    Region Proposal Rectification Towards Robust Instance Segmentation of Biological Images

    Top-down instance segmentation framework has shown its superiority in object detection compared to the bottom-up framework. While it is efficient in addressing over-segmentation, top-down instance segmentation suffers from over-crop problem. However, a complete segmentation mask is crucial for biological image analysis as it delivers important morphological properties such as shapes and volumes. In this paper, we propose a region proposal rectification (RPR) module to address this challenging incomplete segmentation problem. In particular, we offer a progressive ROIAlign module to introduce neighbor information into a series of ROIs gradually. The ROI features are fed into an attentive feed-forward network (FFN) for proposal box regression. With additional neighbor information, the proposed RPR module shows significant improvement in correction of region proposal locations and thereby exhibits favorable instance segmentation performances on three biological image datasets compared to state-of-the-art baseline methods. Experimental results demonstrate that the proposed RPR module is effective in both anchor-based and anchor-free top-down instance segmentation approaches, suggesting the proposed method can be applied to general top-down instance segmentation of biological images.     Difference of gut microbiota between patients with negative and positive HBeAg in chronic hepatitis B and the effect of tenofovir alafenamide on intestinal flora

    BackgroundSevere liver diseases, such as liver fibrosis, cirrhosis, and liver cancer, are mainly caused by hepatitis B virus (HBV). This study investigated the differences between gut microbiota in HBeAg-positive and negative groups of patients with chronic hepatitis B (CHB) and investigated the effect of tenofovir alafenamide (TAF) on gut microbiota.MethodsThis prospective study included patients with CHB not taking nucleoside antivirals (No-NAs group, n = 95) and those taking TAF (TAF group, n = 60). We divided CHB patients into two groups according to the HBeAg status of the subjects on the day of data collection. Phase 1 are HBeAg-negative patients and phase 2 are HBeAg-positive patients. We investigated the improvement of clinical symptoms by TAF, as well as differences in gut microbiota between different groups by 16S rRNA high-throughput sequencing.ResultsGut microbiota demonstrated significant differences between patients with HBeAg-positive and -negative CHB. Both the No-NAs and TAF Phase 2 subgroups demonstrated significantly increased microbiota richness and diversity, showing greater heterogeneity. Additionally, the Phase 2 subgroup exhibited a low abundance of pathways associated with glucose metabolism and amino acid metabolism. The TAF group demonstrated a significantly decreased HBV load, alanine aminotransferase, and aspartate aminotransferase and a significant increase in prealbumin compared with the No-NAs group. No significant difference was found in uric acid, creatinine, blood calcium, inorganic phosphorus, eGFR, and β2-microglobulin concentrations between the two groups. Additionally, the urea level in the TAF group was significantly lower than that in the No-NAs group, but with no significant effect on other indicators such as eGFR and β2-microglobulin.ConclusionThis study revealed significant differences in gut microbiota composition and function between patients with HBeAg-positive and -negative CHB

    Ivermectin induces apoptosis of esophageal squamous cell carcinoma via mitochondrial pathway

    Background: Esophageal squamous cell carcinoma (ESCC) is the most predominant primary malignant tumor among worldwide, especially in China. To date, the successful treatment remains a mainly clinical challenge, it is imperative to develop successful therapeutic agents. Methods: The anti-proliferative effect of ivermectin on ESCC is investigated in cell model and in nude mice model. Cell apoptosis was assessed using flow cytometry, TUNEL assay and western blotting. Mitochondrial dysfunction was determined by reactive oxygen species accumulation, mitochondrial membrane potential and ATP levels. Results: Our results determined that ivermectin significantly inhibited the proliferation of ESCC cells in vitro and in vivo. Furthermore, we found that ivermectin markedly mediated mitochondrial dysfunction and induced apoptosis of ESCC cells, which indicated the anti-proliferative effect of ivermectin on ESCC cells was implicated in mitochondrial apoptotic pathway. Mechanistically, ivermectin significantly triggered ROS accumulation and inhibited the activation of NF-κB signaling pathway and increased the ratio of Bax/Bcl-2. Conclusions: These finding indicated that ivermectin has significant anti-tumour potential for ESSC and may be a potential therapeutic candidate against ESCC

    Transportation systems need more accurate predictions to further optimize traffic network design with the development and application of autonomous driving technology. In this article, we focus on highway traffic flow systems that are often simulated by the modified Greenshields model. However, this model can not perfectly match the true traffic flow due to its underlying simplifications and assumptions, implying that it is inexact. Specifically, some parameters affect the simulation accuracy of the modified Greenshields model, while tuning these parameters to improve the model’s accuracy is called model calibration. The parameters obtained using the L2 calibration have the advantages of high accuracy and small variance for an inexact model. However, the method is calculation intensive, requiring optimization of the integral loss function. Since traffic flow data are often massive, this paper proposes a fast L2 calibration framework to calibrate the modified Greenshields model. Specifically, the suggested method selects a sub-design containing more information on the calibration parameters, and then the empirical loss function obtained from the optimal sub-design is utilized to approximate the integral loss function. A case study highlights that the proposed method preserves the advantages of L2 calibration and significantly reduces the running time
