    Analyzing Poverty in the Southern United States

    This thesis deals with two related topics under the theme of ―Analyzing Poverty in the Southern United States‖. The first part explores the role of government healthcare and education expenditure for poverty reduction, focusing particularly on how these relationships change over space and time in the Southern United States. It is found that healthcare expenditure is a significant contributor to poverty alleviation in both 1990 and 2000. The healthcare expenditure has a relatively high poverty-reducing effect in the Texas cluster and in the west part of the Mississippi Delta cluster in both years, while the poverty-reducing effect of healthcare expenditures disappears in 2000 in the Central Appalachia cluster. The effect of government expenditures on education decreased over time in the west part of the Mississippi Delta cluster but the education expenditure began to have a poverty-reducing effect in the Central Appalachia cluster in 2000. The second part focuses on disentangling the relationship between urban sprawl and poverty in the Southern United States. Results show that an increase in urban sprawl, as measured by wildland-urban interface (WUI), is associated with an increase in the urban poverty rate. The positive interrelationship between urban poverty and area of sprawl in metro counties supports the theoretical framework that urban poverty is both cause and effect of urban sprawl. With no other direct or indirect association between the poverty rate and urban sprawl, the positive interrelationship is explained by the movement of business centers to the suburban areas by sprawl development and immobility of the poor and the middle and upper class households‘ preference for the neighborhoods with lower poverty rates

    Extreme coefficients in Geographically Weighted Regression and their effects on mapping

    This study deals with the issue of extreme coefficients in geographically weighted regression (GWR) and their effects on mapping coefficients using three datasets with different spatial resolutions. We found that although GWR yields extreme coefficients regardless of the resolution of the dataset or types of kernel function, 1) the GWR tends to generate extreme coefficients for less spatially dense datasets, 2) coefficient maps based on polygon data representing aggregated areal units are more sensitive to extreme coefficients, and 3) coefficient maps using bandwidths generated by a fixed calibration procedure are more vulnerable to the extreme coefficients than adaptive calibration.extreme coefficient, fixed and adaptive calibrations, geographically weighted regression, Mapping, Research Methods/ Statistical Methods,

    Inactivation of Medial Prefrontal Cortex Impairs Time Interval Discrimination in Rats

    Several lines of evidence suggest the involvement of prefrontal cortex in time interval estimation. The underlying neural processes are poorly understood, however, in part because of the paucity of physiological studies. The goal of this study was to establish an interval timing task for physiological recordings in rats, and test the requirement of intact medial prefrontal cortex (mPFC) for performing the task. We established a temporal bisection procedure using six different time intervals ranging from 3018 to 4784 ms that needed to be discriminated as either long or short. Bilateral infusions of muscimol (GABAA receptor agonist) into the mPFC significantly impaired animal's performance in this task, even when the animals were required to discriminate between only the longest and shortest time intervals. These results show the requirement of intact mPFC in rats for time interval discrimination in the range of a few seconds

    Spatial patterns of large‐scale land transactions and their potential socio‐environmental outcomes in Cambodia, Ethiopia, Liberia, and Peru

    Recent large‐scale land transactions, often framed as ’land grabbing,’ are historically unprecedented. Millions of hectares of land have changed hands for agriculture‐driven development over the past decade, and their implementation generates substantial risk of land degradation. This paper investigates land transaction patterns and evaluate their potential socio‐environmental impacts in Cambodia, Ethiopia, Liberia, and Peru. We undertake a novel spatially explicit approach to quantify land transactions and conduct scenario‐based analyses to explore their implementation consequences on people, land, and carbon emission. Our results demonstrate that existing global datasets on land transactions substantially underestimate their incidence but can either exaggerate or underreport transacted areas. Although confirming that land transactions are more likely to occur in sparsely populated, poorer, and more forested areas, our scenario‐based analyses reveal that if fully implemented for agricultural development, land transactions in the four countries will affect more than one million people, yield over 2 Gt of carbon emissions, and disrupt vast swathes of forests.     Time Series Analysis on the Conformational Change of c-Src Tyrosine Kinase

    c-Src tyrosine kinase plays an important role in signal transduction pathways, where its activity is regulated by phosphorylation of the two tyrosine residues. We performed targeted molecular dynamics simulation to obtain trajectory of conformational change from inactive to active form. To investigate the conformational change of c-Src tyrosine kinase, we applied network analysis to time series of correlation among residues. The time series of correlation between residues during the conformational change generated by targeted molecular dynamic simulation. With centrality measures such as betweenness centrality, degree centrality, and closeness centrality, we observed a few important residues that significantly contribute to the conformational change of c-Src tyrosine kinase for the different time steps

    Capnography for Assessing Nocturnal Hypoventilation and Predicting Compliance with Subsequent Noninvasive Ventilation in Patients with ALS

    BACKGROUND: Patients with amyotrophic lateral sclerosis (ALS) suffer from hypoventilation, which can easily worsen during sleep. This study evaluated the efficacy of capnography monitoring in patients with ALS for assessing nocturnal hypoventilation and predicting good compliance with subsequent noninvasive ventilation (NIV) treatment. METHODS: Nocturnal monitoring and brief wake screening by capnography/pulse oximetry, functional scores, and other respiratory signs were assessed in 26 patients with ALS. Twenty-one of these patients were treated with NIV and had their treatment compliance evaluated. RESULTS: Nocturnal capnography values were reliable and strongly correlated with the patients' respiratory symptoms (R(2) = 0.211-0.305, p = 0.004-0.021). The duration of nocturnal hypercapnea obtained by capnography exhibited a significant predictive power for good compliance with subsequent NIV treatment, with an area-under-the-curve value of 0.846 (p = 0.018). In contrast, no significant predictive values for nocturnal pulse oximetry or functional scores for nocturnal hypoventilation were found. Brief waking supine capnography was also useful as a screening tool before routine nocturnal capnography monitoring. CONCLUSION: Capnography is an efficient tool for assessing nocturnal hypoventilation and predicting good compliance with subsequent NIV treatment of ALS patients, and may prove useful as an adjunctive tool for assessing the need for NIV treatment in these patients

    Transduction of the MPG-tagged fusion protein into mammalian cells and oocytes depends on amiloride-sensitive endocytic pathway

    BACKGROUND: MPG is a cell-permeable peptide with proven efficiency to deliver macromolecular cargoes into cells. In this work, we examined the efficacy of MPG as an N-terminal tag in a fusion protein to deliver a protein cargo and its mechanism of transduction. RESULTS: We examined transduction of MPG-EGFP fusion protein by live imaging, flow cytometry, along with combination of cell biological and pharmacological methods. We show that MPG-EGFP fusion proteins efficiently enter various mammalian cells within a few minutes and are co-localized with FM4-64, a general marker of endosomes. The transduction of MPG-EGFP occurs rapidly and is inhibited at a low temperature. The entry of MPG-EGFP is inhibited by amiloride, but cytochalasin D and methyl-β-cyclodextrin did not inhibit the entry, suggesting that macropinocytosis is not involved in the transduction. Overexpression of a mutant form of dynamin partially reduced the transduction of MPG-EGFP. The partial blockade of MPG-EGFP transduction by a dynamin mutant is abolished by the treatment of amiloride. MPG-EGFP transduction is also observed in the mammalian oocytes. CONCLUSION: The results show that the transduction of MPG fusion protein utilizes endocytic pathway(s) which is amiloride-sensitive and partially dynamin-dependent. Collectively, the MPG fusion protein could be further developed as a novel tool of "protein therapeutics", with potentials to be used in various cell systems including mammalian oocytes
