    Licarin-B Exhibits Activity Against the Toxoplasma gondii RH Strain by Damaging Mitochondria and Activating Autophagy

    Toxoplasma gondii is an obligate intracellular pathogen that infects warm-blooded animals and humans. However, side effects limit toxoplasmosis treatment, and new drugs with high efficiency and low toxicity need to be developed. Natural products found in plants have become a useful source of drugs for toxoplasmosis. In this study, twenty natural compounds were screened for anti-T. gondii activity by Giemsa staining or real-time fluorescence quantitative polymerase chain reaction (qPCR) in vitro. Among these, licarin-B from nutmeg exhibited excellent anti-T. gondii activity, inhibiting T. gondii invasion and proliferation in a dose-dependent manner, with an EC50 of 14.05 ± 3.96 μg/mL. In the in vivo, licarin-B treatment significantly reduced the parasite burden in tissues compared to no treatment, protected the 90% infected mice from to death at 50 mg/kg.bw. Flow cytometry analysis suggested a significant reduction in T. gondii survival after licarin-B treatment. Ultrastructural changes in T. gondii were observed by transmission electron microscopy (TEM), as licarin-B induced mitochondrial swelling and formation of cytoplasmic vacuoles, an autophagosome-like double-membrane structure and extensive clefts around the T. gondii nucleus. Furthermore, MitoTracker Red CMXRos, MDC, and DAPI staining showed that licarin-B promoted mitochondrial damage, autophagosome formation, and nuclear disintegration, which were consistent with the TEM observations. Together, these findings indicate that licarin-B is a promising anti-T. gondii agent that potentially functions by damaging mitochondria and activating autophagy, leading to T. gondii death

    Agents: An Open-source Framework for Autonomous Language Agents

    Recent advances on large language models (LLMs) enable researchers and developers to build autonomous language agents that can automatically solve various tasks and interact with environments, humans, and other agents using natural language interfaces. We consider language agents as a promising direction towards artificial general intelligence and release Agents, an open-source library with the goal of opening up these advances to a wider non-specialist audience. Agents is carefully engineered to support important features including planning, memory, tool usage, multi-agent communication, and fine-grained symbolic control. Agents is user-friendly as it enables non-specialists to build, customize, test, tune, and deploy state-of-the-art autonomous language agents without much coding. The library is also research-friendly as its modularized design makes it easily extensible for researchers.     Phase Equilibria in the System CaO-SiO<sub>2</sub>-La<sub>2</sub>O<sub>3</sub>-Nb<sub>2</sub>O<sub>5</sub> at 1400 °C

    CaO-SiO2-La2O3-Nb2O5 system is of great significance for the pyrometallurgical utilization of Bayan Obo tailing resources. In the present work, the phase equilibrium of this quaternary system at 1400 °C was determined by a thermodynamic equilibrium experiment. On the basis of the recently determined CaO-La2O3-Nb2O5 phase diagram, some boundary surfaces of primary phase fields of CaO-SiO2-La2O3-Nb2O5 phase diagram were modified; then, the 1400 °C isothermal surface in the primary phase fields of SiO2, CaNb2O6, Ca2Nb2O7, and LaNbO4 was constructed, respectively. On this basis, CaO-SiO2-Nb2O5 pseudo-ternary phase diagrams with w(La2O3) = 5%, 10%, 15%, and 20% were determined, respectively. Considering the importance of equilibrium crystallization reaction type, we proposed a new rule named Tangent Line Rule to judge the univariant reaction type in the quaternary phase diagram. By applying Tangent Line Rule and Tangent Plane Rule previously proposed, some univariant and bivariant crystallization reaction types in the CaO-SiO2-La2O3- Nb2O5 phase diagram were determined, respectively. The current work can provide original data for the establishment of a thermodynamic database of Nb-bearing and REE-bearing slag system; the proposed Tangent Line Rule will promote the application of a spatial quaternary phase diagram

    Growth, Water Use, and Nitrate-15N Uptake of Greenhouse Tomato as Influenced by Different Irrigation Patterns, 15N Labeled Depths, and Transplant Times

    Increasing water use efficiency and reducing nitrogen pollutant discharge are important tasks for modern agriculture. To evaluate the effect of alternate partial root-zone irrigation (APRI) on tomato plant growth, water use efficiency and nitrate-15N uptake, an experiment was conducted from June to December in 2014 under greenhouse condition in northern China. The experiment contained two irrigation patterns [APRI and conventional irrigation (CI)], two 15N labeled depths in soil (10 and 50 cm) and two transplant time (early and late summer). Results showed that, compared to CI, APRI did not significantly (p &gt; 0.05) impact the growth and biomass accumulation in aboveground part of tomato, while it enhanced the root, reflecting by greater length density, and more dry mass. APRI produced marginally lower yields, but saved 34.9% of irrigation water, and gave a 37.6–49.9% higher water use efficiency relative to CI. In addition, APRI improved fruit quality, mainly through increasing the contents of soluble solid (by 12.8–21.6%), and vitamin C (2.8–12.7%), and the sugar/acid ratio (3.5–8.5%). The 15N utilization efficiency (15NUE) in APRI was higher than that in CI, which was more evident when 15N was labeled at 50 cm depth. Significant (p &lt; 0.05) 15N recovery increase of 10.2–13.2% and 15N loss decrease of 35.4–54.6% were found for APRI compared to CI. The overall results suggest that APRI under greenhouse could benefit the nitrate-N recovery and increase the water use efficiency in tomato

    Fast Detection of Striped Stem-Borer (Chilo suppressalis Walker) Infested Rice Seedling Based on Visible/Near-Infrared Hyperspectral Imaging System

    Striped stem-borer (SSB) infestation is one of the most serious sources of damage to rice growth. A rapid and non-destructive method of early SSB detection is essential for rice-growth protection. In this study, hyperspectral imaging combined with chemometrics was used to detect early SSB infestation in rice and identify the degree of infestation (DI). Visible/near-infrared hyperspectral images (in the spectral range of 380 nm to 1030 nm) were taken of the healthy rice plants and infested rice plants by SSB for 2, 4, 6, 8 and 10 days. A total of 17 characteristic wavelengths were selected from the spectral data extracted from the hyperspectral images by the successive projection algorithm (SPA). Principal component analysis (PCA) was applied to the hyperspectral images, and 16 textural features based on the gray-level co-occurrence matrix (GLCM) were extracted from the first two principal component (PC) images. A back-propagation neural network (BPNN) was used to establish infestation degree evaluation models based on full spectra, characteristic wavelengths, textural features and features fusion, respectively. BPNN models based on a fusion of characteristic wavelengths and textural features achieved the best performance, with classification accuracy of calibration and prediction sets over 95%. The accuracy of each infestation degree was satisfactory, and the accuracy of rice samples infested for 2 days was slightly low.
