Mitochondrial dysfunction in adult midbrain dopamine neurons triggers an early immune response

    Get PDF
Dopamine (DA) neurons of the midbrain are at risk to become affected by mitochondrial damage over time and mitochondrial defects have been frequently reported in Parkinson\u27s disease (PD) patients. However, the causal contribution of adult-onset mitochondrial dysfunction to PD remains uncertain. Here, we developed a mouse model lacking Mitofusin 2 (MFN2), a key regulator of mitochondrial network homeostasis, in adult midbrain DA neurons. The knockout mice develop severe and progressive DA neuron-specific mitochondrial dysfunction resulting in neurodegeneration and parkinsonism. To gain further insights into pathophysiological events, we performed transcriptomic analyses of isolated DA neurons and found that mitochondrial dysfunction triggers an early onset immune response, which precedes mitochondrial swelling, mtDNA depletion, respiratory chain deficiency and cell death. Our experiments show that the immune response is an early pathological event when mitochondrial dysfunction is induced in adult midbrain DA neurons and that neuronal death may be promoted non-cell autonomously by the cross-talk and activation of surrounding glial cells

Surface topography on trimming dies and the influence of wear

    No full text
In this paper it is investigated how surface topography on trimming dies influences the wear process. Surfaces on both semi-industrial test dies and dies used in production were analyzed with different surface topography analysis methods. The result shows that the wear process is different on the different sides of the trimming edge. The texture direction of the surface is changed due to wear, with the strongest change on the clearance side of the edge. The roughness on sheet side of the edge is reduced due to a hammering effect from the sheet material. The result on the test dies and the production dies correlates well

Measuring strategies for smooth tool steel surfaces

Comparisons between different measuring strategies were made on three types of smooth tool steel surfaces. Three replica materials were tested to study possibilities within replication techniques. An optical interferometer as well as a mechanical stylus was used to evaluate the surfaces. The results showed that the tested replica materials generated good representations of both the form and the surface roughness (Sq > 300 nm). The evaluated surfaces were quite homogeneous, thus, few measurements are needed to get representative results. However, it was found that caution must be taken regarding manually polished surfaces which can be less homogenous and therefore require more measurements to get representative results

Formal and informal communication in a project organisation: The case of a large construction project

    No full text
In larger projects, where large numbers of project participants are involved, it is important to have a well functioning project organisation. The more complex a project is the more planning and control is needed. The study presented in this paper investigates an installation company’s formal and informal communication when constructing a new power plant for district heating, in G\uf6teborg, Sweden. This is done in order to understand the informal communication and its effect on the project organisation. Literature reviews and interviews with managers involved in the project have been done. The results of the study indicate that the actual organisational structure differs from the planned structure in terms of communication paths. Actors involved, tend to contact each other beside the formal chain of order. A well functioning project team with established informal communication paths might be a possibility to achieve success in projects

Architecting cars as constituents of a system of systems

    No full text
Future transportation systems will be a heterogeneous mix of items with varying connectivity and interoperability. A mix of new technologies and legacy systems will co-exist to realize a variety of scenarios involving not only connected cars but also road infrastructures, pedestrians, cyclists, etc. Future transportation systems can be seen as a System of Systems (SoS), where each constituent system - one of the units that compose an SoS - can act as a standalone system, but the cooperation among the constituent systems enables new emerging and promising scenarios. In this paper we investigate how to architect cars so that they can be constituents of future transportation systems. This work is realized in the context of two Swedish projects coordinated by Volvo Cars and involving some universities and research centers in Sweden and many suppliers of the OEM, including Autoliv, Arccore, Combitech, Cybercom, Knowit, Prevas, \uc5F-Technology, Semcom, and Qamcom
