Comparisons of ELISA and Western blot assays for detection of autophagy flux
We analyzed autophagy/mitophagy flux in vitro (C2C12 myotubes) and in vivo (mouse skeletal muscle) following the treatments of autophagy inducers (starvation, rapamycin) and a mitophagy inducer (carbonyl cyanide m-chlorophenylhydrazone, CCCP) using two immunodetection methods, ELISA and Western blotting, and compared their working range, accuracy, and reliability. The ELISAs showed a broader working range than that of the LC3 Western blots (Table 1). Table 2 showed that data value distribution was tighter and the average standard error from the ELISA was much smaller than those of the Western blot, directly relating to the accuracy of the assay. Test-retest reliability analysis showed good reliability for three individual ELISAs (interclass correlation, ≥ 0.7), but poor reliability for three individual Western blots (interclass correlation, ≤ 0.4) (Table 3). Geometric mean for T-positive definite tensors and associated Riemannian geometry
In this paper, we generalize the geometric mean of two positive definite
matrices to that of third-order tensors using the notion of T-product.
Specifically, we define the geometric mean of two T-positive definite tensors
and verify several properties that "mean" should satisfy including the
idempotence and the commutative property, and so on. Moreover, it is shown that
the geometric mean is a unique T-positive definite solution of an algebraic
Riccati tensor equation and can be expressed as solutions of algebraic Riccati
matrix equations. In addition, we investigate the Riemannian manifold
associated with the geometric mean for T-positive definite tensors, considering
it as a totally geodesic embedded submanifold of the Riemannian manifold
associated with the case of matrices. It is particularly shown that the
geometric mean of two T-positive definite tensors is the midpoint of a unique
geodesic joining the tensors, and the manifold is a Cartan-Hadamard-Riemannian
manifold.Comment: 20 page
IDENTIFYING CRITICAL KINEMATIC PARAMETERS FOR BETTER GOLF PUTTING
In modern golf competition, putting is one of the crucial parts of the game. It has been reported that putting accounts for about 40% of all golf shot played in tournaments (Gwyn & Patch, 1993). Wiren (1992) also indicated that, on average, putting constitutes 38% of all golf strokes in competition and improving putting skills is th e fastest way to lower the score. However, it is also true that most recreational golfers neglect the putting and seldom practice it hard. Despite this revealing statistics and the obvious
importance of competent putting, much of the pedagogical literature is based on the observations and anecdotal evidence provided by top players and coaches (Paradisis & Rees, 2004). Therefore, the purpose of this study was to identify critical kinematic parameters of a putt by comparing putts performed by elite and novice golifers, and nongolfers. The findings might provide valuable information for improving putting performance
Regulatory gaps between LNG carriers and LNG fuelled ships
Given a number of marine vessels treating the liquefied natural gas either as cargo or fuel, this paper examined the regulatory gaps of two different international Codes – the InternationalCodeof the Construction and Equipment of Ships Carrying Liquefied Gases in Bulk and the International Code of Safety for Ships Using Gases or Other Low-flashpoint Fuels – from the regulatory standpoint. Results of the gap analysis have identified and discussed the key areas encountered with regulatory discrepancies or ambiguities that might interrupt the proper design, construction and operation of LNG carrier and LNG fuelled ship. A systematic investigation and harmonisation process across the Codes was proposed to mitigate the potential issues that may arise from the discordant regulations. Also, the International Maritime Organization was suggested to take proactive action to improve such dissonances while a general insight into the importance of filling those gaps was provided for rule-makers and stakeholders
Life cycle assessment of LPG engines for small fishing vessels and the applications of bio LPG fuel in Korea
This study aimed to evaluate the environmental impact of using liquefied petroleum gas (LPG) in small fishing vessels by conducting a life cycle assessment (LCA) in Korea. For the first time in the country, LPG engines designed for small fishing ships were utilized in this study. In addition, this research examined the potential benefits of employing Bio LPG, a renewable LPG produced from two distinct raw materials (crude palm oil (CPO) and refined, bleached, and deodorized (RBD) palm oil), instead of conventional LPG. The LCA findings reveal that utilizing LPG fuel in small fishing vessels can reduce greenhouse gas (GHG) emissions by more than 30% over conventional gasoline and diesel fuels. During the life cycle of vessels that use LPG fuel instead of gasoline and diesel fuels, there is a reduction of 2.2 and 1.2 million tons of GHG emissions, respectively. Moreover, substituting conventional fossil fuels with Bio LPG can result in over 65% reduction in GHG emissions. For the life cycle of boats that use Bio LPG fuel in place of gasoline and diesel fuels, the reduction of GHG emissions was 4.9 million tons and 2.5 million tons for CPO and 5.2 million tons and 2.7 million tons for RBD, respectively. This study not only underscores the substantial advantages of using Bio LPG over conventional fossil fuels but also presents conventional LPG as a way to reduce GHG emissions and promote sustainable practices in the fishing industry
The Rhizome Mixture of Anemarrhena asphodeloides
We investigated the effect of DWac on the gut microbiota composition in mice with 2,3,6-trinitrobenzenesulfonic acid- (TNBS-) induced colitis. Treatment with DWac restored TNBS-disturbed gut microbiota composition and attenuated TNBS-induced colitis. Moreover, we examined the effect of DWac in mice with mesalazine-resistant colitis (MRC). Intrarectal injection of TNBS in MRC mice caused severe colitis, as well as colon shortening, edema, and increased myeloperoxidase activity. Treatment with mesalazine (30 mg/kg) did not attenuate TNBS-induced colitis in MRC mice, whereas treatment with DWac (30 mg/kg) significantly attenuated TNBS-induced colitis. Moreover, treatment with the mixture of mesalazine (15 mg/kg) and DWac (15 mg/kg) additively attenuated colitis in MRC mice. Treatment with DWac and its mixture with mesalazine inhibited TNBS-induced activation of NF-κB and expression of M1 macrophage markers but increased TNBS-suppressed expression of M2 macrophage markers. Furthermore, these inhibited TNBS-induced T-bet, RORγt, TNF-α, and IL-17 expression but increased TNBS-suppressed Foxp3 and IL-10 expression. However, Th2 cell differentiation and GATA3 and IL-5 expression were not affected. These findings suggest that DWac can ameliorate MRC by increasing the polarization of M2 macrophage and correcting the disturbance of gut microbiota and Th1/Th17/Treg, as well as additively attenuating MRC along with mesalazine
Solid–liquid interface temperature measurement of evaporating droplet using thermoresponsive polymer aqueous solution
The present study aims to measure the solid–liquid interface temperature of an evaporating droplet on a heated surface using a thermoresponsive polymer. Poly(N-isopropylacrylamide) (pNIPAM) was used owing to its sensitive optical and mechanical properties to the temperature. We also measured the refractive index variation of the pNIPAM solution by using the surface plasmon resonance imaging (SPRi). In particular, the present study proposed a new method to measure the solid–liquid interface temperature using the correlation among reflectance, refractive index, and temperature. It was found that the reflectance of a pNIPAM solution decreased after the droplet deposition. The solid–liquid interface temperature, estimated from the reflectance, showed a lower value at the center of the droplet, and it gradually increased along the radial direction. The lowest temperature at the contact line region is present because of the maximum evaporative cooling. Moreover, the solid–liquid interface temperature deviation increased with the surface temperature, which means solid–liquid interface temperature should be considered at high temperature to predict the evaporation flux of the droplet accurately
