    Effect of Plasma Viremia on Apoptosis and Immunophenotype of Dendritic Cells Subsets in Acute SIVmac239 Infection of Chinese Rhesus Macaques

    Non-human primates such as Chinese rhesus macaques (Ch Rhs) provide good animal models for research on human infectious diseases. Similar to humans, there are two principal subsets of dendritic cells (DCs) in the peripheral blood of Ch Rhs: myeloid DCs (mDCs) and plasmacytoid DCs (pDCs). In this study, two-color fluorescence-activated cell sorting (FACS) analyses were used to identify the main DC subsets, namely CD1c+ mDCs and pDCs from Ch Rhs. Then, the apoptosis and immunophenotype changes of DCs subsets were first described during the acute phase of SIVmac239 infection. Both the DCs subsets showed decreased CD4 expression and enhanced CCR5 expression; in particular, those of pDCs significantly changed at most time points. Interestingly, the plasma viral loads were negatively correlated with CD4 expression, but were positively correlated with CCR5 expression of pDCs. During this period, both CD1c+ mDCs and pDCs were activated by enhancing expressions of co-stimulatory molecules, accompanied with increase in CCR7. Either CD80 or CD86 expressed on CD1c+ mDCs and pDCs was positively correlated with the plasma viral loads. Our analysis demonstrates that the pDCs were more prone to apoptosis after infection during the acute phase of SIVmac239 infection, which may be due to their high expressions of CD4 and CCR5. Both DCs subsets activated through elevating the expression of co-stimulatory molecules, which was beneficial in controlling the replication of SIV. However, a mere broad immune activation initiated by activated DCs may lead to tragic AIDS progression

    Experimental treatment of radiation pneumonitis with human umbilical cord mesenchymal stem cells

    AbstractObjectiveTo evaluate of the curative effect of human umbilical cord mesenchymal stem cells (hUC-MSCs) on rat acute radiation pneumonitis.MethodsFourty rats were randomly divided into control group, radiation group, stem cell prevention group, stem cell treatment group and prednisone treatment group. All rats except those in the control group were radiated with X ray to establish the acute radiation pneumonitis damage model. The hUC-MSCs cultured in vitro was administrated to the rats of the prevention group via tail vein (1×106 cells/kg BW) 24 h before the radiation, while the same administration was performed in the rats of the treatment group 24 h after the radiation. After 24 h post the radiation, the rats in the radiation group were given 0.4 mL physiological saline, and those in the prednisone group were given 1 mg/kg prednisone. All rats were observed and executed 72 h after the radiation to detect lung histological changes.ResultsAfter the administration of hUC-MSCs, the survival status of the rats in the prevention group and treatment group was obviously better than that in the control group. As shown by the histological staining, the morphology, proliferation activity and bronchial state of lung tissues were better in the prevention group and treatment group than in the control group.ConclusionsThe hUC-MSCs have definite therapeutic effects on acute radiation pneumonitis in rats

    Isospin Effect on the Process of Multifragmentation and Dissipation at Intermediate Energy Heavy Ion Collisions

    In the simulation of intermediate energy heavy ion collisions by using the isospin dependent quantum molecular dynamics, the isospin effect on the process of multifragmentation and dissipation has been studied. It is found that the multiplicity of intermediate mass fragments NimfN_{imf} for the neutron-poor colliding system is always larger than that for the neutron-rich system, while the quadrupole of single particle momentum distribution QzzQ_{zz} for the neutron-poor colliding system is smaller than that of the neutron-rich system for all projectile-target combinations studied at the beam energies from about 50MeV/nucleon to 150MeV/nucleon. Since QzzQ_{zz} depends strongly on isospin dependence of in-medium nucleon-nucleon cross section and weakly on symmetry potential at the above beam energies, it may serve as a good probe to extract the information on the in-medium nucleon-nucleon cross section. The correlation between the multiplicity NimfN_{imf} of intermediate mass fragments and the total numer of charged particles NcN_c has the behavior similar to QzzQ_{zz}, which can be used as a complementary probe to the in-medium nucleon-nucleon cross section.Comment: 18 pages, 9 figure

    Dendritic cell subsets dynamics and cytokine production in SIVmac239-infected Chinese rhesus macaques

    BACKGROUND: Several studies have demonstrated that SIV infection progresses more slowly to experimental AIDS in Chinese rhesus macaques (Ch Rhs) than in Indian rhesus macaques (Ind Rhs). Here we investigated the dynamic and functional changes in dendritic cell (DC) subsets in SIVmac239-infected Ch Rhs. RESULTS: The numbers of both mDC and pDC strongly fluctuated but were not significantly changed during the acute and chronic phases of infection. However, the concentration of both poly (I:C)-induced IL-12 and HSV-1-induced IFN-α significantly increased in the acute phase of infection but returned to normal levels at the chronic phase of infection. The peak of IFN-α emerged earlier than that of IL-12, and it had a significantly positive correlation with IL-12, which indicated that IFN-α may initiate the immune activation. We also found that only the concentration of IFN-α was positively correlated with CD4+ T-cell counts, but it was negatively correlated with viral load. CONCLUSION: High levels of IFN-α in the early stage of infection may contribute to effective control of virus replication, and normal levels of IFN-α during chronic infection may help Ch Rhs resist the disease progression. The change in DC subsets dynamics and cytokine production may help further our understanding of why Ch Rhs are able to live longer without progressing to an AIDS-like illness

    Topical application of TAT-Superoxide Dismutase in Acupoints LI 20 on allergic rhinitis

    Reactive oxygen species are products of cellular metabolism and assigned important roles in biomedical science as deleterious factors in pathologies. In fact, some studies have shown that the therapeutic benefits of taking antioxidants were limited and the potential for therapeutic intervention remains unclear. New evidences showed that ROS have some ability of intercellular transportation. For treating allergic rhinitis, as a novel intracellular superoxide quencher, TAT-SOD applied to acupoints LI 20 instead of directly to nasal cavity can be used to test that. TTA group apply TAT-SOD cream prepared by adding purified TAT-SOD to the vehicle cream to acupoints LI 20, while placebo group used the vehicle cream instead. TTN group applied the same TAT-SOD cream directly to nasal cavity three times daily. Symptom scores were recorded at baseline and days 8 and 15. For the overall efficacy rate, TTA group was 81.0%, while placebo group was 5.9% and TTN was 0%. Malondialdehyde levels decreased observably in TTA group, and superoxide dismutase, catalase, and glutathione peroxidase levels remained basically unaffected. Enzymatic scavenging of the intracellular superoxide at acupoints LI 20 proved to be effective in treating allergic rhinitis, while no improvement was observed with the placebo group and TTN group

    Automatic Search of Meet-in-the-Middle Preimage Attacks on AES-like Hashing

    The Meet-in-the-Middle (MITM) preimage attack is highly effective in breaking the preimage resistance of many hash functions, including but not limited to the full MD5, HAVAL, and Tiger, and reduced SHA-0/1/2. It was also shown to be a threat to hash functions built on block ciphers like AES by Sasaki in 2011. Recently, such attacks on AES hashing modes evolved from merely using the freedom of choosing the internal state to also exploiting the freedom of choosing the message state. However, detecting such attacks especially those evolved variants is difficult. In previous works, the search space of the configurations of such attacks is limited, such that manual analysis is practical, which results in sub-optimal solutions. In this paper, we remove artificial limitations in previous works, formulate the essential ideas of the construction of the attack in well-defined ways, and translate the problem of searching for the best attacks into optimization problems under constraints in Mixed-Integer-Linear-Programming (MILP) models. The MILP models capture a large solution space of valid attacks; and the objectives of the MILP models are attack configurations with the minimized computational complexity. With such MILP models and using the off-the-shelf solver, it is efficient to search for the best attacks exhaustively. As a result, we obtain the first attacks against the full (5-round) and an extended (5.5-round) version of Haraka-512 v2, and 8-round AES-128 hashing modes, as well as improved attacks covering more rounds of Haraka-256 v2 and other members of AES and Rijndael hashing modes
