    The Effects of Urethane on Rat Outer Hair Cells

    The cochlea converts sound vibration into electrical impulses and amplifies the low-level sound signal. Urethane, a widely used anesthetic in animal research, has been shown to reduce the neural responses to auditory stimuli. However, the effects of urethane on cochlea, especially on the function of outer hair cells, remain largely unknown. In the present study, we compared the cochlear microphonic responses between awake and urethane-anesthetized rats. The results revealed that the amplitude of the cochlear microphonic was decreased by urethane, resulting in an increase in the threshold at all of the sound frequencies examined. To deduce the possible mechanism underlying the urethane-induced decrease in cochlear sensitivity, we examined the electrical response properties of isolated outer hair cells using whole-cell patch-clamp recording. We found that urethane hyperpolarizes the outer hair cell membrane potential in a dose-dependent manner and elicits larger outward current. This urethane-induced outward current was blocked by strychnine, an antagonist of the α9 subunit of the nicotinic acetylcholine receptor. Meanwhile, the function of the outer hair cell motor protein, prestin, was not affected. These results suggest that urethane anesthesia is expected to decrease the responses of outer hair cells, whereas the frequency selectivity of cochlea remains unchanged

    Real-time Multi-person Eyeblink Detection in the Wild for Untrimmed Video

    Real-time eyeblink detection in the wild can widely serve for fatigue detection, face anti-spoofing, emotion analysis, etc. The existing research efforts generally focus on single-person cases towards trimmed video. However, multi-person scenario within untrimmed videos is also important for practical applications, which has not been well concerned yet. To address this, we shed light on this research field for the first time with essential contributions on dataset, theory, and practices. In particular, a large-scale dataset termed MPEblink that involves 686 untrimmed videos with 8748 eyeblink events is proposed under multi-person conditions. The samples are captured from unconstrained films to reveal "in the wild" characteristics. Meanwhile, a real-time multi-person eyeblink detection method is also proposed. Being different from the existing counterparts, our proposition runs in a one-stage spatio-temporal way with end-to-end learning capacity. Specifically, it simultaneously addresses the sub-tasks of face detection, face tracking, and human instance-level eyeblink detection. This paradigm holds 2 main advantages: (1) eyeblink features can be facilitated via the face's global context (e.g., head pose and illumination condition) with joint optimization and interaction, and (2) addressing these sub-tasks in parallel instead of sequential manner can save time remarkably to meet the real-time running requirement. Experiments on MPEblink verify the essential challenges of real-time multi-person eyeblink detection in the wild for untrimmed video. Our method also outperforms existing approaches by large margins and with a high inference speed.Comment: Accepted by CVPR 202

    Pituitary tumor transforming gene-1 haplotypes and risk of pituitary adenoma: a case-control study

    <p>Abstract</p> <p>Background</p> <p>It has been suggested that pituitary adenoma results from accumulation of multiple genetic and/or epigenetic aberrations, which may be identified through association studies. As pituitary tumor transforming gene-1 (<it>PTTG1</it>)/securin plays a critical role in promoting genomic instability in pituitary neoplasia, the present study explored the association of <it>PTTG1 </it>haplotypes with the risk of pituitary adenoma.</p> <p>Methods</p> <p>We genotyped five <it>PTTG1 </it>haplotype-tagging SNPs (htSNP) by PCR-RFLP assays in a case-control study, which included 280 Han Chinese patients diagnosed with pituitary adenoma and 280 age-, gender- and geographically matched Han Chinese controls. Haplotypes were reconstructed according to the genotyping data and linkage disequilibrium status of the htSNPs.</p> <p>Results</p> <p>No significant differences in allele and genotype frequencies of the htSNPs were observed between pituitary adenoma patients and controls, indicating that none of the individual <it>PTTG1 </it>SNPs examined in this study is associated with the risk of pituitary adenoma. In addition, no significant association was detected between the reconstructed <it>PTTG1 </it>haplotypes and pituitary adenoma cases or the controls.</p> <p>Conclusions</p> <p>Though no significant association was found between <it>PTTG1 </it>haplotypes and the risk of pituitary adenoma, this is the first report on the association of individual <it>PTTG1 </it>SNPs or <it>PTTG1 </it>haplotypes with the risk of pituitary adenoma based on a solid study; it will provide an important reference for future studies on the association between genetic alterations in <it>PTTG1 </it>and the risk of pituitary adenoma or other tumors.</p

    An Intronic Variant in the GRP78, a Stress-Associated Gene, Improves Prediction for Liver Cirrhosis in Persistent HBV Carriers

    Background: Our previous study indicated that a common variant (rs430397 G>A) in the intron 5 of glucose-regulated protein 78 (GRP78) gene was associated with risk and prognosis of primary hepatocellular carcinoma (HCC), including HBV- and cirrhosis-related HCC. rs430397 polymorphism may be a contributing factor or biomarker of HBV infection or HBV-related cirrhosis. Methodology/Principal Findings: 539 non-HBV-infected individuals, 205 self-limited infection and 496 persistent HBV infection were recruited between January 2001 and April 2005 from the hospitals in Southern China. Genomic DNA was genotyped for rs430397. The associations between the variation and susceptibility to liver cirrhosis (LC) in persistent HBV infection were examined. We observed that individuals carrying allele rs430397A were more likely to become HBV-related LC. When persistently infected patients were divided into four subgroups, patients with phase IV had an increased allele A and genotype AG compared with phase I and/or phase III. Decreased serum albumin and prolonged plasma prothrombin time (PT) were showed in LC patients carrying genotype AA. Furthermore, rs430397 genotype had an increased susceptibility to LC with dose-dependent manners (P-trend = 0.005), and the genotype did constitute a risk factor for the development of advanced LC (Child-Pugh classification C and B, P-trend = 0.021). Conclusions/Significance: rs430397 polymorphism may be a contributing factor to LC in persistent HBV carriers. © 2011 Zhu et al.published_or_final_versio

    The Impact of Social Capital on Multidimensional Poverty of Rural Households in China

    Getting rid of multidimensional poverty is both the people&rsquo;s wish and the direction of governance. Based on 2018 China Household Tracking Survey (CFPS) data, this paper identifies household multidimensional poverty in rural areas using a combination of the MPI index and the A-F method. The relationship between social capital and multidimensional poverty was also empirically analyzed using a Logit model. The results showed that: (1) 1599 multidimensional poverty households were identified, the incidence of multidimensional poverty was 24.94%, and the multidimensional poverty index was 0.103. In terms of poverty incidence, the three highest indicators of poverty incidence are adult education, health, and chronic diseases, reaching 42.06%, 37.65%, and 29.90%, respectively, and mainly concentrated in the education and health care systems. (2) Social capital can significantly reduce the probability of multidimensional poverty in rural households. Among them, social network significantly and negatively affects the occurrence of multidimensional poverty in rural households, social trust in neighbors has a significant negative effect on multidimensional poverty in rural households at the 1% level, and social prestige is positively related to multidimensional poverty in rural households. (3) Age of household head, household size, and income from working outside the home are significantly associated with multidimensional poverty in rural households. This paper expands the scope of social capital theory research and provides new perspectives and empirical evidence for alleviating multidimensional poverty

    Transcriptome Analysis of the Toxic Effects of Amisulbrom and Isoflucypram on Zebrafish (<i>Danio rerio</i>) Larvae

    Fungicides are frequently detected in the water bodies, however, the adverse effects of these fungicides on aquatic lives remain limited. To better understand the adverse effects of amisulbrom (AML) and isoflucypram (ISO) on embryogenesis, zebrafish embryos were exposed to two different fungicides, 0.75 μM amisulbrom (AML) and 2.5 μM isoflucypram (ISO), for 72 h. Transcriptome sequencing was employed to identify differentially expressed genes (DEGs) after AML and ISO exposure. A total of 571 and 3471 DEGs were detected between the libraries of the two fungicides-treated groups and the control, respectively. Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis showed that PPAR signaling pathway, phototransduction, ribosome and p53 signaling pathway were significantly enriched in response to both AML and ISO stress. Moreover, a number of DEGs involved in tyrosine metabolism, phagosome pathway, cell cycle pathway, extracellular matrix (ECM) receptor interaction pathway, and arginine and proline metabolism were specially enriched after exposure to AML; a number of DEGs involved in notch signaling pathway, drug metabolism, alanine, aspartate and glutamate metabolism, amino-acyl−tRNA biosynthesis, and protein processing in endoplasmic reticulum were significantly enriched after exposure to ISO. These results provide novel insights into the toxicological mechanisms underlying fish’s responses to fungicides
