A comparison of the economic value for enteric methane emissions with other biological traits associated with dairy cows
This is the first study to derive the economic value of enteric methane produced by a ruminant animal. There is considerable interest globally in selecting for low methane-emitting ruminant livestock, as methane is a potent greenhouse gas. However, before enteric methane can be included in a genetic selection index for breeding, the economic value for enteric methane needs to be derived. An animal model including a partial budget was used to derive economic values for a range of production and fitness (health and fertility) traits typically used in genetic selection of dairy cows with the addition of enteric methane. This study found that enteric methane (kilograms/lactation) has an economic value of -Ā£1.68 per kg increase in methane per lactation. The economic value for enteric methane was of similar magnitude to the traits of milk fat yield (Ā£1.14 per unit change in milk fat) and mastitis (-Ā£1.55 per % incidence). Based on the variation seen in the dairy cow population in the UK, genetic selection on enteric methane has potential to increase herd profit per cow and reduce emissions. Even if the economic and abatement gains associated with selecting low methane producing livestock are relatively small, reductions in enteric methane emissions appear possible if a reliable and repeatable measure becomes available for use on commercial farms
Alcohol involvement in aggression between intimate partners in New Zealand: a national cross-sectional study
Objectives: To examine the role of alcohol at the time of aggressive incidents between intimate partners in the general population by gender, by estimating (1) prevalence and severity of aggression, and drinking at the time, (2) associations of drinking at the time of the aggression with reported severity, anger and fear, and (3) association of usual drinking patterns with partner aggression. Design: A national survey of 18-70-year-olds using an electoral roll sample obtained self-reported alcohol consumption, partnerās alcohol consumption and details of the most severe partner aggression by the respondent and towards the respondent in the past 2 years. The mean scores for associated severity, anger and fear were analysed by gender and alcohol involvement. Multinomial models estimated associations of drinking patterns with aggression to and from the respondent. Results: The response rate was 49% (n=1925). Men and women reported similar prevalence of victimisation and perpetration of aggression (11-15%). Alcohol was involved in more than 25% of incidents, and reported more by women than by men, particularly male-only drinking when the respondent was the victim. Women reported greater severity, anger and fear with victimisation than men, and drinking was associated with greater reported severity. Heavy episodic drinking by respondents was associated with a threefold increase in victimisation and doubling of perpetration of aggression involving alcohol. Heavy episodic drinking by either partner was also associated with drinking being involved in reported aggression. Conclusions: The experience of intimate-partner aggression in a cross-section of households differs by gender and the involvement of alcohol, and ācountsā of aggressive acts in a population-based survey do not reflect the reality of gender differences. Heavy episodic drinking patterns are associated with more aggression involving alcohol within relationships, and alcohol involvement is associated with increased severity

The impact of genetic selection on greenhouse-gas emissions in Australian dairy cattle
In Australia, dairy cattle account for ~12% of the nationās agricultural greenhouse-gas (GHG) emissions. Genetic selection has had a positive impact, reducing GHG emissions from dairy systems mainly due to increased production per cow, which has led to (1) requiring fewer cows to produce the same amount of milk and (2) lowering emissions per unit of milk produced (emission intensity). The objective of the present study was to evaluate the consequences of previous and current genetic-selection practices on carbon emissions, using realised and predicted responses to selection for key traits that are included in the Australian national breeding objective. A farm model was used to predict the carbon dioxide equivalent (COā-eq) emissions per unit change of these traits, while holding all other traits constant. Estimates of the realised change in annual COā-eq emissions per cow over the past decade were made by multiplying predicted COā-eq emissions per unit change of each trait under selection by the realised rates of genetic gain in each of those traits. The total impact is estimated to be an increase of 55 kg COā-eq/cow.year after 10 years of selection. The same approach was applied to future COā-eq emissions, except predicted rates of genetic gain assumed to occur over the next decade through selection on the Balanced Performance Index (BPI) were used. For an increase of AU100). In fact, cow numbers have decreased over the past decade and production has increased; altogether, we estimate that the net impact has been a reduction of COā-eq emissions of ~1.0% in total emissions from the dairy industry per year. Using two future scenarios of either keeping the number of cows or amount of product static, we predict that net GHG emissions will reduce by ~0.6%/year of total dairy emissions if milk production remains static, compared with 0.3%/year, if cow numbers remain the same and there is genetic improvement in milk-production traits
The social affordances of flashpacking: exploring the mobility nexus of travel and communication
The proliferation of digital devices and online social media and networking technologies has altered the backpacking landscape in recent years. Thanks to the ready availability of online communication, travelers are now able to stay in continuous touch with friends, family and other travelers while on the move. This article introduces the practice of āflashpackingā to describe this emerging trend and interrogates the patterns of connection and disconnection that become possible as corporeal travel and social technologies converge. Drawing on the concepts of āassemblagesā and āaffordancesā, we outline several aspects of this new sociality: virtual mooring, following, collaborating, and (dis)connecting. The conclusion situates this discussion alongside broader questions about the shifting nature of social life in an increasingly mobile and mediated world and suggests directions for future research at the intersection of tourism and technology
Using a Non-Image-Based Medium-Throughput Assay for Screening Compounds Targeting N-myristoylation in Intracellular Leishmania Amastigotes
We have refined a medium-throughput assay to screen hit compounds for activity against N-myristoylation in intracellular amastigotes of Leishmania donovani. Using clinically-relevant stages of wild type parasites and an Alamar blue-based detection method, parasite survival following drug treatment of infected macrophages is monitored after macrophage lysis and transformation of freed amastigotes into replicative extracellular promastigotes. The latter transformation step is essential to amplify the signal for determination of parasite burden, a factor dependent on equivalent proliferation rate between samples. Validation of the assay has been achieved using the anti-leishmanial gold standard drugs, amphotericin B and miltefosine, with EC50 values correlating well with published values. This assay has been used, in parallel with enzyme activity data and direct assay on isolated extracellular amastigotes, to test lead-like and hit-like inhibitors of Leishmania Nmyristoyl transferase (NMT). These were derived both from validated in vivo inhibitors of Trypanosoma brucei NMT and a recent high-throughput screen against L. donovani NMT. Despite being a potent inhibitor of L. donovani NMT, the activity of the lead T. brucei NMT inhibitor (DDD85646) against L. donovani amastigotes is relatively poor. Encouragingly, analogues of DDD85646 show improved translation of enzyme to cellular activity. In testing the high-throughput L. donovani hits, we observed macrophage cytotoxicity with compounds from two of the four NMT-selective series identified, while all four series displayed low enzyme to cellular translation, also seen here with the T. brucei NMT inhibitors. Improvements in potency and physicochemical properties will be required to deliver attractive lead-like Leishmania NMT inhibitors
The American Congress of Rehabilitation Medicine Diagnostic Criteria for Mild Traumatic Brain Injury
Objective: To develop new diagnostic criteria for mild traumatic brain injury (TBI) that are appropriate for use across the lifespan and in sports, civilian trauma, and military settings. Design: Rapid evidence reviews on 12 clinical questions and Delphi method for expert consensus. Participants: The Mild Traumatic Brain Injury Task Force of the American Congress of Rehabilitation Medicine Brain Injury Special Interest Group convened a Working Group of 17 members and an external interdisciplinary expert panel of 32 clinician-scientists. Public stakeholder feedback was analyzed from 68 individuals and 23 organizations. Results: The first 2 Delphi votes asked the expert panel to rate their agreement with both the diagnostic criteria for mild TBI and the supporting evidence statements. In the first round, 10 of 12 evidence statements reached consensus agreement. Revised evidence statements underwent a second round of expert panel voting, where consensus was achieved for all. For the diagnostic criteria, the final agreement rate, after the third vote, was 90.7%. Public stakeholder feedback was incorporated into the diagnostic criteria revision prior to the third expert panel vote. A terminology question was added to the third round of Delphi voting, where 30 of 32 (93.8%) expert panel members agreed that āthe diagnostic label āconcussionā may be used interchangeably with āmild TBIā when neuroimaging is normal or not clinically indicated.ā Conclusions: New diagnostic criteria for mild TBI were developed through an evidence review and expert consensus process. Having unified diagnostic criteria for mild TBI can improve the quality and consistency of mild TBI research and clinical care.</p
