    An Assessment of CO2 Storage and Sea‐Air Fluxes for the Atlantic Ocean and Mediterranean Sea Between 1985 and 2018

    As part of the second phase of the Regional Carbon Cycle Assessment and Processes project (RECCAP2), we present an assessment of the carbon cycle of the Atlantic Ocean, including the Mediterranean Sea, between 1985 and 2018 using global ocean biogeochemical models (GOBMs) and estimates based on surface ocean carbon dioxide (CO2) partial pressure (pCO2 products) and ocean interior dissolved inorganic carbon observations. Estimates of the basin-wide long-term mean net annual CO2 uptake based on GOBMs and pCO2 products are in reasonable agreement (−0.47 ± 0.15 PgC yr−1 and −0.36 ± 0.06 PgC yr−1, respectively), with the higher uptake in the GOBM-based estimates likely being a consequence of a deficit in the representation of natural outgassing of land derived carbon. In the GOBMs, the CO2 uptake increases with time at rates close to what one would expect from the atmospheric CO2 increase, but pCO2 products estimate a rate twice as fast. The largest disagreement in the CO2 flux between GOBMs and pCO2 products is found north of 50°N, coinciding with the largest disagreement in the seasonal cycle and interannual variability. The mean accumulation rate of anthropogenic CO2 (Cant) over 1994–2007 in the Atlantic Ocean is 0.52 ± 0.11 PgC yr−1 according to the GOBMs, 28% ± 20% lower than that derived from observations. Around 70% of this Cant is taken up from the atmosphere, while the remainder is imported from the Southern Ocean through lateral transport

The sample is assumed to be cleaned from objects with quasar properties.     Unequal allelic expression of wild-type and mutated ÎČ-myosin in familial hypertrophic cardiomyopathy

    Familial hypertrophic cardiomyopathy (FHC) is an autosomal dominant disease, which in about 30% of the patients is caused by missense mutations in one allele of the ÎČ-myosin heavy chain (ÎČ-MHC) gene (MYH7). To address potential molecular mechanisms underlying the family-specific prognosis, we determined the relative expression of mutant versus wild-type MYH7-mRNA. We found a hitherto unknown mutation-dependent unequal expression of mutant to wild-type MYH7-mRNA, which is paralleled by similar unequal expression of ÎČ-MHC at the protein level. Relative abundance of mutated versus wild-type MYH7-mRNA was determined by a specific restriction digest approach and by real-time PCR (RT-qPCR). Fourteen samples from M. soleus and myocardium of 12 genotyped and clinically well-characterized FHC patients were analyzed. The fraction of mutated MYH7-mRNA in five patients with mutation R723G averaged to 66 and 68% of total MYH7-mRNA in soleus and myocardium, respectively. For mutations I736T, R719W and V606M, fractions of mutated MYH7-mRNA in M. soleus were 39, 57 and 29%, respectively. For all mutations, unequal abundance was similar at the protein level. Importantly, fractions of mutated transcripts were comparable among siblings, in younger relatives and unrelated carriers of the same mutation. Hence, the extent of unequal expression of mutated versus wild-type transcript and protein is characteristic for each mutation, implying cis-acting regulatory mechanisms. Bioinformatics suggest mRNA stability or splicing effectors to be affected by certain mutations. Intriguingly, we observed a correlation between disease expression and fraction of mutated mRNA and protein. This strongly suggests that mutation-specific allelic imbalance represents a new pathogenic factor for FHC

    The impact of local control on overall survival after stereotactic body radiotherapy for liver and lung metastases from colorectal cancer: a combined analysis of 388 patients with 500 metastases

    Background: The aim of this analysis was to model the effect of local control (LC) on overall survival (OS) in patients treated with stereotactic body radiotherapy (SBRT) for liver or lung metastases from colorectal cancer. Methods: The analysis is based on pooled data from two retrospective SBRT databases for pulmonary and hepatic metastases from 27 centers from Germany and Switzerland. Only patients with metastases from colorectal cancer were considered to avoid histology as a confounding factor. An illness-death model was employed to model the relationship between LC and OS. Results: Three hundred eighty-eight patients with 500 metastatic lesions (lung n = 209, liver n = 291) were included and analyzed. Median follow-up time for local recurrence assessment was 12.1 months. Ninety-nine patients with 112 lesions experienced local failure. Seventy-one of these patients died after local failure. Median survival time was 27.9 months in all patients and 25.4 months versus 30.6 months in patients with and without local failure after SBRT. The baseline risk of death after local failure exceeds the baseline risk of death without local failure at 10 months indicating better survival with LC. Conclusion: In CRC patients with lung or liver metastases, our findings suggest improved long-term OS by achieving metastatic disease control using SBRT in patients with a projected OS estimate of > 12 months
