    Adapting Global Service-Learning Project and Community Partnership Outcomes Using a “Tele-engineering” Approach in Response to the COVID-19 Pandemic

    The Water Supply in Developing Countries (WSDC) service-learning course at Purdue University has fostered a strong partnership with the La Vega region in the Dominican Republic since 2012. During this time, an interdisciplinary group of engineering and science students has helped design drinking water treatment systems and the group has developed water, sanitation, and health (WASH) education materials. These WASH education and water safety approaches often have been conducted in person in the past. However, with the state of the COVID-19 pandemic and the inability to travel in the fall and spring semesters of the 2020–2021 academic year, the students have been exploring (1) the impact of the pandemic on the community schools in the La Vega province, (2) the impact of the pandemic on the current water treatment systems, and (3) possible solutions to implement a “virtual installation” of a water treatment system at our newest partner school in the community of Desecho. The coronavirus pandemic has ushered in a new way that we may approach our service-learning experiences in the future. More specifically, in the future, it may be more effective to serve as a “virtual consulting firm” of engineers and consultants, rather than builders, for the implementation and design of the water treatment systems. In this way, we may facilitate a partnership that fosters community agency and solution-based approaches to technical issues are led by local community members

    Exploring pandemic-related health literacy among adolescents in Germany: a focus group study

    Background Health literacy enables people to cope efficiently with health threats, such as the COVID-19 pandemic. However, little is known about health literacy among adolescents in general and especially in the context of pandemics. This study aimed to explore pandemic-related health literacy among adolescents by addressing cognitive, behavioral, conative, and affective components of the multidimensional health literacy construct. Methods Four online focus groups with 24 adolescents aged 13-17 years from four German federal states were conducted during the COVID-19 pandemic in May and June 2021. Data were analyzed using qualitative content analysis. Results Regarding the cognitive and behavioral components of pandemic-related health literacy, adolescents reported to use a broad range of traditional and digital media and personal information sources. The adolescents considered pandemic-related information to be good and easy to understand, when the information is presented in a concise and structured manner. The participants stated difficulties in finding, understanding, and evaluating pandemic-related information regarding particular protective measures. The adolescents described themselves to be critical when evaluating pandemic-related information and reported a high level of adherence to protective measures. Regarding the conative and affective components of health literacy, the adolescents explained that their wish to protect their loved ones from getting infected was the predominant motive for adherence to protective measures. They were convinced that people of their age play a role in pandemic containment. The adolescents reported sometimes making exceptions from adhering to protective measures to cope with negative feelings they experienced during the pandemic. Conclusions This study provides insights on how measures to improve pandemic-related health literacy among adolescents may be tailored to their needs. Prompt, concise, structured, and comprehensible preparation and communication of pandemic-related information in addition to educational efforts to strengthen health-related cognitive skills and critical health literacy may be supportive to reduce barriers in finding, understanding, and evaluating pandemic-related content.Peer Reviewe

    Die Autor*innen widmen sich in diesem Beitrag der Frage, wie Hochschulen gesellschaftliche Verantwortung ĂŒbernehmen und Erkenntnisse aus ihren beiden Kernmissionen Forschung und Lehre vermehrt gesellschaftlich nutzbar machen können. Dieses akademische Aufgabenfeld, das darauf abzielt basierend auf wissenschaftlichen Erkenntnissen gesellschaftliche Entwicklungen mitzugestalten, wird als Third Mission bezeichnet. Im ersten Teil des Beitrags wird die HeterogenitĂ€t solcher Third-Mission-AktivitĂ€ten und die damit einhergehenden Schwierigkeiten eines einheitlichen inter- und intrauniversitĂ€ren BegriffsverstĂ€ndnisses beschrieben. Anschließend wird diskutiert, wie die Third Mission in bestehende universitĂ€re Strukturen eingebettet werden kann. Dabei wird besonders auf Förderungsmöglichkeiten von Third-Mission-AktivitĂ€ten eingegangen. Abschließend wird am Beispiel dreier UniversitĂ€ten illustriert, wie die oben genannten Herausforderungen systematisch bearbeitet werden können, um die Third Mission auf gesamtinstitutioneller Ebene zu implementieren. (Herausgeber)In this article, the authors address the question of how universities can assume social responsibility and make the findings from their two core missions of research and teaching more useful for society. This academic field of activity, which aims to help shape social developments on the basis of scientific findings, is referred to as the third mission. The first part of this article describes the heterogeneity of such third mission activities and the associated difficulties of a uniform inter- and intra-university understanding of the term. Subsequently, it is discussed how Third Mission can be embedded in existing university structures. Particular attention is paid to the possibilities of funding Third Mission activities. Finally, the example of three universities is used to illustrate how the above-mentioned challenges can be systematically addressed in order to implement the Third Mission at the overall institutional level.     Correlational analysis for identifying genes whose regulation contributes to chronic neuropathic pain

    <p>Abstract</p> <p>Background</p> <p>Nerve injury-triggered hyperexcitability in primary sensory neurons is considered a major source of chronic neuropathic pain. The hyperexcitability, in turn, is thought to be related to transcriptional switching in afferent cell somata. Analysis using expression microarrays has revealed that many genes are regulated in the dorsal root ganglion (DRG) following axotomy. But which contribute to pain phenotype versus other nerve injury-evoked processes such as nerve regeneration? Using the L5 spinal nerve ligation model of neuropathy we examined <b><it>differential </it></b>changes in gene expression in the L5 (and L4) DRGs in five mouse strains with contrasting susceptibility to neuropathic pain. We sought genes for which the degree of regulation correlates with strain-specific pain phenotype.</p> <p>Results</p> <p>In an initial experiment six candidate genes previously identified as important in pain physiology were selected for in situ hybridization to DRG sections. Among these, regulation of the Na<sup>+ </sup>channel α subunit <it>Scn11a </it>correlated with levels of spontaneous pain behavior, and regulation of the cool receptor <it>Trpm8 </it>correlated with heat hypersensibility. In a larger scale experiment, mRNA extracted from individual mouse DRGs was processed on Affymetrix whole-genome expression microarrays. Overall, 2552 ± 477 transcripts were significantly regulated in the axotomized L5DRG 3 days postoperatively. However, in only a small fraction of these was the degree of regulation correlated with pain behavior across strains. Very few genes in the "uninjured" L4DRG showed altered expression (24 ± 28).</p> <p>Conclusion</p> <p>Correlational analysis based on in situ hybridization provided evidence that differential regulation of <it>Scn11a </it>and <it>Trpm8 </it>contributes to across-strain variability in pain phenotype. This does not, of course, constitute evidence that the others are unrelated to pain. Correlational analysis based on microarray data yielded a larger "look-up table" of genes whose regulation likely contributes to pain variability. While this list is enriched in genes of potential importance for pain physiology, and is relatively free of the bias inherent in the candidate gene approach, additional steps are required to clarify which transcripts on the list are in fact of functional importance.</p

    Power estimation for non-standardized multisite studies

    AbstractA concern for researchers planning multisite studies is that scanner and T1-weighted sequence-related biases on regional volumes could overshadow true effects, especially for studies with a heterogeneous set of scanners and sequences. Current approaches attempt to harmonize data by standardizing hardware, pulse sequences, and protocols, or by calibrating across sites using phantom-based corrections to ensure the same raw image intensities. We propose to avoid harmonization and phantom-based correction entirely. We hypothesized that the bias of estimated regional volumes is scaled between sites due to the contrast and gradient distortion differences between scanners and sequences. Given this assumption, we provide a new statistical framework and derive a power equation to define inclusion criteria for a set of sites based on the variability of their scaling factors. We estimated the scaling factors of 20 scanners with heterogeneous hardware and sequence parameters by scanning a single set of 12 subjects at sites across the United States and Europe. Regional volumes and their scaling factors were estimated for each site using Freesurfer's segmentation algorithm and ordinary least squares, respectively. The scaling factors were validated by comparing the theoretical and simulated power curves, performing a leave-one-out calibration of regional volumes, and evaluating the absolute agreement of all regional volumes between sites before and after calibration. Using our derived power equation, we were able to define the conditions under which harmonization is not necessary to achieve 80% power. This approach can inform choice of processing pipelines and outcome metrics for multisite studies based on scaling factor variability across sites, enabling collaboration between clinical and research institutions

    A Dynamic Landscape for Antibody Binding Modulates Antibody-Mediated Neutralization of West Nile Virus

    Neutralizing antibodies are a significant component of the host's protective response against flavivirus infection. Neutralization of flaviviruses occurs when individual virions are engaged by antibodies with a stoichiometry that exceeds a required threshold. From this “multiple-hit” perspective, the neutralizing activity of antibodies is governed by the affinity with which it binds its epitope and the number of times this determinant is displayed on the surface of the virion. In this study, we investigated time-dependent changes in the fate of West Nile virus (WNV) decorated with antibody in solution. Experiments with the well-characterized neutralizing monoclonal antibody (MAb) E16 revealed a significant increase in neutralization activity over time that could not be explained by the kinetics of antibody binding, virion aggregation, or the action of complement. Additional kinetic experiments using the fusion-loop specific MAb E53, which has limited neutralizing activity because it recognizes a relatively inaccessible epitope on mature virions, identified a role of virus “breathing” in regulating neutralization activity. Remarkably, MAb E53 neutralized mature WNV in a time- and temperature-dependent manner. This phenomenon was confirmed in studies with a large panel of MAbs specific for epitopes in each domain of the WNV envelope protein, with sera from recipients of a live attenuated WNV vaccine, and in experiments with dengue virus. Given enough time, significant inhibition of infection was observed even for antibodies with very limited, or no neutralizing activity in standard neutralization assays. Together, our data suggests that the structural dynamics of flaviviruses impacts antibody-mediated neutralization via exposure of otherwise inaccessible epitopes, allowing for antibodies to dock on the virion with a stoichiometry sufficient for neutralization
