    Discovery of a lectin domain that regulates enzyme activity in mouse N-acetylglucosaminyltransferase-IVa (MGAT4A)

    N-Glycosylation is a common post-translational modification, and the number of GlcNAc branches in N-glycans impacts glycoprotein functions. N-Acetylglucosaminyltransferase-IVa (GnT-IVa, also designated as MGAT4A) forms a β1-4 GlcNAc branch on the α1-3 mannose arm in N-glycans. Downregulation or loss of GnT-IVa causes diabetic phenotypes by dysregulating glucose transporter-2 in pancreatic β-cells. Despite the physiological importance of GnT-IVa, its structure and catalytic mechanism are poorly understood. Here, we identify the lectin domain in mouse GnT-IVa’s C-terminal region. The crystal structure of the lectin domain shows structural similarity to a bacterial GlcNAc-binding lectin. Comprehensive glycan binding assay using 157 glycans and solution NMR reveal that the GnT-IVa lectin domain selectively interacts with the product N-glycans having a β1-4 GlcNAc branch. Point mutation of the residue critical to sugar recognition impairs the enzymatic activity, suggesting that the lectin domain is a regulatory subunit for efficient catalytic reaction.     Realization of the mean-field universality class in spin-crossover materials

    In spin-crossover materials, the volume of a molecule changes depending on whether it is in the high-spin (HS) or low-spin (LS) state. This change causes distortion of the lattice. Elastic interactions among these distortions play an important role for the cooperative properties of spin-transition phenomena. We find that the critical behavior caused by this elastic interaction belongs to the mean-field universality class, in which the critical exponents for the spontaneous magnetization and the susceptibility are β=1/2\beta = 1/2 and γ=1\gamma = 1, respectively. Furthermore, the spin-spin correlation function is a constant at long distances, and it does not show an exponential decay in contrast to short-range models. The value of the correlation function at long distances shows different size-dependences: O(1/N)O(1/N), O(1/N)O(1/\sqrt{N}), and constant for temperatures above, at, and below the critical temperature, respectively. The model does not exhibit clusters, even near the critical point. We also found that cluster growth is suppressed in the present model and that there is no critical opalescence in the coexistence region. During the relaxation process from a metastable state at the end of a hysteresis loop, nucleation phenomena are not observed, and spatially uniform configurations are maintained during the change of the fraction of HS and LS. These characteristics of the mean-field model are expected to be found not only in spin-crossover materials, but also generally in systems where elastic distortion mediates the interaction among local states.Comment: 13 pages, 16 figure

    Suppression of growth by multiplicative white noise in a parametric resonant system

    The author studied the growth of the amplitude in a Mathieu-like equation with multiplicative white noise. The approximate value of the exponent at the extremum on parametric resonance regions was obtained theoretically by introducing the width of time interval, and the exponents were calculated numerically by solving the stochastic differential equations by a symplectic numerical method. The Mathieu-like equation contains a parameter α\alpha that is determined by the intensity of noise and the strength of the coupling between the variable and the noise. The value of α\alpha was restricted not to be negative without loss of generality. It was shown that the exponent decreases with α\alpha, reaches a minimum and increases after that. It was also found that the exponent as a function of α\alpha has only one minimum at α≠0\alpha \neq 0 on parametric resonance regions of α=0\alpha = 0. This minimum value is obtained theoretically and numerically. The existence of the minimum at α≠0\alpha \neq 0 indicates the suppression of the growth by multiplicative white noise.Comment: The title and the description in the manuscript are change

    Parametric amplification with a friction in heavy ion collisions

    We study the effects of the expansion of the system and the friction on the parametric amplification of mesonic fields in high energy heavy ion collisions within the linear σ\sigma model . The equation of motion which is similar to Mathieu equation is derived to describe the time development of classical fields in the last stage of a heavy ion collision after the freezeout time. The enhanced mode is extracted analytically by comparison with Mathieu equation and the equation of motion is solved numerically to examine whether soft modes will be enhanced or not. It is found that the strong peak appears around 267 MeV in the pion transverse momentum distribution in cases with weak friction and high maximum temperature. This enhancement can be extracted by taking the ratio between different modes in the pion transverse momentum distribution.Comment: 10 pages, 9 figures LaTeX: appendix adde

    MUC4 and MUC1 expression in adenocarcinoma of the stomach correlates with vessel invasion and lymph node metastasis: an immunohistochemical study of early gastric cancer.

    We have previously reported that MUC4 expression is a poor prognostic factor in various carcinomas. Our previous study also showed that MUC1 expression in gastric cancers, including the early and advanced stages is a poor prognostic factor. In the present study, the expression profiles of MUC4 and MUC1 were examined by immunohistochemistry (IHC) using two anti-MUC4 monoclonal antibodies (MAbs), 8G7 and 1G8, and anti-MUC1 MAb DF3 in 104 gastrectomy specimens of early gastric adenocarcinoma with submucosal invasion (pT1b2), including 197 histological subtype lesions. Before the IHC study of the human specimens, we evaluated the specificity of the two MAbs by Western blotting and IHC of two MUC4 mRNA expressing gastric cancer cell lines. MAb 8G7 reacted clearly, whereas MAb 1G8 did not show any reactivity, in either Western blotting or IHC. In the IHC of the gastric cancers, the expression rates of MUC4/8G7 detected by MAb 8G7, MUC4/1G8 detected by MAb 1G8 and MUC1/DF3 detected by MAb DF3 in well differentiated types (70%, 38/54; 67%, 36/54; 52%, 28/54) were significantly higher than those in poorly differentiated types (18%, 10/55; 36%, 20/55; 13%, 7/55) (

    By studying the dynamics of the metastable magnetization of a statistical mechanical model we propose a switching mechanism of photoinduced magnetization. The equilibrium and nonequilibrium properties of the Blume-Capel (BC) model, which is a typical model exhibiting metastability, are studied by mean field theory and Monte Carlo simulation. We demonstrate reversible changes of magnetization in a sequence of changes of system parameters, which would model the reversible photoinduced magnetization. Implications of the calculated results are discussed in relation to the recent experimental results for prussian blue analogs.Comment: 12 pages, 13 figure
