    Biogeochemistry and geomicrobiology of cold-water coral carbonate mounds - lessons learned from IODP Expedition 307

    Large mound structures associated with cold-water coral ecosystems commonly occur on the slopes of continental margins, for instance, west of Ireland in the Porcupine Seabight, the Gulf of Cadiz or the Straits of Florida. In the Porcupine Seabight over 1500 mounds of up to 5 km in diameter and 250 m height lie at water depths of 600 to 900 m. The cold-water coral reef ecosystems associated with these structures are considered to be “hotspots” of organic carbon mineralization and microbial systems. To establish a depositional and biogeochemical/diagenetic model for cold-water carbonate mounds, Challenger Mound and adjacent continental slope sites were drilled in May 2005 during IODP Expedition 307. One major objective was to test whether deep sub-surface hydrocarbon flow and enhanced microbial activity within the mound structure was important in producing and stabilizing these sedimentary structures.Drilling results showed that the Challenger mound succession (IODP Site U1317) is 130 to 150 meters thick, and mainly consists of floatstone and rudstone facies formed of fine sediments and cold-water branching corals. Pronounced recurring cycles on the scales of several meters are recognized in carbonate content (up to 70% carbonate) and color reflectance, and are probably associated with Pleistocene glacial-interglacial cycles. A role for methane seepage and subsequent anaerobic oxidation was discounted both as a hard-round substrate for mound initiation and as a principal source of carbonate within the mound succession. A broad sulfate-methane transition (approximately 50 m thick) within the Miocene sediments suggested that the zone of anaerobic oxidation of methane principally occurs below the moundbase. In the mound sediments, interstitial water profiles of sulfate, alkalinity, Mg, and Sr suggested a tight coupling between carbonate diagenesis and low rates of microbial sulfate reduction. Overall organic carbon mineralization within cold-water coral mound appeared to be dominated by low rates of iron- and sulfate-reduction that occur in discrete layers within the mound. This was consistent with distributions of total cell-counts, acetate turnover (Webster et al. 2009) and hydrogenase activity (Soffiento et al. 2009). However, biomarker lipid distributions suggested that the Miocene sediments underlying the mound, into which sulfate is diffusing, as well as the sediments from the non-cold water coral reference site (U1318) contain higher abundances of living microbes. The results obtained from Expedition 307 are consistent with a picture emerging from other biogeochemical studies of cold-water coral mound and reef sites. Unless impacted by some external forcing (e.g. fluid flow or erosion event), the microbial activity in the underlying cold-water coral mound sediments is largely decoupled from the highly diverse, active surface ecosystem

    Low-cost UAV monitoring: insights into seasonal volumetric changes of an oyster reef in the German Wadden Sea

    This study aims to quantify the dimensions of an oyster reef over two years via low-cost unoccupied aerial vehicle (UAV) monitoring and to examine the seasonal volumetric changes. No current study investigated via UAV monitoring the seasonal changes of the reef-building Pacific oyster (Magallana gigas) in the German Wadden Sea, considering the uncertainty of measurements and processing. Previous studies have concentrated on classifying and mapping smaller oyster reefs using terrestrial laser scanning (TLS) or hyperspectral remote sensing data recorded by UAVs or satellites. This study employed a consumer-grade UAV with a low spectral resolution to semi-annually record the reef dimensions for generating digital elevation models (DEM) and orthomosaics via structure from motion (SfM), enabling identifying oysters. The machine learning algorithm Random Forest (RF) proved to be an accurate classifier to identify oysters in low-spectral UAV data. Based on the classified data, the reef was spatially analysed, and digital elevation models of difference (DoDs) were used to estimate the volumetric changes. The introduction of propagation errors supported determining the uncertainty of the vertical and volumetric changes with a confidence level of 68% and 95%, highlighting the significant change detection. The results indicate a volume increase of 22 m³ and a loss of 2 m³ in the study period, considering a confidence level of 95%. In particular, the reef lost an area between September 2020 and March 2021, when the reef was exposed to air for more than ten hours. The reef top elevation increased from -15.5 ± 3.6 cm NHN in March 2020 to -14.8 ± 3.9 cm NHN in March 2022, but the study could not determine a consistent annual growth rate. As long as the environmental and hydrodynamic conditions are given, the reef is expected to continue growing on higher elevations of tidal flats, only limited by air exposure. The growth rates suggest a further reef expansion, resulting in an increased roughness surface area that contributes to flow damping and altering sedimentation processes. Further studies are proposed to investigate the volumetric changes and limiting stressors, providing robust evidence regarding the influence of air exposure on reef loss

M. gigas, as an ecosystem engineer, builds reef structures that are characterized by highly complex and variable surfaces consisting of densely packed, sharp-edged individuals connected with cement-like bonds. To investigate the interactions between reef structure, shape and formation and wave as well as tidal currents, an understanding of the surface roughness is essential. This work reports on observations of oyster reefs for which seven new structural classes (Central Reef, Transitional Zone, Cluster I, Cluster II, Patch I, Patch II, and Garland) are proposed. For each class, high resolution Digital Elevation Models (DEMs) have been elaborated based on Structure-from-Motion (SfM) photogrammetry and analyzed using spatial statistics. By determining probability density functions (PDFs), vertical porosity distributions, abundances, orientations and second-order structure functions (SSFs), topographical parameters that influence the hydraulic bed roughness have been determined. The results suggest, that by applying the structural classification and their distinct topographical roughness parameters, the oyster reef surfaces can be described appropriately accounting for their complexity. The roughness accounts to a total roughness height kt = 103 ± 15 mm and root-mean-square roughness height krms = 23 ± 5 mm. These values were found similar across all structural classes, yet the shape of the PDFs reveal differences. With decreasing abundance, the distributions become more positively skewed and are characterized by more extreme outliers. This is reflected in the higher statistical moments, as the skewness ranges between Sk = 0.4–2.1 and the kurtosis between Ku = 2.2–11.5. The analysis of the orientations and the SSFs confirms anisotropic behavior across all structural classes. Further, the SSFs reveal the oyster shells as significant roughness elements with exception of Cluster I and II, where the clusters are identified as significant roughness elements. The provided set of topographical roughness parameters enhances the knowledge of oyster reef surfaces and gives insights into the interactions between biogenic structure and surrounding hydrodynamics. The new intra-reef classification allows for more accurate determination of the overall roughness as well as the population dynamics of the habitat forming oyster. Combined with hydraulic measurements, the results can be used to estimate the hydraulic bed roughness induced by the oyster reef surfaces.     Packed Red Blood Cell Transfusion Associates with Acute Kidney Injury After Transcatheter Aortic Valve Replacement

    Background: Acute kidney injury after cardiac surgery significantly associates with morbidity and mortality. Despite not requiring cardiopulmonary bypass, transcatheter aortic valve replacement patients have an incidence of post-procedural acute kidney injury similar to patients who undergo open surgical aortic valve replacement. Packed red blood cell transfusion has been associated with morbidity and mortality after cardiac surgery. We hypothesized that packed red blood cell transfusion independently associates with acute kidney injury after transcatheter aortic valve replacement, after accounting for other risk factors. Methods: This is a single-center retrospective cohort study of 116 patients undergoing transcatheter aortic valve replacement. Post-transcatheter aortic valve replacement acute kidney injury was defined by Kidney Disease: Improving Global Outcomes serum creatinine-based criteria. Univariate comparisons between patients with and without post-transcatheter aortic valve replacement acute kidney injury were made for clinical characteristics. Multivariable logistic regression was used to assess independent association of packed red blood cell transfusion with post-transcatheter aortic valve replacement acute kidney injury (adjusting for pre-procedural renal function and other important clinical parameters). Results: Acute kidney injury occurred in 20 (17.2%) subjects. Total number of packed red blood cells transfused independently associated with post-procedure acute kidney injury (OR = 1.67 per unit, 95% CI 1.13–2.47, P = 0.01) after adjusting for pre-procedure estimated glomerular filtration rate (OR = 0.97 per ml/min/1.73m2, 95% CI 0.94–1.00, P = 0.05), nadir hemoglobin (OR = 0.88 per g/dL increase, CI 0.61–1.27, P = 0.50), and post-procedure maximum number of concurrent inotropes and vasopressors (OR = 2.09 per inotrope or vasopressor, 95% CI 1.19–3.67, P = 0.01). Conclusion: Packed red blood cell transfusion, along with post-procedure use of inotropes and vasopressors, independently associate with acute kidney injury after transcatheter aortic valve replacement. Further studies are needed to elucidate the pathobiology underlying these associations

    The Alberta Ambassador Program: delivering Health Technology Assessment results to rural practitioners

    BACKGROUND: The purpose of Health Technology Assessment (HTA) is to make the best possible summary of the evidence regarding specific health interventions in order to influence health care and policy decisions. The need for decision makers to find relevant HTA data when it is needed is a barrier to its usefulness. These barriers are highest in rural areas and amongst isolated practitioners. METHODS: A multidisciplinary team developed an interactive case-based instructional strategy on the topic of chronic non-cancer pain (CNCP) management using clinical evidence derived by HTA. The evidence for each of 18 CNCP interventions was distilled into single-sheet summaries. Clinicians and HTA specialists ('Ambassadors') conducted 11 two-hour interactive sessions on CNCP in eight of Alberta's nine health regions. Pre- and post-session evaluations were conducted. RESULTS: The sessions were attended by 130 individuals representing 14 health and administrative disciplines. The ambassador model was well received. The use of content experts as ambassadors was highly rated. The educational strategy was judged to be effective. Awareness of the best evidence in CNCP management was increased. Although some participants reported practice changes as a result of the workshops, the program was not designed to measure changes in patient outcome. CONCLUSION: The ambassador program was successful in increasing awareness of the best evidence in CNCP management, and positively influenced treatment decisions. Its teaching methods were felt to be unique and innovative by participants. Its methods could be applied to other clinical content areas in order to increase the uptake of the results of HTA
