Amyloid precursor protein interaction network in human testis: sentinel proteins for male reproduction
Background
Amyloid precursor protein (APP) is widely recognized for playing a central role in Alzheimer's disease pathogenesis. Although APP is expressed in several tissues outside the human central nervous system, the functions of APP and its family members in other tissues are still poorly understood. APP is involved in several biological functions which might be potentially important for male fertility, such as cell adhesion, cell motility, signaling, and apoptosis. Furthermore, APP superfamily members are known to be associated with fertility. Knowledge on the protein networks of APP in human testis and spermatozoa will shed light on the function of APP in the male reproductive system.
Results
We performed a Yeast Two-Hybrid screen and a database search to study the interaction network of APP in human testis and sperm. To gain insights into the role of APP superfamily members in fertility, the study was extended to APP-like protein 2 (APLP2). We analyzed several topological properties of the APP interaction network and the biological and physiological properties of the proteins in the APP interaction network were also specified by gene ontologyand pathways analyses. We classified significant features related to the human male reproduction for the APP interacting proteins and identified modules of proteins with similar functional roles which may show cooperative behavior for male fertility.
Conclusions
The present work provides the first report on the APP interactome in human testis. Our approach allowed the identification of novel interactions and recognition of key APP interacting proteins for male reproduction, particularly in sperm-oocyte interaction.publishe
Results from the portuguese register
Objective Our aims were to evaluate the correlation between Juvenile Arthritis Disease Activity Score 27-joint reduced count (JADAS27) with erythrocyte sedimentation rate (ESR) and JADAS27 with C-reactive protein (CRP) scores and to test the agreement of both scores on classifying each disease activity state. We also aimed at verifying the correlation of the 2 scores across juvenile idiopathic arthritis (JIA) categories and to check the correlation between JADAS27-ESR and clinical JADAS27 (JADAS27 without ESR). Methods A nationwide cohort of patients with JIA registered in the Portuguese Register, Reuma.pt, was studied. JADAS27-CRP was adapted by replacing ESR with CRP level as the inflammatory marker. JADAS27-CRP was calculated similarly to JADAS27-ESR as the simple linear sum of its 4 components. Pearson's correlations and K statistics were used in the analyses. Results A total of 358 children had full data to calculate JADAS27; 65.4% were female and the mean ± SD disease duration was 11.8 ± 9.1 years. The correlation coefficient between JADAS27-ESR and JADAS27-CRP was 0.967 (P < 0.0001), although the correlation coefficient between ESR and CRP level was 0.335 (P < 0.0001). The strong correlation between JADAS27-ESR and JADAS27-CRP was maintained when compared within each JIA category. The agreement between JADAS27-ESR and JADAS27-CRP across the 4 activity states was very good, showing 91.1% of the observations in agreement; K = 0.867 (95% confidence interval 0.824-0.91). The correlation between JADAS27 with ESR and JADAS27 without ESR was high (r = 0.97, P < 0.0001). Conclusion JADAS27 based on CRP level correlated closely with JADAS27-ESR across all disease activity states and JIA categories, indicating that both measures can be used in clinical practice. Moreover, the correlation of JADAS27 with and without ESR was also high, suggesting that this tool might be useful even in the absence of laboratorial measures.publishersversionpublishe
Prevention of hypertension in patients with pre-hypertension: protocol for the PREVER-prevention trial
<p>Abstract</p> <p>Background</p> <p>Blood pressure (BP) within pre-hypertensive levels confers higher cardiovascular risk and is an intermediate stage for full hypertension, which develops in an annual rate of 7 out of 100 individuals with 40 to 50 years of age. Non-drug interventions to prevent hypertension have had low effectiveness. In individuals with previous cardiovascular disease or diabetes, the use of BP-lowering agents reduces the incidence of major cardiovascular events. In the absence of higher baseline risk, the use of BP agents reduces the incidence of hypertension. The PREVER-prevention trial aims to investigate the efficacy, safety and feasibility of a population-based intervention to prevent the incidence of hypertension and the development of target-organ damage.</p> <p>Methods</p> <p>This is a randomized, double-blind, placebo-controlled clinical trial, with participants aged 30 to 70 years, with pre-hypertension. The trial arms will be chlorthalidone 12.5 mg plus amiloride 2.5 mg or identical placebo. The primary outcomes will be the incidence of hypertension, adverse events and development or worsening of microalbuminuria and of left ventricular hypertrophy in the EKG. The secondary outcomes will be fatal or non-fatal cardiovascular events: myocardial infarction, stroke, heart failure, evidence of new sub-clinical atherosclerosis, and sudden death. The study will last 18 months. The sample size was calculated on the basis of an incidence of hypertension of 14% in the control group, a size effect of 40%, power of 85% and P alpha of 5%, resulting in 625 participants per group. The project was approved by the Ethics committee of each participating institution.</p> <p>Discussion</p> <p>The early use of blood pressure-lowering drugs, particularly diuretics, which act on the main mechanism of blood pressure rising with age, may prevent cardiovascular events and the incidence of hypertension in individuals with hypertension. If this intervention shows to be effective and safe in a population-based perspective, it could be the basis for an innovative public health program to prevent hypertension in Brazil.</p> <p>Trial Registration</p> <p>Clinical Trials <a href="http://www.clinicaltrials.gov/ct2/show/NCT00970931">NCT00970931</a>.</p
Processo alternativo para conservação do leite de coco produzido para consumo comercial
The present study is concerned with acidification of coconut (Cocos nucifera L.) milk with different acids (citric, malic, lactic, phosphoric and tartaric) to pH values between 4.2 and 4.4. The product was stored in 200 ml bottles and under thermal processing in boiling water for 30 minutes. A microbiological and physical-chemical evaluation of the product was carried out immediately after processing and every 30 days after that for a period of 150 days of storage. The acidification and the thermal processing used, proved to be efficient for the preservation of the product and no development of microorganisms or undesirable alterations were observed. All the acids tested proved to be efficient for the acidification of the coconut milk from the physical-chemical, chemical, physical and microbiological points of view.No presente estudo realizou-se a acidificação do leite de coco (Cocos nucifera L.) com distintos ácidos (cítrico, málico, láctico, fosfórico e tartárico) a valores de pH entre 4,2 e 4,4, tendo sido acondicionado em garrafas de 200 ml e submetido a tratamento térmico em água em ebulição por 30 min. Fez-se a avaliação microbiológica e físico-química do produto logo após o processamento e a cada 30 dias, durante um período de 150 dias de estocagem. Os processos de acidificação e pasteurização aplicados mostraram-se eficientes para a conservação do produto, não tendo sido verificado o desenvolvimento de microrganismos e alterações físico-químicas indesejáveis. Todos os ácidos testados mostraram-se eficientes para a acidificação do leite de coco sob o ponto de vista físico-químico, físico e microbiológico

Assessment of rubber tree panels under crowns resistant to South American leaf blight
The objective of this work was to assess the performance of panel clones under crowns resistant to South American leaf blight (Microcyclus ulei). The experiment was carried out with 18 panel clones crown-budded with Hevea pauciflora x H. guianensis, in a Xanthic Ferralsol (Oxisol) in Manaus, AM, Brazil. The following parameters were evaluated: dry rubber yield, plant nutritional status, and anatomical and physiological characteristics of the latex vessels. In the first three years of evaluation, the panel clones IAN 2878, IAN 2903, CNS AM 7905, CNS AM 7905 P1, and PB 28/59 showed the highest dry rubber yield potential, while the clones IAN 6158, IAN 6590, and IAN 6515 should not be recommended for crown budding. Higher potassium and copper foliar content in panel clones were associated to an increase in dry rubber yield. The simultaneous evaluation of anatomical and physiological characteristics of latex is fundamental for the selection of panel clones in the Amazon region. Crown budding is an efficient technology for South American leaf blight management in endemic regions
Anthropocene and "development" : intertwined trajectories since the beginning of the Great Acceleration = Antropoceno e "desenvolvimento" : trajetórias entrelaçadas desde o começo da Grande Aceleração
Objectives: We aim to propose the thesis that the trajectories of the Anthropocene and the current mainstream understandings of development are intertwined from the beginning. It means that the Anthropocene and the “development” are coetaneous: the implementation of development policies for the so-considered underdeveloped regions started to happen at the same time of what is known as The Great Acceleration of production, consumption and environmental degradation in a global level. Method: In this conceptual paper, we adopt a decolonial critique as an analytical lens and argue that different geopolitical positions may be necessary for approaching the issue of the Anthropocene from epistemological reflections that can include the cultural and political context of the production and reproduction of local knowledge. Results: Our theoretical argumentation sheds light on the role of Global North and South relations in shaping the environmental crisis. Latin America (LA) exemplifies the modus operandi of the intertwinement of the practical effects of development policies and the environmental consequences underlying the Anthropocene, in which natural resources are over-explored to satisfy export-oriented trade, from the South toward the North. LA is not only a propitious context to show the validity of our thesis, but also the source of alternatives to such developmental model. Conclusion: The emphasis on development as a cause of the Anthropocene supports The Great Acceleration thesis. The proposition of the name Developmentocene comes from the thesis that development and Anthropocene are coetaneous, the intertwinement of both resulting in the very definition of the new epoch.Objetivo: Buscamos propor a tese de que as trajetórias do Antropoceno e o entendimento atualmente dominante sobre desenvolvimento estão entrelaçadas desde o começo. Isso implica que o Antropoceno e o “desenvolvimento” são coetâneos: a implementação de políticas de desenvolvimento em regiões tidas como subdesenvolvidas começou a acontecer ao mesmo tempo em que teve início A Grande Aceleração da produção, do consumo e da degradação ambiental em nível global. Método: Neste artigo conceitual, nós adotamos a crítica decolonial como lente analítica e argumentamos a necessidade de diferentes posições geopolíticas para abordar a questão do Antropoceno a partir de reflexões epistemológicas que possam incluir o contexto cultural e político de produção e reprodução do conhecimento. Resultados: Nossa argumentação teórica enaltece as relações entre o Norte e o Sul Global no delineamento da crise ambiental. A América Latina (AL) exemplifica o modus operandi do entrelaçamento entre os efeitos práticos das políticas de desenvolvimento e as consequências ambientais subjacentes ao Antropoceno, em que os recursos naturais são explorados além dos limites para satisfazer o comércio para exportações, desde o Sul para o Norte. Nesse quadro, a AL não é apenas um contexto propício para mostrar a validade da nossa tese, mas também a fonte de alternativas a esse modelo de desenvolvimento. Conclusão: A ênfase no desenvolvimento como causa do Antropoceno apoia a tese da Grande Aceleração. A proposição do nome Desenvolvimentoceno advém da tese de que o desenvolvimento e o Antropoceno são coetâneos e que o entrelaçamento de ambos resulta na própria definição da nova época
Bone Morphogenetic Protein-6 (BMP-6) induces atresia in goat primordial follicles cultured in vitro
This study investigated the effects of bone morphogenetic protein 6 (BMP-6) on in vitro primordial follicle development in goats. Samples of goat ovarian cortex were cultured in vitro for 1 or 7 days in Minimum Essential Medium (control medium) supplemented with different concentrations of BMP-6. Follicle survival, activation and growth were evaluated through histology and transmission electron microscopy (TEM). After 7 days of culture, histological analysis demonstrated that BMP-6 enhanced the percentages of atretic primordial follicles when compared to fresh control (day 0). Nevertheless, BMP-6 increased follicular and oocyte diameter during both culture periods. As the culture period progressed from day 1 to day 7, a significant increase in follicle diameter was observed with 1 or 50ng/ml BMP-6. However, on the contrary to that observed with the control medium TEM revealed that follicles cultured for up to 7 days with 1 or 50ng/ml BMP-6 had evident signs of atresia. In conclusion, this study demonstrated that BMP-6 negatively affects the survival and ultrastructure of goat primordial follicles.O presente estudo investigou os efeitos da proteína morfogenética óssea-6 (BMP-6) no desenvolvimento in vitro de folículos primordiais caprinos. Amostras de córtex ovariano de cabras foram cultivados por 1 ou 7 dias em Meio Essencial Mínimo (meio controle) suplementado com diferentes concentrações de BMP-6. As taxas de sobrevivência, ativação e crescimento foram avaliadas por histologia clássica e microscopia eletrônica de transmissão (MET). Após 7 dias de cultivo, a análise histológica demonstrou que a BMP-6 aumentou o percentual de folículos primordiais degenerados no dia 7 quando comparados ao controle fresco (D0). Além disso, houve um aumento significativo do diâmetro folicular e oocitário em ambos os períodos de cultivo em todos os tratamentos na presença de BMP-6. Com a progressão do cultivo do dia 1 para o dia 7, nos tratamentos com 1 ou 50ng/ml de BMP-6, foi observado um aumento significativo no diâmetro folicular. Entretanto, contrário ao observado no meio controle, a MET revelou que os folículos cultivados nesses tratamentos apresentavam sinais evidentes de atresia. Em conclusão, esse estudo demonstrou que a BMP-6 afeta negativamente a sobrevivência e a ultra-estrutura de folículos primordiais caprinos
