    <p>Abstract</p> <p>Background</p> <p>Endothelial inflammatory responses mediated by Toll-like receptors (TLRs), particularly TLR2 and TLR4, play an important role in atherogenesis. While Type 1 diabetes (T1D) promotes the development and progression of atherosclerosis, the effect of T1D on TLR2/4-mediated inflammatory responses in coronary artery endothelial cells (CAECs) remains unclear.</p> <p>Methods</p> <p>We tested the hypothesis that diabetic CAECs have enhanced inflammatory responses to TLR2/4 stimulation. Non-diabetic and diabetic CAECs were treated with TLR2 agonist peptidoglycan and TLR4 agonist lipopolysaccharide. The expression of ICAM-1, IL-6 and IL-8 were analyzed by real-time PCR, immunoblotting and ELISA, and NF-ÎșB activation by immunoblotting and immunostaining. In additional experiments, insulin was added before TLR stimulation to determine whether insulin deficiency alone is responsible for the alteration of TLR2/4-mediated inflammatory responses.</p> <p>Results</p> <p>Stimulation of TLR2 or TLR4 induced NF-ÎșB activation, and the expression of ICAM-1, IL-6 and IL-8. Interestingly, the expression of inflammatory mediators was significantly enhanced in diabetic cells. The enhanced inflammatory responses correlated with augmented NF-ÎșB activation in the absence of a change in TLR2 or TLR4 protein levels. Further, pretreatment of diabetic cells with insulin failed to suppress the enhanced inflammatory responses.</p> <p>Conclusions</p> <p>Diabetic CAECs have enhanced inflammatory responses to stimulation of TLR2 or TLR4, and insulin alone is insufficient to correct the hyper-inflammatory responses. The mechanism underlying the enhanced inflammatory responses appears to be augmentation of pro-inflammatory signaling, rather than up-regulation of levels of TLR2 and TLR4. These findings suggest that diabetic CAECs adopt a hyper-inflammatory phenotype and that this endothelial phenotypic change may predispose coronary artery to atherogenesis.</p

    Multiobjective Optimization Design of a Fractional Order PID Controller for a Gun Control System

    Motion control of gun barrels is an ongoing topic for the development of gun control equipments possessing excellent performances. In this paper, a typical fractional order PID control strategy is employed for the gun control system. To obtain optimal parameters of the controller, a multiobjective optimization scheme is developed from the loop-shaping perspective. To solve the specified nonlinear optimization problem, a novel Pareto optimal solution based multiobjective differential evolution algorithm is proposed. To enhance the convergent rate of the optimization process, an opposition based learning method is embedded in the chaotic population initialization process. To enhance the robustness of the algorithm for different problems, an adapting scheme of the mutation operation is further employed. With assistance of the evolutionary algorithm, the optimal solution for the specified problem is selected. The numerical simulation results show that the control system can rapidly follow the demand signal with high accuracy and high robustness, demonstrating the efficiency of the proposed controller parameter tuning method

    Microalgae-mediated tandem culture of shrimp and bivalve: an environmental and health co-benefits solution for phosphorus recovery and emission reduction

    Phosphorus (P) accumulation in aquaculture systems is damaging our environment beyond acceptable levels. Devising strategies to potentially recover P from aquaculture systems in a reusable bioresource form is paramount and aligns with circular economy policies. In this study, we constructed two culture models, monoculture (Mon) and tandem culture (Tan), using Exopalaemon carinicauda and Mercenaria mercenaria. By monitoring the performance of rearing organisms, P dynamic patterns, and pollutant emissions, we found that: i) Compared to the Mon system, the Tan system demonstrated no differences in the performance of E. carinicauda and M. mercenaria, suggesting that the Tan model was viable in terms of fishery yield; ii) P in the Tan system could be efficiently recovered and removed from water and sediment, as indicated by the lower phosphate concentration in water (0.01 mg L−1), and the decrease in labile P in surface sediment (from 0.04 to 0.02 mg L−1). A combination of assimilatory and dissimilatory processes, mediated by phototrophic (bait-microalgae) and heterotrophic organisms (bivalves), appeared to be the primary mechanism for P utilization and removal; iii) The Tan system reduced pollutant emissions four times lower than the Mon system due to its minimal tailwater discharge (10%, 230 L). The emissions of total P, phosphate, total organic carbon, ammonium, and chemical oxygen demand from the Tan systems were 19 mg m−2 d−1, 2 mg m−2 d−1, 2 g m−2 d−1, 38 mg m−2 d−1, and 11 g m−2 d−1, respectively, 1.3, 1.7, 1.4, 1.3, and 1.2 times lower than those from the Mon systems. The eco-friendly Tan culture model fully exploited the resources of pond culture, a solution with environmental and health co-benefits for P recovery and emission reduction
