    Mach number effects on transonic aeroelastic forces and flutter characteristics

    Transonic aeroelastic stability analysis and flutter calculations are presented for a generic transport-type wing based on the use of the CAP-TSD (Computational Aeroelasticity Program - Transonic Small Disturbance) finite-difference code. The CAP-TSD code was recently developed for transonic unsteady aerodynamic and aeroelastic analysis of complete aircraft configurations. A binary aeroelastic system consisting of simple bending and torsion modes was used to study aeroelastic behavior at transonic speeds. Generalized aerodynamic forces are presented for a wide range of Mach number and reduced frequency. Aeroelastic characteristics are presented for variations in freestream Mach number, mass ratio, and bending-torsion frequency ratio. Flutter boundaries are presented which have two transonic dips in flutter speed. The first dip is the usual transonic dip involving a bending-dominated flutter mode. The second dip is characterized by a single degree-of-freedom torsion oscillation. These aeroelastic results are physically interpreted and shown to be related to the steady state shock location and changes in generalized aerodynamic forces due to freestream Mach number

    The Marine Nature of Nuwuk Lake and Small Ponds of the Peninsula of Point Barrow Alaska

    Discusses a score or more ponds, some transient, some persistent, on this narrow gravel spit, their location, nature, salinity and temperature; their biotas, marine and fresh-water, are outlined. Nuwuk Lake, the largest water body of the locality, approx. 600 ft long, max. depth 18.5 ft, is treated in some detail: its bottom, its formation by converging currents of the Bering and Chukchi Seas, ice conditions, temperature, salinity and O2-content. The biotas: euryhaline, reduced shallow-sea fauna are dealt with and the organisms collected during 1952-1960 are tabulated. Comparison is made with the few halocline lakes known in northern Russia and Scandinavia, notably Mogil'noye on Kil'din Island

    Development of a new real-time PCR for the detection of pilchard orthomyxovirus (POMV) in apparently healthy fish

    Pilchard orthomyxovirus (POMV) is a virus of concern to the Atlantic salmon aquaculture industry in Tasmania. First isolated from wild pilchards in southern Australia in 1998, the virus is now a recognised pathogen of farmed Atlantic salmon (Salmo salar) in Tasmania. While the current real-time PCR for POMV targets segment 5 of the viral genome, recent viral gene expression data suggests that other segments of the POMV genome presented higher transcription levels and thus may be better candidates for the early detection of the virus. This study aimed to design and begin validating a more sensitive reverse transcriptase real-time PCR (RT-qPCR) assay to detect POMV. Primers and probes were developed targeting two independent viral genes derived from segments 7 and 8, which presented higher transcription levels than segment 5 in both cell culture and infected fish. These were compared with the current POMV RT-qPCR. The POMV segment 8 assay had a higher analytical sensitivity than segment 7, detecting at least 1 plasmid copy μl−1, and was 10-fold more sensitive than both POMV segment 7 and 5 assays when analysing nucleic acid from a positive field sample. Both new assays also had high analytical specificity, detecting the 11 POMV isolates tested (inclusivity testing) and not amplifying nucleic acids from other viruses, including ISAV, a related orthomyxovirus. In the latent class model analysis, the diagnostic sensitivity of the segment 8 and 7 assays were higher than segment 5 in 93% and 92% of simulations, respectively. Seven samples (18.4%), all from subclinical fish infected with POMV, returned a positive result only with the segment 8 assay. Both new assays showed reproducible results when applied to aliquots of the same samples tested in three different laboratories. The new POMV segment 8 assay shows promising results as a surveillance tool for detecting POMV in fish without any symptoms.publishedVersio

    Use of Genetic Stock Identification Data for Comparison of the Ocean Spatial Distribution, Size at Age, and Fishery Exposure of an Untagged Stock and Its Indicator: California Coastal versus Klamath River Chinook Salmon

    Managing weak stocks in mixed-stock fisheries often relies on proxies derived from data-rich indicator stocks. For example, full cohort reconstruction of tagged Klamath River fall run Chinook salmon (Oncorhynchus tshawytscha) of northern California, USA, enables the use of detailed models to inform management. Information gained from this stock is also used in the management of the untagged, threatened California Coastal Chinook (CCC) salmon stock, by capping Klamath harvest rates. To evaluate use of this proxy, we used genetic stock identification (GSI) data to compare the two stocks\u27 size-at-age and ocean distribution, two key factors influencing fishery exposure. We developed methods to account for both sampling and genetic assignment uncertainty in catch estimates. We found that, in 2010, the stocks were similar in size-at-age early in the year (age-3 and age-4), but CCC fish were larger later in the year. The stocks appeared similarly distributed early in the year (2010), but more concentrated near their respective source rivers later in the year (2010 and 2011). If these results are representative, relative fishery impacts on the two stocks might scale similarly early in the year but management changes later in the year might have differing impacts on the two stocks

    The total synthesis of (-)-cyanthiwigin F by means of double catalytic enantioselective alkylation

    Double catalytic enantioselective transformations are powerful synthetic methods that can facilitate the construction of stereochemically complex molecules in a single operation. In addition to generating two or more stereocentres in a single reaction, multiple asymmetric reactions also impart increased enantiomeric excess to the final product in comparison with the analogous single transformation. Furthermore, multiple asymmetric operations have the potential to independently construct several stereocentres at remote points within the same molecular scaffold, rather than relying on pre-existing chiral centres that are proximal to the reactive site. Despite the inherent benefits of multiple catalytic enantioselective reactions, their application to natural product total synthesis remains largely underutilized. Here we report the use of a double stereoablative enantioselective alkylation reaction in a concise synthesis of the marine diterpenoid (-)-cyanthiwigin F (ref. 8). By employing a technique for independent, selective formation of two stereocentres in a single stereoconvergent operation, we demonstrate that a complicated mixture of racemic and meso diastereomers may be smoothly converted to a synthetically useful intermediate with exceptional enantiomeric excess. The stereochemical information generated by means of this catalytic transformation facilitates the easy and rapid completion of the total synthesis of this marine natural product

    A One Medicine Mission for an Effective Rabies Therapy

    Despite the disease's long history, little progress has been made toward a treatment for rabies. The prognosis for patient recovery remains dire. For any prospect of survival, patients require aggressive critical care, which physicians in rabies endemic areas may be reluctant or unable to provide given the cost, clinical expertise required, and uncertain outcome. Systematic clinical research into combination therapies is further hampered by sporadic occurrence of cases. In this Perspective, we examine the case for a One Medicine approach to accelerate development of an effective therapy for rabies through the veterinary care and investigational treatment of naturally infected dogs in appropriate circumstances. We review the pathogenesis of rabies virus in humans and dogs, including recent advances in our understanding of the molecular basis for the severe neurological dysfunction. We propose that four categories of disease process need to be managed in patients: viral propagation, neuronal degeneration, inflammation and systemic compromise. Compassionate critical care and investigational treatment of naturally infected dogs receiving supportive therapy that mimics the human clinical scenario could increase opportunities to study combination therapies that address these processes, and to identify biomarkers for prognosis and therapeutic response. We discuss the safety and ethics of this approach, and introduce the Canine Rabies Treatment Initiative, a non-profit organization with the mission to apply a One Medicine approach to the investigation of diagnostic, prognostic, and therapeutic options for rabies in naturally infected dogs, to accelerate transformation of rabies into a treatable disease for all patients

    Science Impacts of the SPHEREx All-Sky Optical to Near-Infrared Spectral Survey: Report of a Community Workshop Examining Extragalactic, Galactic, Stellar and Planetary Science

    SPHEREx is a proposed SMEX mission selected for Phase A. SPHEREx will carry out the first all-sky spectral survey and provide for every 6.2" pixel a spectra between 0.75 and 4.18 μ\mum [with R∼\sim41.4] and 4.18 and 5.00 μ\mum [with R∼\sim135]. The SPHEREx team has proposed three specific science investigations to be carried out with this unique data set: cosmic inflation, interstellar and circumstellar ices, and the extra-galactic background light. It is readily apparent, however, that many other questions in astrophysics and planetary sciences could be addressed with the SPHEREx data. The SPHEREx team convened a community workshop in February 2016, with the intent of enlisting the aid of a larger group of scientists in defining these questions. This paper summarizes the rich and varied menu of investigations that was laid out. It includes studies of the composition of main belt and Trojan/Greek asteroids; mapping the zodiacal light with unprecedented spatial and spectral resolution; identifying and studying very low-metallicity stars; improving stellar parameters in order to better characterize transiting exoplanets; studying aliphatic and aromatic carbon-bearing molecules in the interstellar medium; mapping star formation rates in nearby galaxies; determining the redshift of clusters of galaxies; identifying high redshift quasars over the full sky; and providing a NIR spectrum for most eROSITA X-ray sources. All of these investigations, and others not listed here, can be carried out with the nominal all-sky spectra to be produced by SPHEREx. In addition, the workshop defined enhanced data products and user tools which would facilitate some of these scientific studies. Finally, the workshop noted the high degrees of synergy between SPHEREx and a number of other current or forthcoming programs, including JWST, WFIRST, Euclid, GAIA, K2/Kepler, TESS, eROSITA and LSST.Comment: Report of the First SPHEREx Community Workshop, http://spherex.caltech.edu/Workshop.html , 84 pages, 28 figure
