SCYL2 Genes Are Involved in Clathrin-Mediated Vesicle Trafficking and Essential for Plant Growth
Protein transport between organelles is an essential process in all eukaryotic cells and is mediated by the regulation of processes such as vesicle formation, transport, docking, and fusion. In animals, SCY1-LIKE2 (SCYL2) binds to clathrin and has been shown to play roles in trans-Golgi network-mediated clathrin-coated vesicle trafficking. Here, we demonstrate that SCYL2A and SCYL2B, which are Arabidopsis (Arabidopsis thaliana) homologs of animal SCYL2, are vital for plant cell growth and root hair development. Studies of the SCYL2 isoforms using multiple single or double loss-of-function alleles show that SCYL2B is involved in root hair development and that SCYL2A and SCYL2B are essential for plant growth and development and act redundantly in those processes. Quantitative reverse transcription-polymerase chain reaction and a beta-glucuronidase-aided promoter assay show that SCYL2A and SCYL2B are differentially expressed in various tissues. We also show that SCYL2 proteins localize to the Golgi, trans-Golgi network, and prevacuolar compartment and colocalize with Clathrin Heavy Chain1 (CHC1). Furthermore, bimolecular fluorescence complementation and coimmunoprecipitation data show that SCYL2B interacts with CHC1 and two Soluble NSF Attachment Protein Receptors (SNAREs): Vesicle Transport through t-SNARE Interaction11 (VTI11) and VTI12. Finally, we present evidence that the root hair tip localization of Cellulose Synthase-Like D3 is dependent on SCYL2B. The Development of Web3D-based Open-pit Mine Monitoring System
Large-scale open-pit mines are critical infrastructure for acquiring natural resources. However, this type of mine can experience environmental and safety problems during operations and thus requires continuous monitoring. In this study, a web three-dimensional(3D)-based monitoring system is constructed using geospatial information open platform and open-source geospatial information software which targets open-pit mines in Gangwon-do, Korea. The purpose is to develop a monitoring system of open-pit mines that enables any person to monitor the topographic and environmental changes caused by mine operations and to develop and restore the area’s ecology. Open-pit mines were classified into active or inactive mines and monitoring items and methodologies were established for each type of mine. Cesium which is a WebGL-based open-source platform was chosen as it supports dynamic data visualization and hardware-accelerated graphics related to elapsed time which is the essential factor in the monitoring. The open-pit mine monitoring system was developed based on the geospatial database which contains information required for mine monitoring as time elapses, and by developing the open-source-based system software. The geospatial information database for monitoring consists of digital imagery and terrain data and also includes vector data and the restoration plan datas. The basic geospatial information used in the monitoring includes high resolution ortho image(GSD 0.5 m or above) for all areas of the mines. This is acquired by periodically using an airborne laser scanning system and a LiDAR DEM (grid size 1m × 1 m). In addition, geospatial information data were acquired by using an UAV and terrestrial LiDAR for small-scale areas; these tools are frequently used for rapid and irregular data acquisition. The geospatial information acquired for the monitoring of the open-pit mines represents various spatial resolutions and different terrain data. The database was constructed by converging this geospatial information with the Cesium-based geospatial information open platform of the ESRI World Imagery map and with SDK World Terrain meshes. The problems that resulted from the process of fusing aerial imagery and terrain data were solved in the Cesium-based open source environment. The prototype menu for the monitoring system was designed according to the monitoring item which was determined by the type of mine. The scene of the mine and changes in terrain were controlled and analyzed using the raster function of PostGIS according to the elapsed time. Using the GeoServer, the aerial imagery, terrain and restoration information for each period were serviced using the web standard interface, and the monitoring system was completed by visualizing these elements in Cesium in 3D format according to the elapsed time. This study has established a monitoring methodology for open-pit mines according to the type of mine and proposes a method for upgrading the imagery and terrain data required for monitoring. The study also showed the possibility of developing a Web3D-based open-pit mine monitoring system that is applicable to a wide range of mashup service developments
Surgical management of pilon fractures with large segmental bone defects using fibular strut allografts: a report of two cases
We present two patients with open pilon fractures with large bone defects treated successfully with fibular strut allografts. The patients were initially treated by massive irrigation, wound debridement, and temporary external fixation. After complete wound healing, the bone defects were managed. Because autologous iliac crest or fibular bone grafts were impossible to be harvested due to multiple fractures, the bone defects were reconstructed with fibular strut allografts. Fixation was performed with a periarticular distal tibia locking plate. At 2 months postoperatively, the patients ambulated with partial weight-bearing; at 6 months, they had full range of motion of the ankle joint and full weight-bearing
Liquid crystal display using combined fringe and in-plane electric fields
A high performance liquid crystal display using combined fringe and in-plane horizontal electric fields is proposed. The strong electric fields cause more liquid crystals to reorient almost in plane above and between the pixel electrodes. As a result, the operation voltage is lower and transmittance is higher than those of fringe field switching and in-plane switching modes, while preserving a wide viewing angle. Such a high performance device is particularly attractive for large panel liquid crystal displays
A case of idiopathic isolated hypoglossal nerve palsy in a Korean child
Hypoglossal nerve palsy (HNP) is an uncommon neurological abnormality that can provoke characteristic clinical signs, including unilateral atrophy of the tongue musculature. We present the case of a healthy 11-year-old Korean male who was admitted to the outpatient department of our institution with acute onset dysarthria, tongue fasciculations, and right-sided tongue weakness upon awakening. His evaluation included a virology work-up, neck magnetic resonance imaging (MRI), brain MRI, and otorhinolaryngological physical examination; all tests were normal and showed no evidence of inflammation. Fifteen days after the onset of symptoms, the patient recovered completely. Herein, we report a case of idiopathic isolated HNP in a Korean male
Clinical Characteristics of Radiation Oncology in Korea during Past 10 Years
To understand trends in the clinical characteristics of radiation oncology over the last 10 yr in Korea, annual survey questionnaires were sent to all of Korean radiation oncology facilities since 1990. Questionnaires addressed basic radiation therapy facilities and the clinical information. Responses were obtained from all facilities, and data collected from 1997 to 2006 was analyzed. The numbers of new patients that have undergone radiation therapy and the numbers of hospitals with a department of radiation oncology have steadily increased over the past 10 yr, and totaled 37,215 patients and 60 hospitals, respectively, in 2006. However, the proportion of patients irradiated among total cancer patients has remained below 30% over the last 10 yr. The numbers of prostate cancer, breast cancer, and hepatoma have increased by more than 3 fold over the past 10 yr. Moreover, the percentage of irradiated patients treated by brachytherapy was 10.3% in 1997, but this gradually fell to only 4.2% in 2006. The information collected described the role played by radiation oncology in Korea. Continuous surveys are required to enable trends to be detected
Availability of the Pediatric Emergency Care Applied Research Network (PECARN) rule for computed tomography scanning decision in children younger than 2 years with minor head injury
Purpose Traumatic brain injury is the most common cause of pediatric injury. Although computed tomography (CT) scan is an effective modality for screening fatal craniocerebral trauma, there is growing concern about radiation exposure associated with the consequent cancer particularly in children. We assessed validity of previous large prospective study named Pediatric Emergency Care Applied Research Network (PECARN) retrospectively to determine the necessity of CT scans for children younger than 2 years with minor head injury. Methods We reviewed medical records of children younger than 2 years discharged from our emergency department with S00–09 diagnosis code of ICD-10 from August 2008 to December 2014. Patients who had only soft tissue injury without blunt trauma, did not CT scan take brain CT, whose head trauma was not mild, and who was uncertain to meet the rule were excluded. All included patients were divided into the PECARN rule positive group and negative group. Each group was compared by sensitivity, specificity, positive predictive value and negative predictive value to predict four outcomes of clinically important traumatic brain injury (ciTBI), abnormal CT findings, intracranial hemorrhage, and isolated simple skull fracture. Results A total of 1,491 patients were included, 656 PECARN rule positive and 835 negative patients. There is statistical difference between PECARN rule positive and negative the 2 group for ciTBI (P < 0.001), abnormal CT findings (P < 0.001), intracranial hemorrhage (P < 0.001), and isolated simple skull fracture (P < 0.001) with high sensitivity (100.0%, 89.5%, 91.7%,85.7%) and negative predictive value (100.0%, 99.3%, 99.6%, 99.6%). Conclusion We confirmed that PECARN rule is a useful tool to determine the necessity of CT scan and reduce unnecessary CT scan for children younger than 2 years with minor head injury
Vulvar Skin Metastasis of Lung Squamous Cell Carcinoma
In this study, we examined a case where lung cancer metastasized to the vulvar area. A 79-year-old woman that was a heavy smoker was referred to our department with a short (two-week) history of dyspnea, and with a painful nodule on her right labium majora, which she had noticed almost three years earlier. Histopathologically, the specimen appeared as a poorly differentiated squamous cell carcinoma. The chest X-ray, chest computerized tomography scan, and percutaneous transthoracic needle biopsy via bronchoscopy revealed squamous cell carcinoma in the right upper lobe of the lung. Based on these clinical and histopathological findings, we concluded that her condition was skin metastasis at the right labium majora from the lung cancer, which occurs very rarely
Trends for the Past 10 Years and International Comparisons of the Structure of Korean Radiation Oncology
Objective: Study aims include determination of nationwide structural characteristics of radiation oncology facilities, types of radiation therapy equipment, availability of human resources and trends and comparisons with previous surveys. Methods: An annual nationwide survey was conducted to collect the statistics of infrastructure since 1997. All requested questionnaires have been identical for 10 years. The questionnaires included status on basic radiation therapy facilities, human resources and radiation therapy equipment. Journal and statistical data reviews were performed to evaluate the structure of other countries. Results: Radiation oncology facilities have steadily increased for 10 years and reached 60 sites in 2006. Also a steady increase of 1.5 times for linear accelerators, 5.8 times for computed tomography simulators and 3.0 times for radiation treatment planning systems was noted. Meanwhile, cobalt-60 teletherapy units and hyperthermia equipment had steadily deceased for 10 years. The number of human resources has steadily increased for the past 10 years, especially for radiation therapy technologists. However, radiation therapy equipment and human resources per population are relatively low compared with advanced countries. Conclusions: This study will assist preparation of the administrative planning policy of radiation oncology and should be useful to indicate the direction of future development and educational training programs in Korea and possibly in other countries
Human embryonic stem cells express a unique set of microRNAs.
Abstract Human embryonic stem (hES) cells are pluripotent cell lines established from the explanted inner cell mass of human blastocysts. Despite their importance for human embryology and regenerative medicine, studies on hES cells, unlike those on mouse ES (mES) cells, have been hampered by difficulties in culture and by scant knowledge concerning the regulatory mechanism. Recent evidence from plants and animals indicates small RNAs of approximately 22 nucleotides (nt), collectively named microRNAs, play important roles in developmental regulation. Here we describe 36 miRNAs (from 32 stem-loops) identified by cDNA cloning in hES cells. Importantly, most of the newly cloned miRNAs are specifically expressed in hES cells and downregulated during development into embryoid bodies (EBs), while miRNAs previously reported from other human cell types are poorly expressed in hES cells. We further show that some of the ES-specific miRNA genes are highly related to each other, organized as clusters, and transcribed as polycistronic primary transcripts. These miRNA gene families have murine homologues that have similar genomic organizations and expression patterns, suggesting that they may operate key regulatory networks conserved in mammalian pluripotent stem cells. The newly identified hES-specific miRNAs may also serve as molecular markers for the early embryonic stage and for undifferentiated hES cells
