    Genetic characterization of Measles Viruses in China, 2004

    Genetic characterization of wild-type measles virus was studied using nucleotide sequencing of the C-terminal region of the N protein gene and phylogenetic analysis on 59 isolates from 16 provinces of China in 2004. The results showed that all of the isolates belonged to genotype H1. 51 isolates were belonged to cluster 1 and 8 isolates were cluster 2 and Viruses from both clusters were distributed throughout China without distinct geographic pattern. The nucleotide sequence and predicted amino acid homologies of the 59 H1 strains were 96.5%–100% and 95.7%–100%, respectively. The report showed that the transmission pattern of genotype H1 viruses in China in 2004 was consistent with ongoing endemic transmission of multiple lineages of a single, endemic genotype. Multiple transmission pathways leaded to multiple lineages within endemic genotype

    Molecular epidemiology of measles viruses in China, 1995–2003

    This report describes the genetic characterization of 297 wild-type measles viruses that were isolated in 24 provinces of China between 1995 and 2003. Phylogenetic analysis of the N gene sequences showed that all of the isolates belonged to genotype H1 except 3 isolates, which were genotype A. The nucleotide sequence and predicted amino acid homologies of the 294-genotype H1 strains were 94.7%–100% and 93.3%–100%, respectively. The genotype H1 isolates were divided into 2 clusters, which differed by approximately 2.9% at the nucleotide level. Viruses from both clusters were distributed throughout China with no apparent geographic restriction and multiple co-circulating lineages were present in many provinces. Even though other measles genotypes have been detected in countries that border China, this report shows that genotype H1 is widely distributed throughout the country and that China has a single, endemic genotype. This important baseline data will help to monitor the progress of measles control in China

    Epigenetic Modification of TLRs in Leukocytes Is Associated with Increased Susceptibility to Salmonella enteritidis in Chickens

    Toll-like receptors (TLRs) signaling pathways are the first lines in defense against Salmonella enteritidis (S. enteritidis) infection but the molecular mechanism underlying susceptibility to S. enteritidis infection in chicken remains unclear. SPF chickens injected with S. enteritidis were partitioned into two groups, one consisted of those from Salmonella-susceptible chickens (died within 5 d after injection, n = 6), the other consisted of six Salmonella-resistant chickens that survived for 15 d after injection. The present study shows that the bacterial load in susceptible chickens was significantly higher than that in resistant chickens and TLR4, TLR2-1 and TLR21 expression was strongly diminished in the leukocytes of susceptible chickens compared with those of resistant chickens. The induction of expression of pro-inflammatory cytokine genes, IL-6 and IFN-β, was greatly enhanced in the resistant but not in susceptible chickens. Contrasting with the reduced expression of TLR genes, those of the zinc finger protein 493 (ZNF493) gene and Toll-interacting protein (TOLLIP) gene were enhanced in the susceptible chickens. Finally, the expression of TLR4 in peripheral blood mononuclear cells (PBMCs) infected in vitro with S. enteritidis increased significantly as a result of treatment with 5-Aza-2-deoxycytidine (5-Aza-dc) while either 5-Aza-dc or trichostatin A was effective in up-regulating the expression of TLR21 and TLR2-1. DNA methylation, in the predicted promoter region of TLR4 and TLR21 genes, and an exonic CpG island of the TLR2-1 gene was significantly higher in the susceptible chickens than in resistant chickens. Taken together, the results demonstrate that ZNF493-related epigenetic modification in leukocytes probably accounts for increased susceptibility to S. enteritidis in chickens by diminishing the expression and response of TLR4, TLR21 and TLR2-1

    Differentially expressed genes in a flock of Chinese local-breed chickens infected with a subgroup J avian leukosis virus using suppression subtractive hybridization

    Avian leukosis virus subgroup J (ALV-J) is a new type of virus that mainly induces myeloid leukosis (ML) in chickens. To further elucidate the pathogenesis of ALV-J infection and tumor development, expression profiles from the bone marrow tissue of 15 infected and 18 non-infected birds from a local-breed poultry-farm under naturally infected conditions, were analyzed by suppression-subtractive hybridization. The birds were diagnosed as ML+ (or ML-) by specific ALV-J detection methods, involving serological tests for antigens and antibodies, and RT-PCR to detect viral RNA. A total of 59 partial gene sequences were revealed by differential screening of 496 forward and 384 reverse subtracted cDNA clones. Of these, 22 identified genes, including 8 up-regulated and 14 down-regulated, were related to immune functions, these genes being, MHC B-G antigen, translationally-controlled tumor protein (TPT1/TPTC), transferrin and ferritin, hemoglobin and Carbonic anhydrase. Four of the down-regulated genes were selected for further analysis, in view of their predicted roles in infection and immunity by real-time qRT-PCR, using RNA collected from the same birds as those used for SSH. The four genes were expressed at significantly lower levels (p < 0.001) in ALV-J infected birds than in non-infected ones

    Extreme heatwave over Eastern China in summer 2022: the role of three oceans and local soil moisture feedback

    Eastern China experienced persistent regional extreme heatwaves in the summer of 2022, with disparate spatial features and formation mechanisms in different months. We quantitatively assessed the relative contributions of three oceans, i.e. tropical Indian Ocean and Pacific and North Atlantic, and the local soil moisture–temperature feedback using linear regression. The results showed that the monthly mean atmospheric circulation anomalies failed to explain the extreme heatwave in June 2022. The combined contribution of the tropical Indo-Pacific and North Atlantic sea surface temperature anomalies (SSTAs), together with the local soil moisture–temperature feedback, explaining approximately 10% of the temperature anomalies. In July, the tropical Indo-Pacific SSTAs promoted anomalous atmospheric circulation and extreme heat via meridional circulation originating in the Maritime Continent, accounting for approximately 10% of the temperature anomalies, with North Atlantic SSTAs contributing the same percentage by a mid-latitude steady Rossby wave. Local soil moisture–temperature feedback accounted for 42% of the anomalies. The tropical Indo-Pacific SSTAs produced a strong western North Pacific anticyclone in August, but their direct contribution to the temperature anomalies was negligible. The North Atlantic SSTAs contributed 9% of the total via the mid-latitude steady Rossby wave. Local soil moisture–temperature feedback contributed 66%, suggesting that the July heatwave and drought exerted a significant impact on the subsequent August extreme heatwave. Global warming has greatly facilitated extreme heatwaves, accounting for about 30%–40% of these events in summer 2022. These results also suggest that the climatic effects of tropical Indo-Pacific and North Atlantic SSTAs on Eastern China are evident in the month-to-month variation in summer. Our results thus contribute to the understanding and prediction of extreme heatwaves in Eastern China

    Anti-aging Effect of Ginseng Extract and Its Mechanism on Drosophila melanogaster

    Objective: Based on the model organism of Drosophila melanogaster, the anti-aging effect of ginseng extract and its mechanism on Drosophila melanogaster were investigated. Methods: The composition of ginsenosides in ginseng extract were analyzed by HPLC. Wild Canton-S Drosophila melanogaster were randomly divided into control group and ginseng extract group. The life span test, food intake, triglyceride level, reproductive ability, exercise ability, and the survival time of Drosophila melanogaster under starvation, heat stress and oxidative stress were applied to analyze the anti-aging function of ginseng extract. The transcription levels of genes related to AMP-activated protein kinase (AMPK) pathway and mammalian target of rapamycin (mTOR) pathway were determined by polymerase chain reaction. Results: Ginseng extract contained 13 ginsenosides. Compared with the control group, 7.5 mg/mL ginseng extract could prolong the life span of Drosophila melanogaster by 9.72%, and had no significant effect on the food intake, triglyceride level and fecundity (P>0.05), significantly improved the climbing ability (P<0.05 or P<0.01), and increased the ability to resist stress (P<0.05 or P<0.01). Compared with the control group, the intake of ginseng extract significantly increased the transcript levels of AMPKα, FoxO, Sirt1, LKB1 and CaMKII (P<0.05 or P<0.01), decreased the transcript levels of HDAC4 and SREBP (P<0.05), and enhanced ATP content and ATP/ADP ratio (P<0.05 or P<0.01). In addition, ginseng extract significantly decreased the transcription levels of mTOR-related genes PI3K, TORC, 4E-BP and S6K (P<0.05 or P<0.01), and significantly increased the transcription levels of Atg1, Atg5, Atg8a and Atg8b (P<0.01). Conclusion: Ginseng extract could delay the aging of Drosophila melanogaster by activating AMPK pathway and inhibiting mTOR pathway

    The Effect of Supplemental Concentrate Feeding on the Morphological and Functional Development of the Pancreas in Early Weaned Yak Calves

    This experiment was conducted to investigate the effect of supplemental concentrate feeding on the pancreatic development of yak calves. Twenty one-month-old yak calves with healthy body condition and similar body weight were selected as experimental animals and randomly divided into two groups, five replicates in each group. The control group yak calves were fed milk replacer and alfalfa hay, the experimental group yak calves were fed milk replacer, alfalfa hay and concentrate. The pre-feeding period of this experiment was thirty days, the trial period was one hundred days. At the end of feeding trail, five yak calves from each group were selected and slaughtered and the pancreas tissues of yak calves were collected and determined. The results showed that: (1) Dry matter and body weight of yak calves in the test group were significantly higher than those of the control group. (2) The apparent nutrient digestibility of crude protein, crude fat, calcium and phosphorus in the test group of yak calves was significantly higher than that of the control group, while the apparent nutrient digestibility of neutral detergent fiber and acid detergent fiber in the test group was significantly lower than that of the control group. (3) Pancreatic weight, organ index, total ratio of exocrine part area and total ratio of endocrine area of yak calves in the test group were significantly higher than those in the control group, while the ratio of exocrine area was significantly lower in the test group than that of the control group. (4) The activities of the main pancreatic digestive enzymes: pancreatic amylase, pancreatic lipase, pancreatic protease and chymotrypsin were significantly higher in the test group than those of the control group, as were the hormonal contents of glucagon, insulin and pancreatic polypeptide. (5) The main differential metabolites of the pancreas in the test group were significantly higher than those of the control group, such as D-proline, hypoxanthine, acetylcysteine, gamma-glutamylcysteine, thiazolidine-4-carboxylic acid, piperidinic acid, ellagic acid, nicotinamide, tropolone, D-serine, ribulose-5-phosphate, (+/-)5(6)-epoxyeicosatrienoic acid(EET), 2-hydroxycinnamic acid, L-phenylalanine, creatinine, tetrahydrocorticosterone, pyridoxamine, xanthine, 5-oxoproline, asparagine, DL-tryptophan, in-dole-3-acrylic acid, thymine, trehalose, docosapentaenoic acid, docosahexaenoic acid, fatty acid esters of hydroxy fatty acids(FAHFA) (18:1/20:3), fatty acid esters of hydroxy fatty acids(FAHFA) (18:2/20:4), adrenic acid and xanthosine. In conclusion, supplemental concentrate feeding promoted the good development of morphological and functional properties of the pancreas in early weaned yak calves to improve the digestion and absorption of feed nutrients, so as to enhance the growth and development quality of early weaned yak calves

    Effect of Cage and Floor Housing Systems on Muscle Fiber Characteristics, Carcass Characteristics, and Meat Quality of Slow-Growing Meat-Type Chickens

    This study compared floor (FS) with cage systems (CS) for slow-growing meat-type chickens in terms of muscle fiber characteristics, carcass characteristics, and meat quality. Following the design of a factorial arrangement of 2 housing systems × 2 genders, 180 male and 180 female Beijing You chickens of 8 weeks old were allocated to FS and CS. At the end of 17 weeks, five males and five females from each replicate were selected for measurement. No difference was observed in body, carcass, or eviscerated weight (p > 0.05). FS birds showed higher muscle yield and lower abdominal fat composition (p p > 0.05). On the contrary, intramuscular fat (IMF) content was affected by both in a way that CS birds and females had higher IMF content (p p < 0.05). In conclusion, carcass characteristics, meat quality, and muscle fiber type of slowing-growing broilers are influenced: rearing on the floor may improve muscle development and reduce fat deposition without impairing marketing weight and rearing in cages may improve IMF content
