Possible chromosomal and germline integration of human herpesvirus 7

Copyright: Copyright 2017 Elsevier B.V., All rights reserved.Human herpesvirus 7 (HHV-7) is a betaherpesvirus, and is phylogenetically related to both HHV-6A and HHV-6B. The presence of telomeric repeat sequences at both ends of its genome should make it equally likely to integrate into the human telomere as HHV-6. However, numerous studies have failed to detect germline integration of HHV-7, suggesting an important difference between the HHV-6A/-6B and HHV-7 genomes. In search of possible germline integrated HHV-7, we developed a sensitive and quantitative real-time PCR assay and discovered that primers designed against some parts of the HHV-7 genome can frequently miss HHV-7 positive clinical samples even though they work efficiently in cell-culture-derived HHV-7 positive materials. Using a primer pair against the U90 ORF of HHV-7, we identified a possible case of germline integration of HHV-7 with one copy of viral genome per cell in both peripheral blood cells and hair follicles. Chromosomal integration of HHV-7 in these individuals was confirmed by fluorescence in situ hybridization analysis. Germline integration of HHV-7 was further confirmed by detection of ~2.6 copies of HHV-7 in the hair follicles of one of the parents. Our results shed light on the complex nature of the HHV-7 genome in human-derived materials in comparison to cell-culture-derived materials and show the need for stringent criteria in the selection of primers for epidemiological HHV-7 studies.Peer reviewe
Structural and ultrastructural alterations in human olfactory pathways and possible associations with herpesvirus 6 infection

The olfactory bulb/tract samples from 20 subjects with an unspecified encephalopathy determined by pathomorphological examination of the brain autopsy, 17 healthy age-matched and 16 younger controls were used. HHV-6 DNA was detected in 60, 29, and 19% of cases in these groups, respectively. In the whole encephalopathy group, significantly more HHV-6 positive neurons and oligodendrocytes were found in the gray matter, whereas, significantly more HHV-6 positive astrocytes, oligodendrocytes, microglia/macrophages and endothelial cells were found in the white matter. Additionally, significantly more HHV-6 positive astrocytes and, in particular, oligodendrocytes were found in the white matter when compared to the gray matter. Furthermore, when only HHV-6 PCR+ encephalopathy cases were studied, we observed similar but stronger associations between HHV-6 positive oligodendrocytes and CD68 positive cells in the white matter. Cellular alterations were additionally evidenced by anti-S100 immunostaining, demonstrating a significantly higher number of S100 positive cells in the gray matter of the whole encephalopathy group when compared to the young controls, and in the white matter when compared to both control groups. In spite the decreased S100 expression in the PCR+ encephalopathy group when compared to PCR- cases and controls, groups demonstrated significantly higher number of S100 positive cells in the white compared to the gray matter. Ultrastructural changes confirming the damage of myelin included irregularity of membranes and ballooning of paranodal loops. This study shows that among the cellular targets of the nervous system, HHV-6 most severely affects oligodendrocytes and the myelin made by them.publishersversionPeer reviewe
European Network on Myalgic Encephalomyelitis/Chronic Fatigue Syndrome (EUROMENE): Expert Consensus on the Diagnosis, Service Provision, and Care of People with ME/CFS in Europe.
Designed by a group of ME/CFS researchers and health professionals, the European Network on Myalgic Encephalomyelitis/Chronic Fatigue Syndrome (EUROMENE) has received funding from the European Cooperation in Science and Technology (COST)-COST action 15111-from 2016 to 2020. The main goal of the Cost Action was to assess the existing knowledge and experience on health care delivery for people with Myalgic Encephalomyelitis/Chronic Fatigue Syndrome (ME/CFS) in European countries, and to enhance coordinated research and health care provision in this field. We report our findings and make recommendations for clinical diagnosis, health services and care for people with ME/CFS in Europe, as prepared by the group of clinicians and researchers from 22 countries and 55 European health professionals and researchers, who have been informed by people with ME/CFS
Polycation-mediated gene delivery: challenges and considerations for the process of plasmid DNA transfection
The process of gene delivery has increasingly become a focus in biomedical research due to the growing demand by an ageing global population for targeted therapies of genetic related diseases, such as cancer, immunodeficiencies, and cystic fibrosis. In light of the safety issues that viral vectors still face in progressing through clinical trials, polycations have alternatively attracted keen attention, due to their lower safety risks and ability to interact with cells more effectively and with more stability, comparedwith other chemically designed nonviral vectors. In this review, a reflection of the literature pertaining to various types of polycations designed and optimized is presented, including the obstacles they face in facilitating plasmidDNA transfection. In order to design new polycations or optimize current ones that will be successful— both economically and for use in future clinical therapies—further in vivo research is needed to fully elucidate the mechanisms of each stage in the delivery process.Karl Modra, Sheng Dai, Hu Zhang, Bingyang Shi, Jingxiu B
Epidemiology of participation: an Australian community study
STUDY OBJECTIVE—To determine the levels of participation in social and civic community life in a metropolitan region, and to assess differential levels of participation according to demographic, socioeonomic and health status. To contribute to policy debates on community participation, social capital and health using these empirical data.
DESIGN—Cross sectional, postal, self completed survey on health and participation.
SETTING—Random sample of the population from the western suburbs of Adelaide, the capital city of South Australia, a population of approximately 210 000.
PARTICIPANTS—2542 respondents from a sample of 4000 people aged 18 years and over who were registered on the electoral roll.
MAIN RESULTS—The response rate to the survey was 63.6% (n=2542). Six indices of participation, on range of social and civic activities, with a number of items in each, were created. Levels of participation were highest in the informal social activities index (46.7-83.7% for individual items), and lowest in the index of civic activities of a collective nature (2.4-5.9% for individual items). Low levels of involvement in social and civic activities were reported more frequently by people of low income and low education levels.
CONCLUSIONS—Levels of participation in social and civic community life in an urban setting are significantly influenced by individual socioeconomic status, health and other demographic characteristics. An understanding of the pattern of participation is important to inform social and health policy making. Increasing levels of participation will reduce social exclusion and is likely to improve the overall quality of community life.

Recovery ability of common carp (Cyprinus carpio) after a short-term exposure to terbuthylazine
Effects of a high terbuthylazine concentration (3.3 mg/l) on Cyprinus carpio were studied using
a commercial herbicide formulation Click 500 SC (terbuthylazine 500 g/l). The fish were exposed to
the pesticide for 24 h and allowed to recover for 6 days. Biometric parameters, plasma biochemical
parameters and biomarkers of oxidative stress as well as histopathological changes in selected tissues
were assessed on day 1 and 7. After a 24-h exposure, there were significant alterations found in the
activities of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) as well as in the
plasma concentrations of glucose, natrium, chlorides, calcium and phosphorus. Hepatosomatic index,
plasma albumin and lactate reflected the treatment with a delay. Ion levels and ALT were found to be
restored after a 6-day recovery period, which was too short for AST activity and glucose to diminish to
the control levels. The histopathological examination revealed disorders in the gills of the exposed
fish, however, the changes were not detected after a 6-day recovery period. The study shows high
regeneration potential of the fish
Chronic viral infections in myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS)
Background and main text: Myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) is a complex and controversial clinical condition without having established causative factors. Increasing numbers of cases during past decade have created awareness among patients as well as healthcare professionals. Chronic viral infection as a cause of ME/CFS has long been debated. However, lack of large studies involving well-designed patient groups and validated experimental set ups have hindered our knowledge about this disease. Moreover, recent developments regarding molecular mechanism of pathogenesis of various infectious agents cast doubts over validity of several of the past studies.
Conclusions: This review aims to compile all the studies done so far to investigate various viral agents that could be associated with ME/CFS. Furthermore, we suggest strategies to better design future studies on the role of viral infections in ME/CFS
