,8-methylenedioxy-5H-2,3-benzodiazepine hydrochloride (GYKI 52466), and metabotropic receptor antagonists (+/-)-2-amino-3-phosphonopropionic acid (AP-3) and (+/-)-alpha-methyl-4-carboxyphenylglycine (MCPG). The effects of MK-801, GYKI 52466 and MCPG were blocked by haloperidol.These results suggest that the glutamatergic system involving multiple classes of receptors plays a role in the motivational component of withdrawal from acute morphine dependence, and the function of the glutamatergic system would be closely associated with dopaminergic neurotransmission

    Enhancing the Superconducting Transition Temperature due to Strong-Coupling Effect under Antiferromagnetic Spin Fluctuations in CeRh1-xIrxIn5 : 115In-NQR Study

    We report on systematic evolutions of antiferromagnetic (AFM) spin fluctuations and unconventional superconductivity (SC) in heavy-fermion (HF) compounds CeRh1x_{1-x}Irx_{x}In5_5 via 115^{115}In nuclear-quadrupole-resonance (NQR) experiment. The measurements of nuclear spin-lattice relaxation rate 1/T11/T_1 have revealed the marked development of AFM spin fluctuations as a consequence of approaching an AFM ordered state with increasing Rh content. Concomitantly the superconducting transition temperature TcT_{\rm c} and the energy gap Δ0\Delta_0 increase drastically from Tc=0.4T_{\rm c} = 0.4 K and 2Δ0/kBTc=52\Delta_0/k_{\rm B}T_{\rm c} = 5 in CeIrIn5_5 up to Tc=1.2T_{\rm c} = 1.2 K and 2Δ0/kBTc=8.32\Delta_0/k_{\rm B}T_{\rm c} = 8.3 in CeRh0.3_{0.3}Ir0.7_{0.7}In5_5, respectively. The present work suggests that the AFM spin fluctuations in close proximity to the AFM quantum critical point are indeed responsible for the onset of strong-coupling unconventional SC with the line node in the gap function in HF compounds.Comment: 4pages,5figures,to appear in Phys.     Motivation incorporates several psychological aspects that produce reward-related and learning behaviors. Although reward-related behavior is reported to be mediated by the dopaminergic reward pathway, the involvement of dopaminergic systems in motivated behavior has not been fully clarified. Several experimental methodologies for motivational behavior have been reported, but pharmacological characteristics seem to vary among these methodologies. In this review, we attempt to summarize three main concepts:(1) the relationship of dopamine neuron physiology with motivated behavior, (2) the pharmacological characteristics of the runway intracranial self-stimulation model, and (3) the behavioral distinction of disparate motivated behaviors

    気管支喘息や慢性閉塞性肺疾患の治療において、適切に吸入薬を使用することが重要である。本症例は60歳代女性で、労作時呼吸困難を訴え入院となった。点滴による治療、内服薬服用と同時に吸入薬の使用開始となったが、呼吸機能検査値が低値であり、製薬企業供与の吸入チェック器具により吸入できていないことが判明した。そこで、吸入薬を変更し吸入できていることを確認した。吸気流量が十分得られない場合は積極的にチェック器具により確認を行い、アドヒアランスの改善に努めることが重要である。In the treatment of bronchial asthma and chronic obstructive pulmonary disease, it is important to use inhalation therapy adequately. A 60’s woman was admitted to our hospital due to exertional dyspnea. She was diagnosed as having exacerbation of asthma. Although she was treated with intravenous, oral, and inhalation administration simultaneously after admissi on, her symptom did not improve enough, and her pulmonary function was still low. Application of the inhalation check device supplied by the medicine manufacture company grant demonstrated that she failed to inhale effectively . Therefore the inhaled glucocorticosteroid was changed, and it was confirmed that she was able to inhale successfully. Her successful inhalation resulted in the improvement of her respiratory symptom and pulmonary function. It is suggested that the manipulation is very important factor on inhalation therapy, and it is necessary to strive to improve the adherence for the treatment of bronchial asthma and chronic obstructive pulmonary disease

    Development of an appropriate simple suspension method for valganciclovir medication

    Background Valganciclovir (VGC) is essential for preventing cytomegalovirus infections after transplants in adult and pediatric patients. In pediatric patients, VGC tablets have to be pulverized so that they can be delivered via nasogastric tubes. The “simple suspension method” is usually used to suspend tablets in hot water in Japan. However, the optimal suspension conditions and metering methods for preparing VGC suspensions using the simple suspension method are unclear. The purpose of this study was to clarify these issues. Methods VGC tablets were suspended in water (initial water temperature: 25 °C or 55 °C) using the simple suspension method. The residual rate of VGC after it had been suspended in hot water was determined using HPLC. In addition, the suspended solution was passed through 6, 8, and 12 Fr. gavage tubes. The VGC concentrations of suspensions produced using different preparation methods were also determined using HPLC. Results Cracking the surfaces of VGC tablets and suspending them in water at an initial temperature of 55 °C was effective at dissolving the tablets. The VGC concentration of the suspension remained stable for at least 80 min. Furthermore, the VGC concentration remained stable for 48 h during cold dark storage. Cracking the surfaces of VGC tablets could be a more effective metering method than preparing powder from VGC tablets. In addition, little VGC remained in 6, 8, or 12 Fr. gavage tubes after VGC solution was passed through them. Conclusion The amount of VGC should be measured carefully when preparing VGC solutions using the simple suspension method

    Comparative genomic analysis of 1047 completely sequenced cDNAs from an Arabidopsis-related model halophyte, Thellungiella halophila

    <p>Abstract</p> <p>Background</p> <p><it>Thellungiella halophila </it>(also known as <it>T. salsuginea</it>) is a model halophyte with a small size, short life cycle, and small genome. <it>Thellungiella </it>genes exhibit a high degree of sequence identity with Arabidopsis genes (90% at the cDNA level). We previously generated a full-length enriched cDNA library of <it>T. halophila </it>from various tissues and from whole plants treated with salinity, chilling, freezing stress, or ABA. We determined the DNA sequences of 20 000 cDNAs at both the 5'- and 3' ends, and identified 9569 distinct genes.</p> <p>Results</p> <p>Here, we completely sequenced 1047 <it>Thellungiella </it>full-length cDNAs representing abiotic-stress-related genes, transcription factor genes, and protein phosphatase 2C genes. The predicted coding sequences, 5'-UTRs, and 3'-UTRs were compared with those of orthologous genes from Arabidopsis for length, sequence similarity, and structure. The 5'-UTR sequences of <it>Thellungiella </it>and Arabidopsis orthologs shared a significant level of similarity, although the motifs were rearranged. While examining the stress-related <it>Thellungiella </it>coding sequences, we found a short splicing variant of <it>T. halophila </it><it>salt overly sensitive 1 </it>(<it>ThSOS1</it>), designated <it>ThSOS1S</it>. ThSOS1S contains the transmembrane domain of ThSOS1 but lacks the C-terminal hydrophilic region. The expression level of <it>ThSOS1S </it>under normal growth conditions was higher than that of <it>ThSOS1</it>. We also compared the expression levels of Na<sup>+</sup>-transport-system genes between <it>Thellungiella </it>and Arabidopsis by using full-length cDNAs from each species as probes. Several genes that play essential roles in Na<sup>+ </sup>excretion, compartmentation, and diffusion (<it>SOS1</it>, <it>SOS2</it>, <it>NHX1</it>, and <it>HKT1</it>) were expressed at higher levels in <it>Thellungiella </it>than in Arabidopsis.</p> <p>Conclusions</p> <p>The full-length cDNA sequences obtained in this study will be essential for the ongoing annotation of the <it>Thellungiella </it>genome, especially for further improvement of gene prediction. Moreover, they will enable us to find splicing variants such as <it>ThSOS1S </it>(AB562331).</p
