    Dynamics of the formation of a hydrogel by a pathogenic amyloid peptide: islet amyloid polypeptide

    Many chronic degenerative diseases result from aggregation of misfolded polypeptides to form amyloids. Many amyloidogenic polypeptides are surfactants and their assembly can be catalysed by hydrophobic-hydrophilic interfaces (an air-water interface in-vitro or membranes in-vivo). We recently demonstrated the specificity of surface-induced amyloidogenesis but the mechanisms of amyloidogenesis and more specifically of adsorption at hydrophobic-hydrophilic interfaces remain poorly understood. Thus, it is critical to determine how amyloidogenic polypeptides behave at interfaces. Here we used surface tensiometry, rheology and electron microscopy to demonstrate the complex dynamics of gelation by full-length human islet amyloid polypeptide (involved in type II diabetes) both in the bulk solution and at hydrophobic-hydrophilic interfaces (air-water interface and phospholipids). We show that the hydrogel consists of a 3D supramolecular network of fibrils. We also assessed the role of solvation and dissected the evolution over time of the assembly processes. Amyloid gelation could have important pathological consequences for membrane integrity and cellular functions

    Site occupation constraints in mean-field approaches of quantum spin systems at finite temperature

    We study the effect of site occupation on the description of quantum spin systems at finite temperature and mean-field level. We impose each lattice site to be occupied by a single electron. This is realized by means of a specific prescription. The outcome of the prescription is compared to the result obtained by means of a projection procedure which fixes the site occupation to one particle per site on an average. The comparison is performed for different representations of the Hamiltonian in Fock space leading to different types of mean-field solutions. The behaviour of order parameters is analyzed for each choice of the mean-field and constraint which fixes the occupation rate at each site. Sizable quantitative differences between the outcomes obtained with the two different constraints are observed.Comment: 18 pages, 2 figure

    With support from The Wallace Foundation, the Harvard Family Research Project and P/PV conducted a study of almost 200 out-of-school-time (OST) programs in six cities to better understand how they promote sustained participation among older youth.The resulting data indicated that two of the most important practices distinguishing high-retention programs were: ample leadership opportunities for youth and high levels of staff efforts to keep informed about participants' lives outside the programs. The study also compares and contrasts effective practices for middle school- versus high school-aged youth, noting the shortcomings of "one-size-fits-all" strategies.     Radiatively-induced Magnetic moment in four-dimensional anisotropic QED in an external magnetic field

    We discuss one-loop radiatively-induced magnetic moment in four-dimensional quantum electrodynamics (QED) with anisotropic coupling, and examine various cases which may be of interest in effective gauge theories of antiferromagnets, whose planar limit coresponds to highly anisotropic QED couplings. We find a different scaling with the magnetic field intensity in case there are extra statistical gauge interactions in the model with spontaneous symmetry breaking. Such a case is encountered in the CP-1 sigma-model sector of effective spin-charge separated gauge theories of antiferromagnetic systems. Our work provides therefore additional ways of possible experimental probing of the gauge nature of such systems.Comment: 14 pages Latex, no figure

    Critical examination is made on the relation between the mass shift of vector mesons in nuclear medium and the vector-meson −- nucleon scattering length. We give detailed comparison between the QCD sum rule approach by two of the present authors (Phys. Rev. {\bf C46} (1992) R34) and the scattering-length approach by Koike (Phys.     In vivo parasitological measures of artemisinin susceptibility

    Parasite clearance data from 18,699 patients with falciparum malaria treated with an artemisinin derivative in areas of low (n=14,539), moderate (n=2077), and high (n=2083) levels of malaria transmission across the world were analyzed to determine the factors that affect clearance rates and identify a simple in vivo screening measure for artemisinin resistance. The main factor affecting parasite clearance time was parasite density on admission. Clearance rates were faster in high-transmission settings and with more effective partner drugs in artemisinin-based combination treatments (ACTs). The result of the malaria blood smear on day 3 (72 h) was a good predictor of subsequent treatment failure and provides a simple screening measure for artemisinin resistance. Artemisinin resistance is highly unlikely if the proportion of patients with parasite densities of <100,000 parasites/microL given the currently recommended 3-day ACT who have a positive smear result on day 3 is <3%; that is, for n patients the observed number with a positive smear result on day 3 does not exceed (n + 60)/24

    Factors associated with alcohol reduction in harmful and hazardous drinkers following alcohol brief intervention in Scotland: a qualitative enquiry

    Background: Alcohol Brief Intervention (ABI) uses a motivational counselling approach to support individuals to reduce excessive alcohol consumption. There is growing evidence on ABI’s use within various health care settings, although how they work and which components enhance success is largely unknown. This paper reports on the qualitative part of a mixed methods study. It explores enablers and barriers associated with alcohol reduction following an ABI. It focuses on alcohol’s place within participants’ lives and their personal perspectives on reducing consumption. There are a number of randomised controlled trials in this field though few ABI studies have addressed the experiences of hazardous/harmful drinkers. This study examines factors associated with alcohol reduction in harmful/hazardous drinkers following ABI. Methods: This qualitative study was underpinned by a realist evaluation approach and involved semistructured interviews with ten harmful or hazardous alcohol drinkers. Participants (n = 10) were from the intervention arm of a randomised controlled trial (n = 124). All had received ABI, a 20 min motivational counselling interview, six months previously, and had reduced their alcohol consumption. Interviews were recorded, transcribed verbatim and thematically analysed. Results: Participants described their views on alcohol, its’ place in their lives, their personal perspectives on reducing their consumption and future aspirations. Conclusions: The findings provide an insight into participants’ views on alcohol, ABI, and the barriers and enablers to change. Participants described a cost benefit analysis, with some conscious consideration of the advantages and disadvantages of reducing intake or abstaining from alcohol. Findings suggest that, whilst hospital admission can act as a catalyst, encouraging individuals to reflect on their alcohol consumption through ABI may consolidate this, turning this reflective moment into action. Sustainability may be enhanced by the presence of a ‘significant other’ who encourages and experiences benefit. In addition having a purpose or structure with activities linked to employment and/or social and leisure pursuits offers the potential to enhance and sustain reduced alcohol consumption.
