Modulation of HU-DNA interactions by salt concentration and applied force.
HU is one of the most abundant proteins in bacterial chromosomes and participates in nucleoid compaction and gene regulation. We report experiments using DNA stretching that study the dependence of DNA condensation by HU on force, salt and HU concentration. Previous experiments at sub-physiological salt levels revealed that low concentrations of HU could compact DNA, whereas larger HU concentrations formed a DNA-stiffening complex. Here we report that this bimodal binding behavior depends sensitively on salt concentration. Only the compaction mode was observed for 150 mM and higher NaCl levels, i.e. for physiological salt concentrations. Similar results were obtained for the more physiological salt K-glutamate. Real-time studies of dissociation kinetics revealed that HU unbound slowly (minutes to hours under the conditions studied) but completely for salt concentrations at or above 100 mM NaCl; the lifetime of HU complexes was observed to increase with the HU concentration at which the complexes were formed, and to decrease with salt concentration. Higher salt levels of 300 mM NaCl completely eliminated observable HU binding to DNA. Finally, we observed that the dissociation kinetics depend on force applied to the DNA: increased applied force in the sub-piconewton range accelerates dissociation, suggesting a mechanism for DNA tension to regulate chromosome structure and gene expression
Contingent Valuation of Sports Stadiums and Arenas: Temporal Embedding and Order Effect
Using the Contingent Valuation Method, this paper estimates the value of public goods the National Football Leagueâs Jaguars produce for Jacksonville, Florida, including the value of elevating Jacksonville to âmajor leagueâ status and the value of improving racial relations. It also estimates the incremental value of public goods potentially produced by a National Basketball Association team in Jacksonville. The present value of public goods created by the Jaguars is 22.8 million. The results add to the growing body of CVM literature indicating that sports public goods probably cannot justify the large public expenditures on stadiums and arenas.
The History of the Mysterious Eclipses of KH 15D: Asiago Observatory, 1967-1982
We are gathering archival observations to determine the photometric history
of the unique and unexplained eclipses of the pre-main-sequence star KH 15D.
Here we present a light curve from 1967-1982, based on photographic plates from
Asiago Observatory. During this time, the system alternated periodically
between bright and faint states, as observed today. However, the bright state
was 0.9 mag brighter than the modern value, and the fractional variation
between bright and faint states (Delta I = 0.7 mag) was smaller than observed
today (3.5 mag). A possible explanation for these findings is that the system
contains a second star that was previously blended with the eclipsing star, but

Extracting Radial Velocities of A- and B-type Stars from Echelle Spectrograph Calibration Spectra
We present a technique to extract radial velocity measurements from echelle
spectrograph observations of rapidly rotating stars ( km
s). This type of measurement is difficult because the line widths of
such stars are often comparable to the width of a single echelle order. To
compensate for the scarcity of lines and Doppler information content, we have
developed a process that forward-models the observations, fitting the radial
velocity shift of the star for all echelle orders simultaneously with the
echelle blaze function. We use our technique to extract radial velocity
measurements from a sample of rapidly rotating A- and B-type stars used as
calibrator stars observed by the California Planet Survey observations. We
measure absolute radial velocities with a precision ranging from 0.5-2.0 km
s per epoch for more than 100 A- and B-type stars. In our sample of 10
well-sampled stars with radial velocity scatter in excess of their measurement
uncertainties, three of these are single-lined binaries with long observational
baselines. From this subsample, we present detections of two previously unknown
spectroscopic binaries and one known astrometric system. Our technique will be
useful in measuring or placing upper limits on the masses of sub-stellar
companions discovered by wide-field transit surveys, and conducting future
spectroscopic binarity surveys and Galactic space-motion studies of massive
and/or young, rapidly-rotating stars.Comment: Accepted to ApJ

Herschel Observations of Debris Discs Orbiting Planet-hosting Subgiants
Debris discs are commonly detected orbiting main-sequence stars, yet little
is known regarding their fate as the star evolves to become a giant. Recent
observations of radial velocity detected planets orbiting giant stars highlight
this population and its importance for probing, for example, the population of
planetary systems orbiting intermediate mass stars. Our Herschel survey
observed a subset of the Johnson et al program subgiants, finding that 4/36
exhibit excess emission thought to indicate debris, of which 3/19 are
planet-hosting stars and 1/17 are stars with no current planet detections.
Given the small numbers involved, there is no evidence that the disc detection
rate around stars with planets is different to that around stars without
planets. Our detections provide a clear indication that large quantities of
dusty material can survive the stars' main-sequence lifetime and be detected on
the subgiant branch, with important implications for the evolution of planetary
systems and observations of polluted or dusty white dwarfs. Our detection rates
also provide an important constraint that can be included in models of debris

Controlled rotation mechanism of DNA strand exchange by the Hin serine recombinase.
DNA strand exchange by serine recombinases has been proposed to occur by a large-scale rotation of halves of the recombinase tetramer. Here we provide the first direct physical evidence for the subunit rotation mechanism for the Hin serine invertase. Single-DNA looping assays using an activated mutant (Hin-H107Y) reveal specific synapses between two hix sites. Two-DNA "braiding" experiments, where separate DNA molecules carrying a single hix are interwound, show that Hin-H107Y cleaves both hix sites and mediates multi-step rotational relaxation of the interwinding. The variable numbers of rotations in the DNA braid experiments are in accord with data from bulk experiments that follow DNA topological changes accompanying recombination by the hyperactive enzyme. The relatively slow Hin rotation rates, combined with pauses, indicate considerable rotary friction between synapsed subunit pairs. A rotational pausing mechanism intrinsic to serine recombinases is likely to be crucial for DNA ligation and for preventing deleterious DNA rearrangements
Multiple interfaces between a serine recombinase and an enhancer control site-specific DNA inversion.
Serine recombinases are often tightly controlled by elaborate, topologically-defined, nucleoprotein complexes. Hin is a member of the DNA invertase subclass of serine recombinases that are regulated by a remote recombinational enhancer element containing two binding sites for the protein Fis. Two Hin dimers bound to specific recombination sites associate with the Fis-bound enhancer by DNA looping where they are remodeled into a synaptic tetramer competent for DNA chemistry and exchange. Here we show that the flexible beta-hairpin arms of the Fis dimers contact the DNA binding domain of one subunit of each Hin dimer. These contacts sandwich the Hin dimers to promote remodeling into the tetramer. A basic region on the Hin catalytic domain then contacts enhancer DNA to complete assembly of the active Hin tetramer. Our results reveal how the enhancer generates the recombination complex that specifies DNA inversion and regulates DNA exchange by the subunit rotation mechanism.
