Using Fluid-Mobile Elements to Decipher an Aqueous History Preserved in the Sedimentary Rocks of Gale Crater, Mars
Fluid-mobile elements preserve a record of the aqueous history of Mars in the sedimentary rocks of Gale Crater, and they are traceable with the rover Curiosity’s Alpha Particle X-ray Spectrometer (APXS). This is the fundamental principle behind the scientific questions addressed in the four main chapters of this dissertation. First, we conducted a field study at Hawai’i, an established Mars analogue, to investigate sedimentary processes and alteration in rocks comparable to those in Gale Crater. We conclude that open system, circumneutral weathering is very limited in Gale rocks; acid sulfate alteration and sedimentary mixing of diverse rocks is likely. Second, the Hawaiian samples were analyzed by Particle Induced X-ray Emission spectroscopy (PIXE) to evaluate the technique as an analogue for APXS. Third, we developed a novel, non-standard APXS calibration to determine the composition of a very thin layer of airfall dust on the rover. The global dust is enriched in the fluid-mobile elements sulfur and chlorine, but at a ratio found to be constant around the planet. This enables deconvolution of modern dust from underlying rock in APXS measurements to determine inherent sulfur and chlorine content in ancient rocks. Fourth, we used fortuitous zinc and germanium enrichments in Gale Crater rocks to establish a geochemical tracer pair for fluid processes in Gale. We propose a model in which the two trace elements were enriched together by hydrothermal fluids in the sediment source region, transported into the crater, and then fractionated by low temperature diagenetic fluids. The work presented here constrains fluid events affecting Gale sediments and can aid in unraveling fluid histories as Curiosity’s traverse continues
Joint Genetic Analysis of Conception and Maintenance of Pregnancy in Dairy Cattle Using a Linear-Threshold Model
A joint analysis of days open and calving success was implemented to further enhance the identification of cows with greater genetic merit for reproductive performance. Calving success is a categorical trait with similar genetic variation as days open and is analyzed jointly with days open to make effective use of the positive genetic correlation between these two traits. The joint analysis can identify cows with greater genetic merit for conception at an earlier stage of lactation and greater success at maintenance of pregnancy. The joint analysis also enhances the reliability of detecting true genetic differences between cows for fertility because it makes more effective use of all data
Genetic Analyses of Days Open Using a Random Regression Model
The purpose of this study was to investigate the feasibility of using a linear random regression model for the analysis of female fertility as expressed by days open. The analysis provided insight about the kind and amount of variation over the trajectory for age at calving from 24 to 72 mo. Correlations between permanent environmental effects changed from positive to negative values with greater distance between days open in young cows and days open in older cows; r(24,36)=0.88, r(24,72)=-.65. The random regression animal model proved to be a better alternative to a repeatability animal model for prediction of breeding values for days open. Random regression enables breeders to pick the optimum age at calving to select female replacements with enhanced fertility or fewer days open per lactation. Further research is needed to evaluate the advantages of using random regression model on the genetic analysis of days open
An Investigation of Rotorcraft Stability-Phase Margin Requirements in Hover
A cooperative study was performed to investigate the handling quality effects from reduced flight control system stability margins, and the trade-offs with higher disturbance rejection bandwidth (DRB). The piloted simulation study, perform on the NASA-Ames Vertical Motion Simulator, included three classes of rotorcraft in four configurations: a utility-class helicopter; a medium-lift helicopter evaluated with and without an external slung load; and a large (heavy-lift) civil tiltrotor aircraft. This large aircraft also allowed an initial assessment of ADS-33 handling quality requirements for an aircraft of this size. Ten experimental test pilots representing the U.S. Army, Marine Corps, NASA, rotorcraft industry, and the German Aerospace Center (DLR), evaluated the four aircraft configurations, for a range of flight control stability-margins and turbulence levels, while primarily performing the ADS-33 Hover and Lateral Reposition MTEs. Pilot comments and aircraft-task performance data were analyzed. The preliminary stability margin results suggest higher DRB and less phase margin cases are preferred as the aircraft increases in size. Extra care will need to be taken to assess the influence of variability when nominal flight control gains start with reduced margins. Phase margins as low as 20-23 degrees resulted in low disturbance-response damping ratios, objectionable oscillations, PIO tendencies, and a perception of an incipient handling qualities cliff. Pilot comments on the disturbance response of the aircraft correlated well to the DRB guidelines provided in the ADS-33 Test Guide. The A D-3S3 mid-term response-to-control damping ratio metrics can be measured and applied to the disturbance-response damping ratio. An initial assessment of LCTR yaw bandwidth shows the current Level 1 boundary needs to be relaxed to help account for a large pilot off-set from the c.g. Future efforts should continue to investigate the applicability/refinement of the current ADS-33 requirements to large vehicles, like an LCTR
A global Mars dust composition refined by the Alpha-Particle X-ray Spectrometer in Gale Crater
Modern Martian dust is similar in composition to the global soil unit and bulk basaltic Mars crust, but it is enriched in S and Cl. The Alpha Particle X-ray Spectrometer (APXS) on the Mars Science Laboratory Curiosity rover analyzed air fall dust on the science observation tray (o-tray) in Gale Crater to determine dust oxide compositions. The o-tray dust has the highest concentrations of SO3 and Cl measured in Mars dust (SO3 8.3%; Cl 1.1 wt %). The molar S/Cl in the dust (3.35 ± 0.34) is consistent with previous studies of Martian dust and soils (S/Cl = 3.7 ± 0.7). Fe is also elevated ~25% over average Mars soils and the bulk crust. These enrichments link air fall dust with the S-, Cl-, and Fe-rich X-ray amorphous component of Gale Crater soil. Dust and soil have the same S/Cl, constraining the surface concentrations of S and Cl on a global scale
PTF10fqs: A Luminous Red Nova in the Spiral Galaxy Messier 99
The Palomar Transient Factory (PTF) is systematically charting the optical
transient and variable sky. A primary science driver of PTF is building a
complete inventory of transients in the local Universe (distance less than 200
Mpc). Here, we report the discovery of PTF10fqs, a transient in the luminosity
"gap" between novae and supernovae. Located on a spiral arm of Messier 99, PTF
10fqs has a peak luminosity of Mr = -12.3, red color (g-r = 1.0) and is slowly
evolving (decayed by 1 mag in 68 days). It has a spectrum dominated by
intermediate-width H (930 km/s) and narrow calcium emission lines. The
explosion signature (the light curve and spectra) is overall similar to thatof
M85OT2006-1, SN2008S, and NGC300OT. The origin of these events is shrouded in
mystery and controversy (and in some cases, in dust). PTF10fqs shows some
evidence of a broad feature (around 8600A) that may suggest very large
velocities (10,000 km/s) in this explosion. Ongoing surveys can be expected to
find a few such events per year. Sensitive spectroscopy, infrared monitoring
and statistics (e.g. disk versus bulge) will eventually make it possible for

Acute vasoreactivity testing in pediatric idiopathic pulmonary arterial hypertension:an international survey on current practice
The aim of this study was to determine practice patterns and inter-institutional variability in how acute vasoreactivity testing (AVT) is performed and interpreted in pediatrics throughout the world. A survey was offered to physicians affiliated with the Pediatric & Congenital Heart Disease Taskforce of the Pulmonary Vascular Research Institute (PVRI), the Pediatric Pulmonary Hypertension Network (PPHNET), or the Spanish Registry for Pediatric Pulmonary Hypertension (REHIPED), from February to December 2016. The survey requested data about the site-specific protocol for AVT and subsequent management of pediatric patients with idiopathic pulmonary arterial hypertension (IPAH) or heritable PAH (HPAH). Twenty-eight centers from 13 countries answered the survey. AVT is performed in most centers using inhaled nitric oxide (iNO). Sitbon criteria was used in 39% of the centers, Barst criteria in 43%, and other criteria in 18%. First-line therapy for positive AVT responders in functional class (FC) I/II was calcium channel blocker (CCB) in 89%, but only in 68% as monotherapy. Most centers (71%) re-evaluated AVT-positive patients hemodynamics after 6-12 months; 29% of centers re-evaluated based only on clinical criteria. Most centers (64%) considered a good response as remaining in FC I or II, with near normalization of pulmonary arterial pressure and pulmonary vascular resistance, but a stable FC I/II alone was sufficient criteria in 25% of sites. Protocols and diagnostic criteria for AVT, and therapeutic approaches during follow-up, were highly variable across the world. Reported clinical practice is not fully congruent with current guidelines, suggesting the need for additional studies that better define the prognostic value of AVT for pediatric IPAH patients
Elemental Composition and Chemical Evolution of Geologic Materials in Gale Crater, Mars: APXS Results From Bradbury Landing to the Vera Rubin Ridge
The Alpha Particle X-ray Spectrometer (APXS) on the rover Curiosity has analyzed the composition of geologic materials along a >20-km traverse in Gale crater on Mars. The APXS dataset after 6.5 Earth years (2,301 sols) includes 712 analyses of soil, sand, float, bedrock, and drilled/scooped fines. We present the APXS results over this duration and provide stratigraphic context for each target. We identify the best APXS measurement of each of the 22 drilled and scooped samples that were delivered to the instruments Chemistry and Mineralogy (CheMin; X-ray diffractometer) and Sample Analysis at Mars (SAM; mass spectrometer and gas chromatograph) during this period. The APXS results demonstrate that the basaltic and alkali-rich units in the Bradbury group (sols 0-750) show minimal alteration indicating an arid climate. In contrast, the Murray formation of the Mount Sharp group (sols ∼750-2,301) has compositions indicating pervasive alteration. Diagenetic features are common and show fluid interaction with the sediment after (and possibly during) lithification. A sandstone unit, the Stimson formation, overlies part of the Murray formation. This has a composition similar to the basaltic sand and soil, suggesting a shared source. Cross-cutting, fracture-associated haloes are evidence of late-stage fluid alteration after lithification of the sediment. The APXS dataset, evaluated in concert with the full science payload of Curiosity, indicates that Gale crater was habitable, and that liquid water was stable for extended periods.We are indebted to NASA-JPL, the Canadian Space Agency, and Australian Research Council (DP150104604) for supporting our work and the MSL mission. macroH2A2 antagonizes epigenetic programs of stemness in glioblastoma
Self-renewal is a crucial property of glioblastoma cells that is enabled by the choreographed functions of chromatin regulators and transcription factors. Identifying targetable epigenetic mechanisms of self-renewal could therefore represent an important step toward developing effective treatments for this universally lethal cancer. Here we uncover an epigenetic axis of self-renewal mediated by the histone variant macroH2A2. With omics and functional assays deploying patient-derived in vitro and in vivo models, we show that macroH2A2 shapes chromatin accessibility at enhancer elements to antagonize transcriptional programs of self-renewal. macroH2A2 also sensitizes cells to small molecule-mediated cell death via activation of a viral mimicry response. Consistent with these results, our analyses of clinical cohorts indicate that high transcriptional levels of this histone variant are associated with better prognosis of high-grade glioma patients. Our results reveal a targetable epigenetic mechanism of self-renewal controlled by macroH2A2 and suggest additional treatment approaches for glioblastoma patients
