    Effect of germination on nutritional quality of soybean

    Abstract Soybean are rich in functional nutrients such as protein, Essential amino acid, Polyunsaturated fatty acid, minerals, vitamins and dietary fibre which can help maintain optimal body weight, prevent Alzheimer and cardiovascular disease. At the same time, it also contains some anti-nutritional factors which have adverse effects on the digestion, absorption and utilization of nutrients. Cellular and metabolic events are induced by water absorption during seed germination. After absorbing water, soybean dry seeds begin to mobilize organelles and enzyme activities, repair and activate physiological and biochemical processes, and start basic metabolism. After that, by controlling the key factors of metabolism, the cells complete the process of material transformation, energy metabolism, ROS balance, cell wall acidification and looseness, hypocotyl elongation, etc. and finish germination. As a result, cell metabolism causes significant changes in proteins, lipids, sugars, vitamins and minerals, and increases in some beneficial functional nutritional factors for humans and animals, the other anti-nutritional factors decreased obviously, which greatly improved the taste quality and economic value of soybean. This paper introduced the germination physiology, functional nutrients and their metabolic pathways during soybean germination. This study has important reference value for the study of theory and practice technology of soybean germination

    Screening of the Key Genes and Signalling Pathways for Diabetic Nephropathy Using Bioinformatics Analysis

    BackgroundThis study aimed to identify biological markers for diabetic nephropathy (DN) and explore their underlying mechanisms.MethodsFour datasets, GSE30528, GSE47183, GSE104948, and GSE96804, were downloaded from the Gene Expression Omnibus (GEO) database. The differentially expressed genes (DEGs) were identified using the “limma” package, and the “RobustRankAggreg” package was used to screen the overlapping DEGs. The hub genes were identified using cytoHubba of Cytoscape. Logistic regression analysis was used to further analyse the hub genes, followed by receiver operating characteristic (ROC) curve analysis to predict the diagnostic effectiveness of the hub genes. Correlation analysis and enrichment analysis of the hub genes were performed to identify the potential functions of the hub genes involved in DN.ResultsIn total, 55 DEGs, including 38 upregulated and 17 downregulated genes, were identified from the three datasets. Four hub genes (FN1, CD44, C1QB, and C1QA) were screened out by the “UpSetR” package, and FN1 was identified as a key gene for DN by logistic regression analysis. Correlation analysis and enrichment analysis showed that FN1 was positively correlated with four genes (COL6A3, COL1A2, THBS2, and CD44) and with the development of DN through the extracellular matrix (ECM)–receptor interaction pathway.ConclusionsWe identified four candidate genes: FN1, C1QA, C1QB, and CD44. On further investigating the biological functions of FN1, we showed that FN1 was positively correlated with THBS2, COL1A2, COL6A3, and CD44 and involved in the development of DN through the ECM–receptor interaction pathway. THBS2, COL1A2, COL6A3, and CD44 may be novel biomarkers and target therapeutic candidates for DN

    Physicochemical properties, antioxidant activities and hypoglycemic effects of soluble dietary fibers purified from Lentinula edodes

    Abstract Lentinula edodes (L. edodes), which imparts various health benefits to humans, is considered a novel source of soluble dietary fiber (SDF). In this study, ultrasonic-assisted hot-water method was used to extract SDF (U-SDF) from L. edodes, and physicochemical, antioxidant and hypoglycemic properties of the U-SDF were investigated. Physicochemical properties of U-SDF showed that water solubility, water holding capacity, swelling capacity, and oil holding capacity were higher than that the SDF extracted using hot water method without ultrasonication. The DPPH, •OH, and •O2- radical clearance rates indicated that U-SDF exhibited better antioxidant activities. U-SDF also exhibited notable α-amylase and α-glucosidase inhibition activities. Treatment with U-SDF alleviated glucose and peroxidation metabolism disorders in vivo. Histological analysis indicated that U-SDF improved the oxidative tissue damage in diabetic mice. These results provided a theoretical basis for the development and utilization of SDF derived from L. edodes

    The importance of NOx control for peak ozone mitigation based on a sensitivity study using CMAQ‐HDDM‐3D model during a typical episode over the Yangtze River delta region, China.

    In recent years, ground-level ozone (O3) has been one of the main pollutants hindering air quality compliance in China's large city-clusters including the Yangtze River Delta (YRD) region. In this work, we utilized the process analysis (PA) and the higher-order decoupled direct method (HDDM-3D) tools embedded in the Community Multiscale Air Quality model (CMAQ) to characterize O3 formation and sensitivities to precursors during a typical O3 pollution episode over the YRD region in July 2018. Results indicate that gas-phase chemistry contributed dominantly to the ground-level O3 although a significant proportion was chemically produced at the middle and upper boundary layer before reaching the surface via diffusion process. Further analysis of the chemical pathways of O3 and Ox formation provided deep insights into the sensitivities of O3 to its precursors that were consistent with the HDDM results. The first-order sensitivities of O3 to anthropogenic volatile organic compounds (AVOC) were mainly positive but small, and temporal variations were negligible compared with those to NOx. During the peak O3 time in the afternoon, the first- and second-order sensitivities of O3 to NOx were significantly positive and negative, respectively, suggesting a convex response of O3 to NOx over most areas including Shanghai, Hangzhou, Nanjing and Hefei. These findings further highlighted an accelerated decrease in ground-level O3 in the afternoon corresponding to continuous decrease of NOx emissions in the afternoon. Therefore, over the YRD region including its metropolises, NOx emission reductions will be more important in reducing the afternoon peak O3 concentration compared with the effect of VOC emission control alone

    One-Pot Green Synthesis and Bioapplication ofl-Arginine-Capped Superparamagnetic Fe3O4 Nanoparticles

    Water-solublel-arginine-capped Fe3O4 nanoparticles were synthesized using a one-pot and green method. Nontoxic, renewable and inexpensive reagents including FeCl3,l-arginine, glycerol and water were chosen as raw materials. Fe3O4 nanoparticles show different dispersive states in acidic and alkaline solutions for the two distinct forms of surface bindingl-arginine. Powder X-ray diffraction and X-ray photoelectron spectroscopy were used to identify the structure of Fe3O4 nanocrystals. The products behave like superparamagnetism at room temperature with saturation magnetization of 49.9 emu g−1 and negligible remanence or coercivity. In the presence of 1-ethyl-3-(dimethylaminopropyl) carbodiimide hydrochloride, the anti-chloramphenicol monoclonal antibodies were connected to thel-arginine-capped magnetite nanoparticles.     A Plant Virus Ensures Viral Stability in the Hemolymph of Vector Insects through Suppressing Prophenoloxidase Activation

    Large ratios of vector-borne plant viruses circulate in the hemolymph of their vector insects before entering the salivary glands to be transmitted to plants. The stability of virions in the hemolymph is vital in this process. Activation of the proteolytic prophenoloxidase (PPO) to produce active phenoloxidase (PO) is one of the major innate immune pathways in insect hemolymph. How a plant virus copes with the PPO immune reaction in its vector insect remains unclear. Here, we report that the PPO affects the stability of rice stripe virus (RSV), a notorious rice virus, in the hemolymph of a vector insect, the small brown planthopper. RSV suppresses PPO activation using viral nonstructural protein. Once the level of PO activity is elevated, RSV is melanized and eliminated from the hemolymph. Our work gives valuable clues for developing novel strategies for controlling the transmission of vector-borne plant viruses.Most plant viruses require vector insects for transmission. Viral stability in the hemolymph of vector insects is a prerequisite for successful transmission of persistent plant viruses. However, knowledge of whether the proteolytic activation of prophenoloxidase (PPO) affects the stability of persistent plant viruses remains elusive. Here, we explored the interplay between rice stripe virus (RSV) and the PPO cascade of the vector small brown planthopper. Phenoloxidase (PO) activity was suppressed by RSV by approximately 60%. When the PPO cascade was activated, we found distinct melanization around RSV particles and serious damage to viral stability in the hemolymph. Viral suppression of PO activity was derived from obstruction of proteolytic cleavage of PPOs by binding of the viral nonstructural protein NS3. These results indicate that RSV attenuates the PPO response to ensure viral stability in the hemolymph of vector insects. Our research provides enlightening cues for controlling the transmission of vector-borne viruses

    Genome-wide identification and expression analysis of calmodulin and calmodulin-like genes in passion fruit (Passiflora edulis) and their involvement in flower and fruit development

    Abstract Background The calmodulin (CaM) and calmodulin-like (CML) proteins play regulatory roles in plant growth and development, responses to biotic and abiotic stresses, and other biological processes. As a popular fruit and ornamental crop, it is important to explore the regulatory mechanism of flower and fruit development of passion fruit. Results In this study, 32 PeCaM/PeCML genes were identified from passion fruit genome and were divided into 9 groups based on phylogenetic analysis. The structural analysis, including conserved motifs, gene structure and homologous modeling, illustrates that the PeCaM/PeCML in the same subgroup have relative conserved structural features. Collinearity analysis suggested that the expansion of the CaM/CML gene family likely took place mainly by segmental duplication, and the whole genome replication events were closely related with the rapid expansion of the gene group. PeCaM/PeCMLs were potentially required for different floral tissues development. Significantly, PeCML26 had extremely high expression levels during ovule and fruit development compared with other PeCML genes, suggesting that PeCML26 had potential functions involved in the development of passion fruit flowers and fruits. The co-presence of various cis-elements associated with growth and development, hormone responsiveness, and stress responsiveness in the promoter regions of these PeCaM/PeCMLs might contribute to their diverse regulatory roles. Furthermore, PeCaM/PeCMLs were also induced by various abiotic stresses. This work provides a comprehensive understanding of the CaM/CML gene family and valuable clues for future studies on the function and evolution of CaM/CML genes in passion fruit. Conclusion A total of 32 PeCaM/PeCML genes were divided into 9 groups. The PeCaM/PeCML genes showed differential expression patterns in floral tissues at different development stages. It is worth noting that PeCML26, which is highly homologous to AtCaM2, not only interacts with multiple BBR-BPC TFs, but also has high expression levels during ovule and fruit development, suggesting that PeCML26 had potential functions involved in the development of passion fruit flowers and fruits. This research lays the foundation for future investigations and validation of the potential function of PeCaM/PeCML genes in the growth and development of passion fruit
