Ninety Years of Perennial Forage Grass Breeding for the Canadian Prairie Provinces
The majority of Canadian beef production takes place in the three prairie provinces of western Canada and seeded pasture and hay land provides a large proportion of the feed for these cattle. This region is characterized by low to moderate rainfall (300-500 mm annually) and extremely cold winters; thus, forage species grown must have high tolerance to cold and drought. Perennial grass breeding for the region began in 1922 and has continued for ninety years in two programs. Major seeded grass species were introduced into Canada from Eurasia. Significant developments from this program include: the development of Fairway crested wheatgrass (Agropyron cristatum L.) (1932) which was used to revegetate more than 0.5 million ha of eroded land; the introduction and development of improved cultivars of meadow bromegrass (Bromus riparius Rehm.) (1987) which were rapidly adopted by beef producers, making it the most popular grazing species in the prairie provinces; the development of hybrid bromegrass (B. riparius X B. inermis Leyss.) cultivars (2000) which have become popular dual purpose hay/pasture grasses; and the development of green wheatgrass (Elymus hoffmanii K.B. Jensen and K.H. Asay), which is highly tolerant to soil salinity. Recently, there has been increased demand for native Canadian grasses to be used for forage, conservation and reclamation purposes and cultivars or ecological varieties of several species have been released or are under development. Efficacy, Safety, and Timing of Anticoagulant Thromboprophylaxis for the Prevention of Venous Thromboembolism in Patients With Acute Spinal Cord Injury: A Systematic Review
Study Design: Systematic review. Objectives: The objective of this study was to answer 5 key questions: What is the comparative effectiveness and safety of (1a) anticoagulant thromboprophylaxis compared to no prophylaxis, placebo, or another anticoagulant strategy for preventing deep vein thrombosis (DVT) and pulmonary embolism (PE) after acute spinal cord injury (SCI)? (1b) Mechanical prophylaxis strategies alone or in combination with other strategies for preventing DVT and PE after acute SCI? (1c) Prophylactic inferior vena cava filter insertion alone or in combination with other strategies for preventing DVT and PE after acute SCI? (2) What is the optimal timing to initiate and/or discontinue anticoagulant, mechanical, and/or prophylactic inferior vena cava filter following acute SCI? (3) What is the cost-effectiveness of these treatment options? Methods: A systematic literature search was conducted to identify studies published through February 28, 2015. We sought randomized controlled trials evaluating efficacy and safety of antithrombotic strategies. Strength of evidence was evaluated using the Grading of Recommendations Assessment, Development and Evaluation (GRADE) system. Results: Nine studies satisfied inclusion criteria. We found a trend toward lower risk of DVT in patients treated with enoxaparin. There were no significant differences in rates of DVT, PE, bleeding, and mortality between patients treated with different types of low-molecular-weight heparin or between low-molecular-weight heparin and unfractionated heparin. Combined anticoagulant and mechanical prophylaxis initiated within 72 hours of SCI resulted in lower risk of DVT than treatment commenced after 72 hours of injury. Conclusion: Prophylactic treatments can be used to lower the risk of venous thromboembolic events in patients with acute SCI, without significant increase in risk of bleeding and mortality and should be initiated within 72 hours. © 2017, © The Author(s) 2017
Band gap reduction in GaNSb alloys due to the anion mismatch
The structural and optoelectronic properties in GaNxSb1–x alloys (0<=x<0.02) grown by molecular-beam epitaxy on both GaSb substrates and AlSb buffer layers on GaAs substrates are investigated. High-resolution x-ray diffraction (XRD) and reciprocal space mapping indicate that the GaNxSb1–x epilayers are of high crystalline quality and the alloy composition is found to be independent of substrate, for identical growth conditions. The band gap of the GaNSb alloys is found to decrease with increasing nitrogen content from absorption spectroscopy. Strain-induced band-gap shifts, Moss-Burstein effects, and band renormalization were ruled out by XRD and Hall measurements. The band-gap reduction is solely due to the substitution of dilute amounts of highly electronegative nitrogen for antimony, and is greater than observed in GaNAs with the same N content
Species-level classification of the vaginal microbiome
Background
The application of next-generation sequencing to the study of the vaginal microbiome is revealing the spectrum of microbial communities that inhabit the human vagina. High-resolution identification of bacterial taxa, minimally to the species level, is necessary to fully understand the association of the vaginal microbiome with bacterial vaginosis, sexually transmitted infections, pregnancy complications, menopause, and other physiological and infectious conditions. However, most current taxonomic assignment strategies based on metagenomic 16S rDNA sequence analysis provide at best a genus-level resolution. While surveys of 16S rRNA gene sequences are common in microbiome studies, few well-curated, body-site-specific reference databases of 16S rRNA gene sequences are available, and no such resource is available for vaginal microbiome studies. Results
We constructed the Vaginal 16S rDNA Reference Database, a comprehensive and non-redundant database of 16S rDNA reference sequences for bacterial taxa likely to be associated with vaginal health, and we developed STIRRUPS, a new method that employs the USEARCH algorithm with a curated reference database for rapid species-level classification of 16S rDNA partial sequences. The method was applied to two datasets of V1-V3 16S rDNA reads: one generated from a mock community containing DNA from six bacterial strains associated with vaginal health, and a second generated from over 1,000 mid-vaginal samples collected as part of the Vaginal Human Microbiome Project at Virginia Commonwealth University. In both datasets, STIRRUPS, used in conjunction with the Vaginal 16S rDNA Reference Database, classified more than 95% of processed reads to a species-level taxon using a 97% global identity threshold for assignment. Conclusions
This database and method provide accurate species-level classifications of metagenomic 16S rDNA sequence reads that will be useful for analysis and comparison of microbiome profiles from vaginal samples. STIRRUPS can be used to classify 16S rDNA sequence reads from other ecological niches if an appropriate reference database of 16S rDNA sequences is available
Efficacy and safety of methylprednisolone sodium succinate in acute spinal cord injury: a systematic review
Study Design: Systematic review and meta-analysis. Objective: The objective of this study was to conduct a systematic review to assess the comparative effectiveness and safety of high-dose methylprednisolone sodium succinate (MPSS) versus no pharmacological treatment in patients with traumatic spinal cord injury (SCI). Methods: A systematic search was performed in PubMed and the Cochrane Collaboration Library for literature published between January 1956 and June 17, 2015. Included studies ere critically appraised, and Grades of Recommendation Assessment, Development and Evaluation methods were used to determine the overall quality of evidence for primary outcomes. Previous systematic reviews on this topic were collated and evaluated using the Assessment of Multiple Systematic Reviews scoring system. Results: The search yielded 723 citations, 13 of which satisfied inclusion criteria. Among these, 6 were primary research articles and 7 were previous systematic reviews. Based on the included research articles, there was moderate evidence that the 24-hour NASCIS II (National Acute Spinal Cord Injury Studies) MPSS regimen has no impact on long-term neurological recovery when all postinjury time points are considered. However, there is also moderate evidence that subjects receiving the same MPSS regimen within 8 hours of injury achieve an additional 3.2 points (95% confidence interval = 0.10 to 6.33; P = .04) of motor recovery compared with patients receiving placebo or no treatment. Conclusion: Although safe to administer, a 24-hour NASCIS II MPSS regimen, when all postinjury time points are considered, has no impact on indices of long-term neurological recovery. When commenced within 8 hours of injury, however, a high-dose 24-hour regimen of MPSS confers a small positive benefit on long-term motor recovery and should be considered a treatment option for patients with SCI
Runx1 promotes scar deposition and inhibits myocardial proliferation and survival during zebrafish heart regeneration.
Runx1 is a transcription factor that plays a key role in determining the proliferative and differential state of multiple cell types, during both development and adulthood. Here, we report how Runx1 is specifically upregulated at the injury site during zebrafish heart regeneration, and that absence of runx1 results in increased myocardial survival and proliferation, and overall heart regeneration, accompanied by decreased fibrosis. Using single cell sequencing, we found that the wild-type injury site consists of Runx1-positive endocardial cells and thrombocytes that induce expression of smooth muscle and collagen genes. Both these populations cannot be identified in runx1 mutant wounds that contain less collagen and fibrin. The reduction in fibrin in the mutant is further explained by reduced myofibroblast formation and upregulation of components of the fibrin degradation pathway, including plasminogen receptor annexin 2A as well as downregulation of plasminogen activator inhibitor serpine1 in myocardium and endocardium, resulting in increased levels of plasminogen. Our findings suggest that Runx1 controls the regenerative response of multiple cardiac cell types and that targeting Runx1 is a novel therapeutic strategy for inducing endogenous heart repair.BHF, Wellcome, MR

1. This account presents information on all aspects of the biology of Geranium pratense L. (Meadow Crane's‐Bill). The main topics are presented within the standard framework of the Biological Flora of Britain and Ireland : distribution, habitat, communities, responses to biotic factors, responses to environment, structure and physiology, phenology, floral and seed characters, herbivores and disease, history and conservation.2. Geranium pratense is a perennial gynodioecious forb of neutral grassland. In Britain and Ireland, it is particularly abundant on roadside verges, railway embankments, the margins of watercourses and woodland rides. It is generally intolerant of grazing and is absent or scarce in livestock‐grazed grassland. Geranium pratense is widespread in England, Wales and Scotland but is scarce in Ireland. It has an extensive native range in Europe and Asia, extending eastwards to Russia, north‐western China and Mongolia. It has been widely introduced to new sites within its native range and has been introduced to Canada, the USA and New Zealand. 3. Geranium pratense usually occurs on free‐draining soils but also infrequently where drainage is impeded. The soils are often nutrient‐rich and weakly acidic to weakly alkaline. The underlying geology is usually non‐acidic sedimentary rocks or superficial deposits. 4. Geranium pratense is protandrous and is pollinated by various insects of the orders Hymenoptera, Diptera and Lepidoptera, particularly bumblebees, solitary bees, hoverflies and butterflies. Eleven species of phytophagous insect have been recorded on G. pratense in Britain and Ireland.5. Geranium pratense has little or no capacity for vegetative spread. Primary seed dispersal is ballistic and seeds may be flung over distances of up to several metres. The species has a transient seed bank, that is germination typically takes place in the winter and spring after seed production, after the physically dormant seeds have become permeable. Seedling establishment is higher in vegetation‐free gaps than in undisturbed grassland vegetation.6. There has been no significant change in its distribution between the late 1950s and 2019, although since 2000, it has expanded its range, mainly via introductions, in northern and western Scotland, west Wales and in Ireland.
