    Snakes and ladders: localized solutions of plane Couette flow

    We demonstrate the existence of a large number of exact solutions of plane Couette flow, which share the topology of known periodic solutions but are localized in space. Solutions of different size are organized in a snakes-and-ladders structure strikingly similar to that observed for simpler pattern-forming PDE systems. These new solutions are a step towards extending the dynamical systems view of transitional turbulence to spatially extended flows.Comment: submitted to Physics Review Letter

    The effects of different parameterizations of Markov-switching in a CIR model of bond pricing

    We examine several discrete-time versions of the Cox, Ingersoll and Ross (CIR) model for the term structure, in which the short rate is subject to discrete shifts. Our empirical analysis suggests that careful consideration of which parameters of the short-term interest rate equation that are allowed to be switched is crucial. Ignoring this issue may result in a parameterization that produces no improvement (in terms of bond pricing) relative to the standard CIR model, even when there are clear breaks in the data

    Approaches to overcome flow cytometry limitations in the analysis of cells from veterinary relevant species

    BACKGROUND: Flow cytometry is a powerful tool for the multiparameter analysis of leukocyte subsets on the single cell level. Recent advances have greatly increased the number of fluorochrome-labeled antibodies in flow cytometry. In particular, an increase in available fluorochromes with distinct excitation and emission spectra combined with novel multicolor flow cytometers with several lasers have enhanced the generation of multidimensional expression data for leukocytes and other cell types. However, these advances have mainly benefited the analysis of human or mouse cell samples given the lack of reagents for most animal species. The flow cytometric analysis of important veterinary, agricultural, wildlife, and other animal species is still hampered by several technical limitations, even though animal species other than the mouse can serve as more accurate models of specific human physiology and diseases. RESULTS: Here we present time-tested approaches that our laboratory regularly uses in the multiparameter flow cytometric analysis of ovine leukocytes. The discussed approaches will be applicable to the analysis of cells from most animal species and include direct modification of antibodies by covalent conjugation or Fc-directed labeling (Zenon™ technology), labeled secondary antibodies and other second step reagents, labeled receptor ligands, and antibodies with species cross-reactivity. CONCLUSIONS: Using refined technical approaches, the number of parameters analyzed by flow cytometry per cell sample can be greatly increased, enabling multidimensional analysis of rare samples and giving critical insight into veterinary and other less commonly analyzed species. By maximizing information from each cell sample, multicolor flow cytometry can reduce the required number of animals used in a study

    Epigenetic silencing of SOCS3 expression contributes to fibrosis in Crohn’s disease

    Identified risk polymorphisms affecting the Jak-STAT3 pathway in patients with Crohn’s disease could affect TGF-β1 and collagen I expression and in the pathway’s negative regulator, SOCS3. Genetic factors, however, account for only ~25% of disease. Epigenetic events also shape gene expression. Recent experiments showed that autocrine IL-6 production in mesenchymal cells, subepithelial myofibroblasts (SEMF) and muscle cells, of patients with fibrostenotic Crohn’s disease causes sustained Jak-STAT3 activity, excess TGF-β1 and Collagen I production and fibrosis. SOCS3 paradoxically decreased in these cells. We now identify epigenetic mechanisms that silence SOCS3 expression in SEMF of patients with fibrostenotic Crohn’s disease. In a previous experiment, using isolated SEMF of normal ileum and affected ileum from patients with each Crohn’s phenotype, inflammatory (Montreal B1), fibrostenotic (B2) and penetrating (B3), we confirmed decreased SOCS3 protein levels were unique to B2 patients. Expression of miR-19b increased in SEMF of affected ileum. SOCS3 transcriptional activity decreased after transfection of miR-19b mimic and increased when antagomiR-19b was expressed. Epigenetic silencing of SOCS3 in ileal SEMF of patients with fibrostenotic Crohn’s disease occurs by increased miR-19b mediated inhibition of SOCS3

    On the Direct Detection of Dark Matter Annihilation

    We investigate the direct detection phenomenology of a class of dark matter (DM) models in which DM does not directly interact with nuclei, {but rather} the products of its annihilation do. When these annihilation products are very light compared to the DM mass, the scattering in direct detection experiments is controlled by relativistic kinematics. This results in a distinctive recoil spectrum, a non-standard and or even absent annual modulation, and the ability to probe DM masses as low as a \sim10 MeV. We use current LUX data to show that experimental sensitivity to thermal relic annihilation cross sections has already been reached in a class of models. Moreover, the compatibility of dark matter direct detection experiments can be compared directly in EminE_{{\rm min}} space without making assumptions about DM astrophysics, mass, or scattering form factors. Lastly, when DM has direct couplings to nuclei, the limit from annihilation to relativistic particles in the Sun can be stronger than that of conventional non-relativistic direct detection by more than three orders of magnitude for masses in a 2-7 GeV window.Comment: 4 pages, 3 figures, PRL versio

    Halo Independent Direct Detection of Momentum-Dependent Dark Matter

    We show that the momentum dependence of dark matter interactions with nuclei can be probed in direct detection experiments without knowledge of the dark matter velocity distribution. This is one of the few properties of DM microphysics that can be determined with direct detection alone, given a signal of dark matter in multiple direct detection experiments with different targets. Long-range interactions arising from the exchange of a light mediator are one example of momentum-dependent DM. For data produced from the exchange of a massless mediator we find for example that the mediator mass can be constrained to be 10\lesssim 10 MeV for DM in the 20-1000 GeV range in a halo-independent manner.Comment: 15 pages, 4 figures; updated to match published versio

    A promising biotechnical approach to pest management of the western corn rootworm in Illinois maize fields shielded with a MCA kairomone baited trap line

    The leaf beetle Diabrotica virgifera virgifera (Coleoptera: Chrysomelidae), (D.v.v.), also called the western corn rootworm, is endemic to the New World and ranks among the top ten insect pests in worldwide grain production. D.v.v. causes annual damages of 1 billion US Dollars and is a notoriously difficult insect pest to control and manage, as entomological history of the past 50 years amply demonstrates (METCALF 1986). Considering recent emphasis on environmentally compatible and sustainable management strategies, entomologists and practitioners are encouraged to pay increased attention to novel approaches such as biotechnial methods which today are characterized by preferential use of signal compounds. Fortunately, both insect and plants provide a wide variety of such natural resources. In the case of D.v.v., sex pheromonesand plant kairomones as specific attractants and management tools are relatively well investigated through numerous contributions by GUSS et al. (1982), METCALF & METCALF (1992), METCALF (1994) and many recent publications on the advance and spread of D.v.v. within Europe (BERGER 1995-2004, HUMMEL 2003). Principle of MSD method: In this paper, the plant kairomone 4-methoxycinnamaldehyde (MCA), a specific attractant for D.v.v., is being used as a tool within the newly proposed "MSD" strategy. It combines a two pronge approach consisting as the well known mass trapping with the novel shielding and deflecting, called in short "diversion" and introduced here for the first time. An invisible “curtain” or “fence” of MCA vapor released from a MCA trap line establishes a behavioral barrier which the flying beetles cannot easily pass without being 1. either caught in one of the high capacity traps or 2. being diverted elsewhere. The net effect is a significant reduction in adult population density and oviposition within the MCA treated field as compared to an untreated control field. These effects can be experimentally measured by 1. adult beetle counts on maize plants, 2. by counts in independent monitoring traps baited with the D.v.v. sex pheromone, and 3. by egg counts taken in soil samples.Umweltgerechter und nachhaltiger Pflanzenschutz erfordert eine Vielzahl verschiedener Strategien zum Management von Schadinsekten auf ihren Wirtspflanzen. Insekten- und pflanzeneigene Signalstoffe sowie ihre synthetischen Analoga bieten einen nahezu unerschöpflichen Vorrat spezifischer Lockwirkungen und Interventionsmöglichkeiten an, der bisher leider nur unzureichend genutzt wird. Kürzliche Entdeckungen sowohl neuer Kairomon-Lockstoffe als auch neuer Verfahrensschritte für das Management des Westlichen Maiswurzelbohrers Diabrotica virgifera virgifera LeConte (Coleoptera: Chrysomelidae) (D.v.v.) erweitern das Spektrum anwendbarer Management- Optionen für diesen schwer bekämpfbaren Schädling im nordamerikanischen Maisanbau. Seit seiner Einschleppung nach Europa (ČAMPRAG & BAČA 1995) wird D.v.v. neuerdings auch zunehmend zum Problemschädling an Mais in Europa. Die neue "MSD"-Technik besteht aus einer Kombination von Massenabfang ("mass trapping"), Abschirmung ("shielding") und Umlenkung ("diversion") der Blattkäfer. Fallen hoher Fangkapazität, die das Kairomon 4-Methoxyzimtaldehyd (MCA) und Cucurbitacin- Pulver als fraßfördernden Stoff enthalten, können bei Aufstellung als "Fallenzaun" relativ geringer Dichte die Käfer so umlenken, dass zwischen den beiden Seiten dieses Zauns eine unsichtbare Geruchs-Barriere entsteht. Diese führt zu einer messbaren und signifikanten Verminderung der Käferzahl zwischen MCA behandelten Feldabschnitten gegenüber ihren unbehandelten Kontrollen. In 0,27 ha bzw. 0,12 ha großen Maisversuchsfeldern der Standorte Urbana und Champaign des US-Staates Illinois ließen sich während der Monate August und September 2003 und 2004 nach MCA-Behandlung gegenüber Kontrollen deutliche und signifikante Verminderungen von D.v.v. an Hand dreier Kriterien nachweisen: 1. Käferzahlen auf Maispflanzen innerhalb des "MSD-Feldes", 2. Käferzahlen in Sexuallockstoff-Fallen im MSD Feld und 3. die Zahl abgelegter D.v.v.-Eier im Boden des MSD Feldes nehmen ab. Der beobachtete Effekt läßt sich nicht allein auf die Populationsverminderung infolge hoher Abfangzahlen zurückführen. Es gibt darüber hinaus einen Abschirm- und Umlenkeffekt, dessen sinnes- und verhaltensphysiologische Mechanismen zusätzliche künftige Erforschungen erforderlich machen

    Recognition of micro-scale deformation structures in glacial sediments - pattern perception, observer bias and the influence of experience

    It is a scientist's mission to try to remain unbiased. However, certain factors play a role in scientific analyses that are not controlled by conscious thought. These factors are potentially very important in areas of science where interpretations are based on a scientist's ability to identify patterns or structures. One such area is the micromorphology of glacial sediments. In this paper we investigate the role of an analyst's experience in relation to pattern perception with specific reference to turbate microstructures in glacial diamictons. An experiment was conducted on 52 participants, which demonstrated that, as may be expected, more experienced (glacial) micromorphologists tend to exhibit a higher sensitivity-to-signal, but that complete novices, if given clear instructions, can reach levels of sensitivity similar to those of experts. It also showed, perhaps more surprisingly, that response bias does not decrease with experience. We discuss psychological factors, such as the drive for success and consistency, that may have contributed to these results and investigate their possible implications in the micromorphological analysis and interpretation of glacial sediments

    A comparative study of herbage intake, ingestive behaviour and diet selection, and effects of condensed tannins upon body and wool growth in lambs grazing Yorkshire fog (Holcus lanatus) and annual ryegrass (Lolium multiflorum) dominant swards

    An experiment was carried out from August to early November 1994 to examine differences in diet selection, herbage intake, grazing behaviour and animal performance between weaned lambs rotationally grazing swards of annual ryegrass (Lolium multiflorum)/white clover (Trifolium repens) and Yorkshire fog (Holcus lanatus)/T. repens with or without Lotus corniculatus. There were four replicate groups of six lambs per treatment. The effects of condensed tannins (CT) on lamb production were assessed by twice-daily oral administration of 10g polyethylene glycol (PEG; molecular weight 4000) to half the lambs on each sward. The Lotus content of all swards was very low, and results are presented here for main sward comparisons meaned over lotus treatments. Overall mean estimates of pre-grazing herbage mass and sward surface height for the annual ryegrass and Yorkshire fog swards respectively, were 5820 v. 4360 +/- 190 kg DM/ha (P , P < 0.01) and liveweight gain (141 v. 120 +/- 4.3 g per lamb per day, P < 0.01), although differences in carcass weight (17.9 v. 18.2 +/- 0.3 kg) and FEC transformed values (9.6 v. 11.0 +/- 06 eggs/g fresh faeces) were not significant. The effects of CT on animal performance were greater in Yorkshire fog swards. CT had no significant effects on diet selection, herbage intake and grazing behaviour patterns
